The deubiquitinase USP11 is a versatile and conserved regulator of autophagy

Autophagy is a major cellular quality control system responsible for the degradation of proteins and organelles in response to stress and damage to maintain homeostasis. Ubiquitination of autophagy-related proteins or regulatory components is important for the precise control of autophagy pathways. Here, we show that the deubiquitinase ubiquitin-specific protease 11 (USP11) restricts autophagy and that KO of USP11 in mammalian cells results in elevated autophagic flux. We also demonstrate that depletion of the USP11 homolog H34C03.2 in Caenorhabditis elegans triggers hyperactivation of autophagy and protects the animals against human amyloid-β peptide 42 aggregation-induced paralysis. Growth and reproduction data of Plesionika narval (Decapoda, Caridea, Pandalidae) off the Island of Ustica (southern Tyrrhenian Sea)

[We here report some data on the biology and population structure of Plesionika narval (Fabricius, 1787) captured along the coast of the island of Ustica in the southern Tyrrhenian Sea. Specimens were collected monthly from January to October 2000 using commercial traps at depths ranging from 15 to 100 metres. The carapace length (CL) of females ranged between 7.0 and 26.0 mm, and between 7.0 and 17.0 mm for males. Three modes were identified in the cumulative frequency distribution for females but only two for males. In females, the third modal value was observed only at the deeper sample sites. The kinetics of transport of a new fluoroquinolone antibiotic (CNV97100) and its analogs were characterized using the Caco-2 cell culture model. Unidirectional permeabilities of these analogs were greater (p < 0.05) than that of ciprofloxacin. The absorptive permeabilities (P(AB)) of 4'-N-substituted analogs (CNV97101-104) were 400-600% greater, whereas the secretory permeability (P(BA)) was 25-80% greater than unsubstituted analogs because CNV97101-104 were poor substrates for efflux transporters (efflux ratio approximately 1). No evidence for efficacy of intrathecal verapamil in the treatment of tonic-clonic status epilepticus

In two patients with refractory generalized tonic-clonic status epilepticus, 15 mg of the calcium antagonist verapamil was given by suboccipital intrathecal administration. During a 2–6-h observation period, seizure frequency did not decrease. However, in one patient, verapamil induced severe hypotension. Subsequent thiopental anesthesia suppressed convulsions immediately. Our results indicate that, in contradistinction to animal studies with different administration techniques, intrathecal administration of verapamil does not produce any anticonvulsant effect in humans.

Oncogenic HPV types are major human carcinogens. Under hypoxia, HPV-positive cancer cells can repress the viral E6/E7 oncogenes and induce a reversible growth arrest. This response could contribute to therapy resistance, immune evasion, and tumor recurrence upon reoxygenation. Here, we uncover evidence that HPV oncogene repression is mediated by hypoxia-induced activation of canonical PI3K/mTORC2/AKT signaling. Effects of the organic calcium antagonists verapamil and flunarizine on pentylenetetrazol induced paroxysmal depolarizations were tested in organotypic neocortical explants taken from neonatal rats. In these in vitro experiments the papaverin derivative verapamil depressed, and finally abolished, epileptic discharges in all cases. The piperazine derivative flunarizine, however, which is known to suppress epileptic discharges in hippocampal CA3 neurons (Bingmann and Speckmann 1986), showed no significant antiepileptic effects in the explanted neocortical neurons. One potential alternative material to replace hydroxyapatite (HAp) as a coating material for plasma-sprayed coatings on implants for hip replacement is fluorapatite (FAp). FAp has advantages over HAp regarding the capability of being chemically stable during the coating process.
