Intra- and extra-cellular DNA damage by harmine and 9-methyl-harmine
2014
It is known that b-carbolines are able to produce photosensitized damage in cell-free DNA, but there is little information on their effects on cellular DNA. Therefore, we have analyzed the DNA damage produced by harmine and 9-methyl-harmine under UVA irradiation in V79 cells, together with the associated generation of micronuclei and photocytotoxicity. The results indicate that the most frequent photoproducts generated in the cellular DNA are modified purines such as 8-oxo-7,8-dihydroguanine. Only relatively few single-strand breaks were observed. CPDs were absent, although they were generated in cell-free DNA irradiated under the same conditions. The overall extent of DNA damage in the cel…
Experimental and theoretical studies on polar Diels–Alder reactions of 1-nitronaphathalene developed in ionic liquids
2013
The reactions between 1-nitronaphthalene 1 and two nucleophilic dienes, Danishefsky´s diene 2 and 1-trimethylsilyloxy-1,3-butadiene 3, to yield phenanthrenols 7 and 8 developed in protic and aprotic ionic liquids (ILs) with an imidazolium cation base permit to conclude that the reactions occur in better experimental conditions than in molecular solvents. The yields obtained with protic ILs are better than those observed with aprotic ILs. In this sense, the higher yields obtained with aprotic ILs, (working at 60ºC, 48 h) are comparable with the lower yields observed in protic ILs (60 ºC, 24 h). DFT calculations for the reaction of 1-nitronaphthalene 1 with diene 3, in the absence and in the …
Indole-substituted 2,4-diamino-5,8-dihydropyrido[2,3-d]pyrimidines from one-pot process and evaluation of their ability to bind dopamine receptors
2018
A series of novel 7-indole substituted 2,4-diamino-5,8-dihydropyrido[2,3-d]pyrimidine analogous to the 2,4-diaminopteridine core were synthesized by the three-component one-pot cyclocondensation between 2,4,6-triaminopyrimidine, 3-(2-cyanoacetyl)indole and aromatic aldehydes. The reactions, which exhibited good performance, proceeded in EtOH using indium (III) chloride as catalyst under microwave irradiation, in short reaction times. On the basis of certain structural similarity of these compounds with known ligands of the D2 dopamine receptors (D2DR), the study of these compounds as possible ligands of dopamine D2 and D1 receptors was carried out. Three of them showed moderate affinity to …
Silica Nanoparticles for Insect Pest Control.
2019
To date, control strategies used against insect pest species are based on synthetic insecticide applications. In addition, the efficacy of these treatments could be decreased due to insecticide resistance in insect populations. Also, the irrational use of chemical control strategies has negative consequences of non-target organisms and threatening human health. Designing nanomaterial for pest insect control is a promising alternative to traditional insecticide formulations. In particular, it has been proven that silica nanoparticles have the potential for molecules delivery, release control improvement and also their toxicity as insecticide alone. In this work, we summarized the state of kn…
Ionic liquids and microwave irradiation as synergistic combination for polar Diels–Alder reactions using properly substituted heterocycles as dienoph…
2012
For the DA reactions between nitropyrrole derivatives and isoprene, the combination of microwave irradiation and protic ionic liquid, has a notable synergistic effect. DFT calculations suggest that a supplementary energy ranking in the IR region of 1700-1400 cm-1 can favour the structural changes in the reactants to reach the TS. Fil: Mancini, Pedro Maximo Emilio. Nitropyrroles, Diels-Alder reactions assisted by microwave irradiation and solvent effect. An experimental and theoretical study
2017
The behaviors of N-tosylnitropyrroles acting as electrophilic dienophiles in polar Diels-Alder reactions joint to different dienes of increeased nucleophilicity are analyzed. The reactions were developed under microwave irradiation using toluene or protic ionic liquids (PILs) as solvents and in free solvent conditions. In all the cases explored we observed good yields in short reaction times. For these reactions, the free solvent condition and the use of protic ionic liquids as solvents offer similar results. However, the free solvent conditions favor environmental sustainability. The role of PILs in these polar Diels-Alder reactions has been theoretically studied within the Molecular Elect…
Computer-guided drug repurposing: Identification of trypanocidal activity of clofazimine, benidipine and saquinavir
2015
In spite of remarkable advances in the knowledge on Trypanosoma cruzi biology, no medications to treat Chagas disease have been approved in the last 40 years and almost 8 million people remain infected. Since the public sector and non-profit organizations play a significant role in the research efforts on Chagas disease, it is important to implement research strategies that promote translation of basic research into the clinical practice. Recent international public-private initiatives address the potential of drug repositioning (i.e. finding second or further medical uses for known-medications) which can substantially improve the success at clinical trials and the innovation in the pharmac…
One-Year Changes in Urinary Microbial Phenolic Metabolites and the Risk of Type 2 Diabetes—A Case-Control Study
2022
The intake of polyphenols has been associated with a risk reduction of type 2 diabetes. Nevertheless, to the best of our knowledge, the molecules that might be metabolically active after ingestion are only starting to be investigated regarding this metabolic disease. To investigate the association between one-year changes in urinary microbial phenolic metabolites (MPM) and the incidence of type 2 diabetes, we performed a case-control study using data and samples of the PREDIMED trial including 46 incident type 2 diabetes cases of 172 randomly selected participants. Eight urinary MPMs were quantified in urine by liquid chromatography coupled to mass spectrometry and used to assess their asso…
Light-responsive hybrid material based on luminescent core-shell quantum dots and steroidal organogel
2016
We report the synthesis of a smart novel hybrid with reversible photoswitchable luminescence properties modulated by light. The combination of a low molecular weight organogelator (LMOG) and CdSe/ZnS core-shell semiconductor nanoparticles capped with trioctylphosphine oxide ligands produces a luminescent, stable and transparent material. Modulation of the luminescence properties was successfully achieved using a diarylethene photochromic compound, with good resistance to fatigue ca. 22 cycles. Interestingly, the morphology of the organogel fibers was preserved in the hybrid, while a partial luminescence quenching of the nanoparticle was observed. This material could have implication for on-…
Fe3O4@Au@mSiO2 as an enhancing nanoplatform for Rose Bengal photodynamic activity
2017
A novel nanoplatform composed of three types of materials with different functionalities, specifically core-shell Fe3O4@Au nanoparticles encapsulated near the outer surface of mesoporous silica (mSiO2) nanoparticles, has been successfully synthesised and used to enhance the efficiency of a photosensitiser, namely Rose Bengal, in singlet oxygen generation. Fe3O4 is responsible for the unusual location of the Fe3O4@Au nanoparticle, while the plasmonic shell acts as an optical antenna. In addition, the mesoporous silica matrix firmly encapsulates Rose Bengal by chemical bonding inside the pores, thus guaranteeing its photostability, and in turn making the nanosystem biocompatible. Moreover, th…
