Hydroquinone: O-glucosyltransferase from cultivated Rauvolfia cells: enrichment and partial amino acid sequences.

Plant cell suspension cultures of Rauvolfia are able to produce a high amount of arbutin by glucosylation of exogenously added hydroquinone. A four step purification procedure using anion exchange, hydrophobic interaction, hydroxyapatite-chromatography and chromatofocusing delivered in a yield of 0.5%, an approximately 390 fold enrichment of the involved glucosyltransferase. SDS-PAGE showed a M(r) for the enzyme of 52 kDa. Proteolysis of the pure enzyme with endoproteinase LysC revealed six peptide fragments with 9-23 amino acids which were sequenced. Sequence alignment of the six peptides showed high homologies to glycosyltransferases from other higher plants.

Mboat7 down-regulation by hyper-insulinemia induces fat accumulation in hepatocytes.

Background: Naturally occurring variation in Membrane-bound O-acyltransferase domain-containing 7 (MBOAT7), encoding for an enzyme involved in phosphatidylinositol acyl-chain remodelling, has been associated with fatty liver and hepatic disorders. Here, we examined the relationship between hepatic Mboat7 down-regulation and fat accumulation. Methods: Hepatic MBOAT7 expression was surveyed in 119 obese individuals and in experimental models. MBOAT7 was acutely silenced by antisense oligonucleotides in C57Bl/6 mice, and by CRISPR/Cas9 in HepG2 hepatocytes. E' mostrata l'applicazione di un sistema combinato di letture strumentali finalizzate all'ottenimento di una diagnosi attraverso un approccio dall'elevato rigore scientifico. Le analisi strumentali sono svolte nella chiesa di Sant'Anna a Palermo. The graphs show the application of a combined system of instrumental readings aimed to obtain a diagnosis through a high scientific and rigorous approach. The instrumental tests are executed in the St. Anne church in Palermo.

Anderson–Fabry disease (AFD) is a rare disease with an incidenceof approximately 1:117,000 male births. Lysosomal accumulation of globotriaosylceramide (Gb3) is the element characterizing Fabry disease due to a hereditary deficiency α-galactosidase A (GLA) enzyme. The accumulation of Gb3 causes lysosomal dysfunction that compromises cell signaling pathways. Deposition of sphingolipids occurs in the autonomic nervous system, dorsal root ganglia, kidney epithelial cells, vascular system cells, and myocardial cells, resulting in organ failure. Promoter architecture and transcriptional regulation of Abf1-dependent ribosomal protein genes inSaccharomyces cerevisiae

In Saccharomyces cerevisiae, ribosomal protein gene (RPG) promoters display binding sites for either Rap1 or Abf1 transcription factors. Unlike Rap1-associated promoters, the small cohort of Abf1-dependent RPGs (Abf1-RPGs) has not been extensively investigated. We show that RPL3, RPL4B, RPP1A, RPS22B and RPS28A/B share a common promoter architecture, with an Abf1 site upstream of a conserved element matching the sequence recognized by Fhl1, a transcription factor which together with Ifh1 orchestrates Rap1-associated RPG regulation. Trapani ha avviato un processo di profondo rinnovamento che in un breve arco di tempo ha visto la riqualificazione del porto, del centro storico e della città, riverberandosi anche sul suo territorio. Il processo ha preso avvio in occasione dello svolgimento nel porto di Trapani delle regate dell’America’s Cup nel 2005/06, per proseguire e radicarsi nel territorio grazie ad una programmazione integrata tra più soggetti della pubblica amministrazione, alla capacità di attrarre investimenti esogeni e al coinvolgimento dei privati e della popolazione locale. In this study we investigated the in vitro behaviour, morphostructure and extracellular matrix synthesis of human dental follicular stem cells (hDFSCs) isolated from human dental bud, which resulted to be positive for mesenchymal markers (CD29, CD90, CD146 and CD166) by FACS analysis. Cells were analysed by light and electronic microscopy to evaluate their biological response either at week 1, that is before differentiation, or at weeks 3–6, when they had been cultured in osteogenic medium onto a highly porous natural scaffold material (Bio-Oss®). Green, blue and violet colours of Lycaenidae examined by us — with the exception of Rapala arata (Theclinae) — are due to iridescent scales which are arranged before a dark background. The iridescent scales are constructed according to the Urania-type and may be diverted from pigmented scales by laying in equidistant lamellae and little cuticular bars, which maintain the distance between the lamellae. The ribs in the longitudinal axis of the scales contain lamellae and bars only in the Plebejinae, in Lycaeninae and Theclinae they are small and sit on the plain upperside of the scale with a narrow base.
