Metabolic Escape Routes of Cancer Stem Cells and Therapeutic Opportunities

2020

Although improvement in early diagnosis and treatment ameliorated life expectancy of cancer patients, metastatic disease still lacks effective therapeutic approaches. Resistance to anticancer therapies stems from the refractoriness of a subpopulation of cancer cells—termed cancer stem cells (CSCs)—which is endowed with tumor initiation and metastasis formation potential. CSCs are heterogeneous and diverge by phenotypic, functional and metabolic perspectives. Intrinsic as well as extrinsic stimuli dictated by the tumor microenvironment (TME)have critical roles in determining cell metabolic reprogramming from glycolytic toward an oxidative phenotype and vice versa, allowing cancer cells to th…

Cancer Stem Cells and Neuroblastoma: Characteristics and Therapeutic Targeting Options

2019

The majority of embryonal tumors or childhood blastomas derive from pluripotent progenitors or fetal stem cells that acquire cancer stem cell (CSC) properties: multipotency, self-renewal ability, metastatic potential, chemoresistance, more pronounced levels of drug transporters, enhanced DNA-damage repair mechanisms, and a quiescent state. Neuroblastoma (NB) is considered a neuroendocrine tumor and is the most common extracranial neoplasm in children. NB pathogenesis has frequently been associated with epigenetic dysregulation and a failure to implement a differentiation program. The origin, characteristics, and isolation of the CSC subpopulation in NB are still incompletely understood, des…

Cell Stress, Metabolic Reprogramming, and Cancer

2018

The hallmarks of cancer comprise six biological capabilities acquired during the multistep development of human tumors: sustaining proliferative signaling, evading growth suppressors, resisting cell death, enabling replicative immortality, inducing angiogenesis, and activating invasion and metastasis (1). Mitochondria, beyond being the site of aerobic respiration, are at the crossroads of a variety of metabolic and signaling pathways resulting key regulatory organelles in cell life and death decision. Thus, it is no surprise that genomic, functional, and structural mitochondrial alterations have been associated with cancer and that mitochondria have become a pharmacological target in cancer…

Self-assembly concepts for multicompartment nanostructures.

2015

Compartmentalization is ubiquitous to many biological and artificial systems, be it for the separate storage of incompatible matter or to isolate transport processes. Advancements in the synthesis of sequential block copolymers offer a variety of tools to replicate natural design principles with tailor-made soft matter for the precise spatial separation of functionalities on multiple length scales. Here, we review recent trends in the self-assembly of amphiphilic block copolymers to multicompartment nanostructures (MCNs) under (semi-)dilute conditions, with special emphasis on ABC triblock terpolymers. The intrinsic immiscibility of connected blocks induces short-range repulsion into discre…

Processing and Mechanical Properties of Natural Fiber Reinforced Thermoplastic Starch Biocomposites

2007

Natural fiber reinforced starch polymers are processed by compression molding. Potato, sweet potato, and corn starch are used as matrices. Three types of natural fibers, namely sisal, jute, and cabuya, are used in concentrations varying from 2.5 to 12.5% w/w in the composites. Different plasticizers are used for the starch polymers, such as water and glycerol. Mechanical properties are assessed by tensile and impact tests. In both cases, improved mechanical properties are obtained at increasing fiber contents. Tensile strength appears to be markedly improved with the addition of 10% by weight of sisal fibers, while the best results for impact strength are obtained for cabuya fibers.

Oxo and Hydroxo Tantalocene Complexes:  Synthesis and Reactivity. X-ray Molecular Structures of [(η5-Cp*)TaCl(OH)(η5-C5H4SiMe3)]+Cl-, (η5-Cp*)TaCl(O)…

2005

Exposure of a THF solution of tantalocene dichloride complexes (η 5 -Cp*)TaCl 2 (η 5 -C 5 H 4 -SiMe 3 ) (1) and (η 5 -Cp*)TaCl 2 (η 5 -C 5 H 5 ) (2) to the ambient atmosphere for 12 h leads to the formation of new tantalum(V) cationic hydroxo complexes [(η 5 -Cp*)TaCl(OH)(η 5 -C 5 H 4 SiMe 3 )] + -Cl- (3) and [(η 5 -Cp*)TaCl(OH)(η 5 -C 5 H 5 )]+Cl - (4). This oxidation reaction is more rapid under oxygen atmosphere since the hydroxo complexes are obtained in better yields within only 15 min. The hydroxo complexes are easily deprotonated by base to generate oxo complexes 5 and 6, which, in turn, react with trimethylsilyl triflate, giving the corresponding cationic silylated complexes 7 and 8…

Derivatives of (-)-Isosteviol with Expanded Ring D and Various Oxygen Functionalities

2012

(–)-Isosteviol is a unique ex-chiral-pool building block that is readily available. Both functional groups are aligned in a concave manner. The methyl moiety on the backbone also points in this direction, creating a strong asymmetric environment close to these functional groups. The slightly divergent orientation of the keto and carboxy functions limits its use in the construction of supramolecular architectures as optically pure divalent building blocks. By selective transformations, ring D of (–)-isosteviol can be expanded and equipped with oxygen-containing functionalities, providing a variety of useful and rigid building blocks with defined stereochemistry.

Ethanol-Based Post-processing of Generator-Derived 68Ga Toward Kit-Type Preparation of 68Ga-Radiopharmaceuticals

2014

Post-processing by means of a cation-exchanger–based protocol is an efficient strategy for purification and concentration of generator-derived 68Ga. It ensures the removal of 68Ge before 68Ga-radiopharmaceutical preparation and high labeling yields of 68Ga-labeled radiopharmaceuticals for routine medical application. Methods: In an effort to overcome the problem associated with acetone in the currently applied method, we have investigated the feasibility of replacing it with ethanol. The purification of 68Ga from coeluted metallic impurities (68Ge4+, Fe3+, Zn2+, and Ti4+) on various cation-exchange columns has been investigated with a variety of post-processing solutions. As a proof of prin…

The Renaissance of the 68Ge/68Ga Radionuclide Generator Initiates New Developments in 68Ga Radiopharmaceutical Chemistry

2010

⁶⁸Ge/⁶⁸Ga radionuclide generators have been investigated for almost fifty years now, since the cyclotron-independent availability of positron emitting ⁶⁸Ga via the ⁶⁸Ge/⁶⁸Ga system had always attracted researches working in basic nuclear chemistry as well as radiopharmaceutical chemistry. However, it took decades and generations of research (and researchers) to finally approach a reliable level of ⁶⁸Ge/⁶⁸Ga generator designs, adequate to the modern requirements of radiometal labeling chemistry. ⁶⁸Ga radiopharmacy now is awaking from a sort of hibernation. The exciting perspective for the ⁶⁸Ge/⁶⁸Ga generator, now - more than ever, asks for systematic chemical, radiochemical, technological an…
