Sporogen, S14-95, and S-curvularin, three inhibitors of human inducible nitric-oxide synthase expression isolated from fungi.
2003
The induction of human inducible nitric-oxide synthase (iNOS) expression depends (among other factors) on activation of the signal transducer and activator of transcription 1 (STAT1) pathway. Therefore, the STAT1 pathway may be an appropriate target for the development of inhibitors of iNOS expression. HeLa S3 cells transiently transfected with a gamma-activated site (GAS)/interferon-stimulated response element-driven reporter gene construct were used as the primary screening system. Using this system, three novel inhibitors of interferon-gamma-dependent gene expression, namely, sporogen, S14-95, and S-curvularin, were isolated from different Penicillium species. These three compounds also …
Effects of ouabain on human bronchial muscle in vitro
2003
The effects of ouabain, an inhibitor of the plasmalemmal Na(+)/K(+)-ATPase activity, were examined in human isolated bronchus. Ouabain produced concentration-dependent contraction with -logEC(50)=7.16+/-0.11 and maximal effect of 67+/-4% of the response to acetylcholine (1 mM). Ouabain (10 microM)-induced contraction was epithelium-independent and was not depressed by inhibitors of cyclooxygenase and lipoxygenase, antagonists of muscarinic, histamine H(1)-receptors and alpha-adrenoceptors, or neuronal Na(+) channel blockade. The inhibition of ouabain contraction in tissues bathed in K(+)-free medium, and the inhibition by ouabain of the K(+)-induced relaxation confirm that the contractile a…
“Symptoms associated with environmental factors” (SAEF) – Towards a paradigm shift regarding “idiopathic environmental intolerance” and related pheno…
2020
Health conditions characterized by symptoms associated with chemical, physical and biological environmental factors unrelated to objectifiable pathophysiological mechanisms are often labelled by the general term “idiopathic environmental intolerances”. More specific, exposure-related terms are also used, e.g. “multiple chemical sensitivities”, “electromagnetic hypersensitivity” and “candidiasis hypersensitivity”. The prevalence of the conditions varies from a few up to more than 50%, depending on definitions and populations. Based on evolving knowledge within this field, we provide arguments for a paradigm shift from terms focusing on exposure and intolerance/(hyper-)sensitivity towards a t…
Functional characterization of protein variants of the human multidrug transporter ABCC2 by a novel targeted expression system in fibrosarcoma cells
2012
The multidrug resistance-associated protein 2 (MRP2/ABCC2) is involved in the efflux of endogenous and xenobiotic substrates, including several anticancer and antiviral drugs. The functional consequences of ABCC2 protein variants remain inconsistent, which may be due to shortcomings of the in vitro assays used. To study systematically the functional consequences of nonsynonymous ABCC2 variants, we used a novel “Screen and Insert” (ScIn) technology to achieve stable and highly reproducible expression of 13 ABCC2 variants in HT1080 cells. Western blotting revealed lower (30–65%) ABCC2 expression for D333G, R1174H, and R1181L as compared with wild type (WT; 100%), whereas the linked variant V1…
Morphological determination of the phototrophic community composition of biological soil crusts in coastal sand dunes in northern Germany
2022
This dataset comprises the microbial community composition of biological soil crusts in north-German sand dunes. For this we obtained enrichment cultures of phototrophic microorganisms, by placing fragments of biocrusts of the same Petri dishes as used for sequencing, in Petri dishes with Bold Basal (1N BBM) agarized medium (Bischoff and Bold 1963). Cultures were grown under standard laboratory conditions: with a 12-hour alteration of light and dark phases and irradiation of 25 μmol photons m-2 s-1 at a temperature 20 ± 5 ºС. Microscopic study of these raw cultures began in the third week of cultivation. Morphological examinations were performed using Olympus BX53 light microscope with Noma…
A Novel Technique Using Polytetrafluoroethylene Tape to Solve Screw Loosening Complication in Implant-Supported Single Crowns
2020
This study aimed to analyze a novel technique to make screws with greater untightening resistance and to solve screw loosening in implant-supported single crowns. Thirty grade IV titanium straight abutments were screwed onto 30 external hex implants using grade IV titanium screws (30 Ncm). They were exposed to cyclic loading (300,000 cycles, 200 N). Samples were divided into 4 groups (15 samples per group): new screws (SCREW group) (control), reused screws (rSCREW group), new screws wrapped with polytetrafluoroethylene (PTFE) tape (PTFE group), and reused screws wrapped with PTFE tape (rPTFE group). Reverse torque values (RTVs) were recorded with a digitally calibrated implant motor. Mean R…
The MARA-LEB ion transport system
2020
Abstract A low-energy branch is under development for the MARA vacuum-mode recoil separator at the Accelerator Laboratory of the University of Jyvaskyla. This development will allow for the study of proton-rich nuclei through laser ionisation spectroscopy and mass measurements. After stopping and extraction from a buffer gas cell, the ions of interest will be accelerated and transported to dedicated experimental setups by an ion transport system consisting of several focusing, accelerating and mass-separating elements. This article presents the current design and simulations for the ion transport.
