The role of public biological resource centers in providing a basic infrastructure for microbial research

2009

Public collections of microorganisms have been established since the late 19th century, and currently 573 service collections are registered at the World Data Center for Microorganisms (www.wdcm.org). All together, they hold more than 1.5 million microorganisms. By implementing guidelines compiled by the Organisation for Economic Co-operation and Development (OECD), many public service collections evolve into professional ex situ repositories of biodiversity and distribution nodes for known, validated and precisely identified microbial resources and associated information to legitimate end-users. These Biological Resource Centers (BRCs) may be the preferred mechanism for the appropriate exp…

Análisis de resultados del Programa de Control de Calidad Externo SEIMC de carga viral del VIH-1, VHC y VHB. AÑO 2018

2020

BACKGROUND Human immunodeficiency virus type 1 (HIV-1) and hepatitis B (HBV) and C virus (HCV) viral load determinations are among the most relevant markers for the follow-up of patients infected with these viruses. External quality control tools are crucial to ensure the accuracy of the results obtained by microbiology laboratories. This article summarised the results obtained from the 2017 SEIMC External Quality Control Programme for HIV-1, HCV, and HBV viral loads and HCV genotyping. METHODS AND RESULTS In the HIV-1 programme, a total of five standards were sent. One standard consisted of seronegative human plasma, while the remaining four contained plasma from three different viremic pa…

Mixed-model of ANOVA for measurement reproducibility in proteomics

2009

This work is a statistical analysis of reproducibility of a MALDI-TOF mass spectrometry experiment. Its aim is to evaluate measurement variability and compare peak intensities from two types of MALDI-TOF platforms. We compared and commented on the abilities of Principal Component Analysis and mixed-model analysis of variance to evaluate the biological variability and the technical variability of peak intensities in different patients. The properties and hypotheses of both methods are summarized and applied to spectra from plasma of patients with Hodgkin lymphoma. Principal Component Analysis checks rapidly the balance between the two variabilities; however, a mixed-model analysis of varianc…

Pharmacy and pharmacology of biosimilars

2008

Biosimilar medicines are biological medicinal products that can obtain a marketing authorization in the EU after the original product (biological reference medicine) has run out of patent. As a prerequisite, studies including clinical trials are to be conducted to compare the quality, safety, and efficacy of the biosimilar and reference medicine. Due to the specific characteristics of biopharmaceuticals like complex 3-dimensional (glyco) protein structure, immunogenicity, production in living organisms, which causes heterogeneity, complex manufacturing process and analysis, interchangeability of the biosimilar with its reference drug product is not guaranteed. In addition, INN (internationa…

On the internal multivariate quality control of analytical laboratories. A case study: the quality of drinking water

2001

Abstract Multivariate statistical process control (MSPC) tools, based on principal component analysis (PCA), partial least squares (PLS) regression and other regression models, are used in the present study for automatic detection of possible errors in the methods used for routine multiparametric analysis in order to design an internal Multivariate Analytical Quality Control (iMAQC) program. Such tools could notice possible failures in the analytical methods without resorting to any external reference since they use their own analytical results as a source for the diagnosis of the method's quality. Pseudo-univariate control charts provide an attractive alternative to traditional univariate …

Ythdf is a N6‐methyladenosine reader that modulates Fmr1 target mRNA selection and restricts axonal growth in Drosophila

2021

Abstract N6‐methyladenosine (m6A) regulates a variety of physiological processes through modulation of RNA metabolism. This modification is particularly enriched in the nervous system of several species, and its dysregulation has been associated with neurodevelopmental defects and neural dysfunctions. In Drosophila, loss of m6A alters fly behavior, albeit the underlying molecular mechanism and the role of m6A during nervous system development have remained elusive. Here we find that impairment of the m6A pathway leads to axonal overgrowth and misguidance at larval neuromuscular junctions as well as in the adult mushroom bodies. We identify Ythdf as the main m6A reader in the nervous system,…

Noise identification and sampling frequency determination for precise Pb isotopic measurements by quadrupole-based Inductively Coupled Plasma Mass Sp…

2000

Analytical precision of the isotope ratios measured by quadrupole-based ICP-MS is drastically controlled by the low- frequency noises which originate from nebulisation and vaporisation processes, and from sample introduction systems. The unde- sirable influence of these latter can be however reduced by choosing efficiently the operating parameters. In the present study , the choice of the stabilisation time necessary in peak jump mode, and the one of the number of sweeps are discussed in the light of noise power spectra obtained with various speeds of the peristaltic pump used as sample introduction system. Obviously, the settings proposed are probably efficient only on our own ICP-MS, but …

Towards the ultimate minimum particle diameter of silica packings in capillary electrochromatography.

2000

Porous silica beads with an average particle diameter between 0.2 and 3 microm have been applied as packing material in capillary electrochromatography (CEC). The experiments were directed to investigate whether it is really feasible and as promising as expected to use such small particles. In CEC, plate heights of H approximately/= 1-2 d(p) can be achieved which is smaller than the plate heights usually attained in high-performance liquid chromatography. Using a capillary packed with 0.5 microm silica beads we achieved a plate height of H = 3 d(p) indicating the presence of dispersive effects like Joule heating. Calculations demonstrate that at a field strength of about 800 V cm(-1) one th…

An enhanced memetic differential evolution in filter design for defect detection in paper production.

2008

This article proposes an Enhanced Memetic Differential Evolution (EMDE) for designing digital filters which aim at detecting defects of the paper produced during an industrial process. Defect detection is handled by means of two Gabor filters and their design is performed by the EMDE. The EMDE is a novel adaptive evolutionary algorithm which combines the powerful explorative features of Differential Evolution with the exploitative features of three local search algorithms employing different pivot rules and neighborhood generating functions. These local search algorithms are the Hooke Jeeves Algorithm, a Stochastic Local Search, and Simulated Annealing. The local search algorithms are adap…
