eIF2α confers cellular tolerance to S. aureus α-toxin

We report on the role of conserved stress-response pathways for cellular tolerance to a pore forming toxin. First, we observed that small molecular weight inhibitors including of eIF2α-phosphatase, jun-N-terminal kinase (JNK), and PI3-kinase sensitized normal mouse embryonal fibroblasts (MEFs) to the small pore forming S. aureus α-toxin. Sensitization depended on expression of mADAM10, the murine ortholog of a proposed high-affinity receptor for α-toxin in human cells. Similarly, eIF2α (S51A/S51A) MEFs, which harbor an Ala knock-in mutation at the regulated Ser51 phosphorylation site of eukaryotic translation initiation factor 2α, were hyper-sensitive to α-toxin. A new methodology for automated extraction of repeated patterns in time-series data is presented, aimed in particular at the analysis of musical sequences. The basic principles consists in a search for closed patterns in a multi-dimensional parametric space. It is shown that this basic mechanism needs to be articulated with a periodic pattern discovery system, implying therefore a strict chronological scanning of the time-series data. Thanks to this modelling global pattern filtering may be avoided and rich and highly pertinent results can be obtained. Our knowledge on fungal diversity and fungal ecology is however still very limited, in part because surveying and identifying fungi is time demanding and requires expert knowledge. First-generation extracorporeal shock-wave sources disintegrate 97% of kidney stones [1, 2]. Recently, in selected patients gallbladder and common bile duct stones were also treated. The technique available so far, however, requires immersion of the patient’s body in a tank of degassed water. The procedure is therefore inconvenient, time consuming, and relatively expensive.
