Cell surface display of rat invariant γ chain: detection by monoclonal antibodies directed against a C-terminal γ chain segment

A series of 14 monoclonal antibodies (mAb) directed against the C-terminal part of the rat invariant gamma chain (amino acid 142-216) was generated using distinct fusion proteins that contain this gamma segment for immunization and hybridoma screening. Additional fusion protein were prepared carrying discrete regions of the gamma chain. Employing these reagents confirmed that the obtained mAb do indeed recognize the C-terminal portion of the invariant chain, as demonstrated by Western blot analysis. Antibodies to DNA are important markers of various autoimmune diseases and can be pathogenic; however, their generation is not understood. We previously reported that anti-DNA antibodies could be induced in mice by idiotypic immunization to PAb-421, an antibody to a DNA-binding domain of p53. We now report that two monoclonal antibodies of moderate affinity (K(D) asymptotically equal to 10(-7)), raised from PAb-421-immunized mice, specifically recognized both PAb-421 and DNA. In the present study, the pattern of some extracellular matrix glycoproteins was determined in traumatic anterior cruciate ligament ruptures (n = 17) and in normal ligaments (n = 11). 6 microns cryo-sections were incubated with monoclonal antibodies recognizing tenascin, laminin, and fibronectin, and FITC-labeled secondary antibodies. Immunohistochemistry revealed that tenascin, fibronectin and laminin are sparsely distributed in normal anterior cruciate ligaments, but strongly expressed in ruptured ligaments with a time-restricted pattern. The purpose of this qualitative study was to explore in-service teachers’ first experiences with imaginary dialogues – a form of mathematical writing where students are introduced to a written and unfinished dialogue between two imaginary persons discussing a mathematical problem. Students are supposed to continue working with the problem and to complete the initial dialogue between these persons. Guanylyl cyclases (GC) catalyze the formation of the intracellular signal molecule cyclic GMP from GTP. For some years it has been known that the heme-containing soluble guanylyl cyclase (sGC) is stimulated by NO and NO-containing compounds. A differential proteomic analysis, based on 2-DE and MS procedures, was performed on Amycolatopsis balhimycina DSM5908, the actinomycete producing the vancomycin-like antibiotic balhimycin. A comparison of proteomic profiles before and during balhimycin production characterized differentially and constitutively expressed protein isoforms, which were associated to 203 ORFs in the A. balhimycina genome.
