Assessment of polycyclic aromatic hydrocarbons (PAHs) in soil of a Natural Reserve (Isola delle Femmine) (Italy) located in front of a plant for the …

2009

Isola delle Femmine Natural Reserve is a very little isle about 15 km from the centre of Palermo, in front of a plant for the production of cement and about 600 m from coast. In the present research, profiles soil PAHs were obtained for 16 sites within the reserve and for 8 stations on the rural soil taken as reference. summation Sigma PAHs, in the soil of investigated area, ranged from 35 to 545 microg/kg. With the aim to find the origin of PAHs in the soil of Isola delle Femmine, we compare the distribution of single analytes in the investigated area with those of the reference rural area (Monte Raffo Rosso), with those of atmospheric urban particulate collected at Palermo along with repo…

Influences of heat seal lacquer thickness on the quality of blister packages.

2011

A sealability of aluminium lidding foils against formable polymer materials of blister packages is usually achieved by a coating of aluminium with certain grammages of heat seal lacquers. To investigate influences of their thickness on quality of blister packages, lidding foils with different grammages of two lacquer types were manufactured. Sealing experiments (variation of temperature, pressure and sealing time) were performed. Sealed seam strengths were determined with mechanical tensile tests, tightness of cold form blisters were analysed by means of helium leakage tests. Time-dependent moisture uptake of stored blisters was monitored with micro-gas chromatography. By means of a simple …

Factors that influence the quality of metabolomics data in in vitro cell toxicity studies: a systematic survey

2021

Abstract REACH (Registration, Evaluation, Authorization and Restriction of Chemicals) is a global strategy and regulation policy of the EU that aims to improve the protection of human health and the environment through the better and earlier identification of the intrinsic properties of chemical substances. It entered into force on 1st June 2007 (EC 1907/2006). REACH and EU policies plead for the use of robust high-throughput "omic" techniques for the in vitro investigation of the toxicity of chemicals that can provide an estimation of their hazards as well as information regarding the underlying mechanisms of toxicity. In agreement with the 3R’s principles, cultured cells are nowadays wide…

Improving Biological Dyes and Stains: Quality Testing Versus Standardization

1994

This paper discusses the impact of both standardization and quality testing of dyes and stains in biology and medicine. After the brief review of why standardized dyes and strains are not presently available commercially, two types of testing and ways of improving dye quality are described. National or international organizations could be established to define standardization of dyes and stains. Standardization would be specifically defined as a list of physico-chemical parameters such as elaborated in this paper. Commercial batches of comparable quality may be labeled by the supplier as "standard dye," a procedure currently performed by the European Council for Clinical and Laboratory Stan…

Cryopreservation of MHC Multimers: Recommendations for Quality Assurance in Detection of Antigen Specific T Cells

2015

Fluorescence-labeled peptide-MHC class I multimers serve as ideal tools for the detection of antigen-specific T cells by flow cytometry, enabling functional and phenotypical characterization of specific T cells at the single cell level. While this technique offers a number of unique advantages, MHC multimer reagents can be difficult to handle in terms of stability and quality assurance. The stability of a given fluorescence-labeled MHC multimer complex depends on both the stability of the peptide-MHC complex itself and the stability of the fluorochrome. Consequently, stability is difficult to predict and long-term storage is generally not recommended. We investigated here the possibility of…

Monitoring of chicken meat freshness by means of a colorimetric sensor array

2012

A new optoelectronic nose to monitor chicken meat ageing has been developed. It is based on 16 pigments prepared by the incorporation of different dyes (pH indicators, Lewis acids, hydrogenbonding derivatives, selective probes and natural dyes) into inorganic materials (UVM-7, silica and alumina). The colour changes of the sensor array were characteristic of chicken ageing in a modi¿ed packaging atmosphere (30% CO2¿70% N2). The chromogenic array data were processed with qualitative (PCA) and quantitative (PLS) tools. The PCA statistical analysis showed a high degree of dispersion, with nine dimensions required to explain 95% of variance. Despite this high dimensionality, a tridimensional re…

Development of extracellular vesicle-based medicinal products: A position paper of the group “Extracellular Vesicle translatiOn to clinicaL perspecti…

2021

International audience; Extracellular vesicles (EV) are emergent therapeutic effectors that have reached clinical trial investigation. To translate EV-based therapeutic to clinic, the challenge is to demonstrate quality, safety, and efficacy, as required for any medicinal product. EV research translation into medicinal products is an exciting and challenging perspective. Recent papers, provide important guidance on regulatory aspects of pharmaceutical development, defining EVs for therapeutic applications and critical considerations for the development of potency tests. In addition, the ISEV Task Force on Regulatory Affairs and Clinical Use of EV-based Therapeutics as well as the Exosomes C…

Quality control of gold nanoparticles as pharmaceutical ingredients

2019

Abstract Nanoparticles are being developed for a wide range of medical applications such as, controlled release, drug delivery systems or imagery, theranostics, implants…. For the moment, there is no legal definition of nanoparticles or nanomaterials for therapeutic use. The specific case of gold nanoparticles is not an exception: their current definition as nanoparticle material does not correspond to classic pharmaceutical ingredients as described in Pharmacopoeias. In this study, more than 30 different batches of citrate stabilized gold nanoparticles (AuNP) were synthesized and analyzed thanks to both classical approaches (UV–Vis spectrophotometry, dynamic light scattering coupled or not…

Receiving water quality assessment: comparison between simplified and detailed integrated urban modelling approaches

2010

Urban water quality management often requires use of numerical models allowing the evaluation of the cause–effect relationship between the input(s) (i.e. rainfall, pollutant concentrations on catchment surface and in sewer system) and the resulting water quality response. The conventional approach to the system (i.e. sewer system, wastewater treatment plant and receiving water body), considering each component separately, does not enable optimisation of the whole system. However, recent gains in understanding and modelling make it possible to represent the system as a whole and optimise its overall performance. Indeed, integrated urban drainage modelling is of growing interest for tools to …

Pressurized liquid extraction combined with capillary electrophoresis–mass spectrometry as an improved methodology for the determination of sulfonami…

2007

A new analytical method, based on capillary electrophoresis and tandem mass spectrometry (CE-MS2), is proposed and validated for the identification and simultaneous quantification of 12 sulfonamides (SAs) in pork meat. The studied SAs include sulfathiazole, sulfadiazine, sulfamethoxypyridazine, sulfaguanidine, sulfanilamide, sulfadimethoxyne, sulfapyridine, sulfachloropyridazine, sulfisoxazole, sulfasalazine, sulfabenzamide and sulfadimidine. Different parameters (i.e. separation buffer, sheath liquid, electrospray conditions) were optimized to obtain an adequate CE separation and high MS sensitivity. MS2 experiments using an ion trap as analyzer, operating in the selected reaction monitori…
