Effects of endothelin-1 on the cerebrovascular bed of the goat.

Abstract The aim of the present study was to analyze the effects of endothelin-1 (ET-1) in the cerebrovascular bed of the goat, the importance of endothelial integrity and the contribution of extracellular Ca2+ to these responses. We measured isometric tension and 45Ca2+ uptake in isolated middle cerebral arteries, and cerebral blood flow (CBF) in unanesthetized animals. ET-1 elicited concentration-dependent contractions which were potentiated in arteries without endothelium. C-peptide has been shown to increase skin microcirculation in patients with Type I (insulin-dependent) diabetes mellitus and to activate endothelial nitric oxide synthase. Current treatments for pulmonary arterial hypertension (PAH) include pulmonary vasodilators which may also inhibit PASMC proliferation. The aim of this study was to compare the antiproliferative effects of multiple drugs on rat and human PASMC (rPASMC and hPMASC, respectively) in vitro. rPASMCs and hPASMC were starved for 24 h, then treated with different inhibitors and incubated for 48 h in 1% foetal calf serum plus endothelin-1, 5-HT and U46619. To investigate retrograde delivery of cardioplegic solutions as a means of enhancing myocardial protection in the presence of coronary artery occlusion, a two-part experimental model was devised. In part 1 (in vitro) the possibility of retroperfusing the entire myocardium during acute occlusion of the left anterior descending artery (LAD) was assessed. After two-stage nephrectomy in rats the potassium concentration in the red blood corpuscles (RBC) decreases from 10.6 ± 0.3 to 5.5 ± 0.3 mEq per 100 ml of RBC within 48 h. The decrease is accompanied by a much smaller increase in the plasma potassium concentration; the hyperkaliemia in nephrectomized rats is less pronounced than in nephrectomized dogs or anuric humans. Na+ in RBC increases by about 44% after nephrectomy; while there is only a very slight decrease of Na+ in plasma.
