Obtention and characterization of resistant starch from creole faba bean (Vicia faba L. creole) as a promising functional ingredient

Abstract A procedure easy to upscale industrially aimed at obtaining resistant starch (RS) type III (RS-III) was optimized, using a native faba bean from Mexico (Vicia faba L. creole) as RS source for the first time. Pullulanase debranching treatment (6–18 enzyme units (U)/g starch; 0–27 h) and retrogradation process (−30 °C, 2 °C or 20 °C; 1–6 days) was optimized for faba beans. Optimal conditions were determined at 18 U/g for 27 h and a retrogradation process at 20 °C for 6 days. Optimised method for the analysis of phenolic compounds from caper (Capparis spinosa L.) berries and monitoring of their changes during fermentation

In this work, an ad hoc method to identify and quantify polyphenols from caper berries was developed on high-performance liquid chromatography/electrospray ionisation source/mass spectrometry (HPLC-ESI-MS). The method was applied during fermentation carried out with Lactobacillus pentosus OM13 (Trial S) and without starter (Trial C). A total of five polyphenols were identified. All samples contained high concentrations of rutin. Epicatechin was found in untreated fruits, on the contrary quercetin was detected during fermentation. Abstract Desulfurococcus amylolyticus is an anaerobic and hyperthermophilic crenarchaeon that can use various carbohydrates as energy sources. We found a gene encoding a glycoside hydrolase family 57 amylolytic enzymes (DApu) in a putative carbohydrate utilization gene cluster in the genome of D. amylolyticus . This gene has an open reading frame of 1,878 bp and consists of 626 amino acids with a molecular mass of 71 kDa. Recombinant DApu (rDApu) completely hydrolyzed pullulan to maltotriose by attacking α-1,6-glycosidic linkages, and was able to produce glucose and maltose from soluble starch and amylopectin. Abstract Isolation and identification of sterols from culture extracts of the fungus Aureobasidium (Dematium) pullulans yielded metabolites with key function in sterol synthesis. By proof of squalene, lanosterol/dihydrolanosterol, desmosterol, ergosterol, cholesterol, stigmasterol and β-sitosterol it was possible to work out a general synthesis-scheme for the building-up of sundry sterols in Ascomycetes.

Pullulan and dextran: uncommon composition dependent Flory-Huggins interaction parameters of their aqueous solutions.

Vapor pressure measurements were performed for aqueous solutions of pullulan ( M w 280 kg/mol) and dextran ( M w 60 and 2100 kg/mol, respectively) at 25, 37.5, and 50 degrees C. The Flory-Huggins interaction parameters obtained from these measurements, plus information on dilute solutions taken from the literature, show that water is a better solvent for pullulan than for dextran. Furthermore, they evince uncommon composition dependencies, including the concurrent appearance of two extrema, a minimum at moderate polymer concentration and a maximum at high polymer concentration. Large scale fractionation of pullulan and dextran

Abstract A recently developed large scale fractionation technique named continuous spin fractionation (CSF) was applied to fractionate pullulan and dextran. 450 g of pullulan with a broad molecular weight distribution were fractionated using water as solvent and acetone as precipitant. In this study, we have in five CSF runs prepared three fractions with apparent M ¯ w * values ranging from 17.6 to 413 kg mol−1. Seventy grams of dextran were fractionated with a mixed solvent of water plus methanol. Five fractionation steps resulted in four samples with M ¯ w values between 4.36 and 18.2 kg mol−1.

International audience; This work was performed to investigate on the yeast ecology of durum wheat to evaluate the interaction between kernel yeasts and the commercial baker's yeast Saccharomyces cerevisiae during dough leavening. Yeast populations were studied in 39 genotypes of durum wheat cultivated in Sicily. The highest level of kernel yeasts was 2.9 Log CFU/g. A total of 413 isolates was collected and subjected to phenotypic and genotypic characterization. Twenty-three yeast species belonging to 11 genera have been identified. Growth and macromolecular content of the dimorphic fungus Aureobasidium pullulans and the effect of hydroxyurea and other inhibitors

The growth kinetics and the macromolecular content of the yeast and ethanol-induced hyphal forms of Aureobasidium pullulans were studied. During the morphological transition from yeasts to hyphae, both the protein and RNA content decreased significantly, the mycelial form containing only 76% of the amount of protein in the yeasts, and 38% of the RNA. The DNA was the only component tested whose level increased during the transition. Among several compounds inhibiting macromolecular synthesis, only hydroxyurea showed a remarkable effect on the morphology of A. pullulans, inducing the mycelial morphology. Succinate-bonded pullulan: An efficient and reusable super-sorbent for cadmium-uptake from spiked high-hardness groundwater.

Abstract Chemically modified pullulan was evaluated for its sorption efficiency and selectivity to remove cadmium (Cd) from spiked high-hardness groundwater (GW). Pullulan esterified with succinic anhydride using dimethylaminopyridine showed a fairly high degree of substitution value as confirmed by 1 H NMR spectroscopy. Pullulan succinate (Pull-Suc) was converted into the sodium salt (Pull-Suc-Na). The effect of contact time (5–200 min) and pH (2–8) on Cd-uptake by the sorbent (Pull-Suc-Na) was investigated.
