Comparison of the genotoxic and apoptosis-inducing properties of ganciclovir and penciclovir in Chinese hamster ovary cells transfected with the thym…

2000

We studied the genotoxic and apoptosis-inducing properties of ganciclovir (GCV) and penciclovir (PCV) using Chinese hamster ovary cells stably transfected with the thymidine kinase (tk) gene of herpes simplex virus-1 (HSV-1). Cells expressing HSVtk were 300 and 100 times more sensitive than their isogenic HSVtk- counterparts to the cytotoxic effects of GCV and PCV, respectively. Using radiolabeled drugs, GCV was found to be incorporated into the genomic DNA much more effectively than PCV. GCV was highly potent in inducing chromosomal aberrations compared with PCV, which provoked less sister chromatid exchanges and chromosomal changes using equimolar or equitoxic doses. For both agents, apop…

Comparative analysis of DNA breakage, chromosomal aberrations and apoptosis induced by the anti-herpes purine nucleoside analogues aciclovir, gancicl…

2002

Nucleoside analogues have been used in antiviral therapy and suicide cancer gene therapy. Therefore, it is of importance to compare their potential cytotoxic and genotoxic action. Using metabolically competent CHO cells expressing the thymidine kinase gene of herpes simplex virus type 1 (CHO-HSVtk cells) as a model system, the induction of DNA breaks was compared with the induction of structural chromosomal aberrations and apoptosis/necrosis after exposure to the anti-herpes nucleoside analogues aciclovir (ACV), ganciclovir (GCV) and penciclovir (PCV). After continuous treatment of CHO-HSVtk cells with the drugs, LD(10) in a colony-forming assay was 50, 0.5 and 1 microM for ACV, GCV and PCV…

The role of red yeast rice (RYR) supplementation in plasma cholesterol control: A review and expert opinion.

2019

1. Preamble : Hypercholesterolemia is a major risk factor for atherosclerotic cardiovascular disease (ASCVD) [1]. Increased levels of low density lipoprotein cholesterol (LDL-C) are associated with an increased risk of coronary heart disease (CHD) and many clinical trials have shown that reducing LDL-C levels significantly reduced the CHD and CVD risk [[2], [3], [4], [5]]. Thus LDL-C-lowering is the main approach for the management of cardiovascular disease. Current guidelines suggest LDL-C levels targets based on the individual CV risk; such targets can be achieved by several means, which include both lifestyle changes and pharmacological approaches [6], with statins being the cornerstone …

Prevention of Upper Gastrointestinal Bleeding in Cirrhotic Patients

1987

The prevalence of varices in patients with cirrhosis is stated to be about 50% and the risk of variceal bleeding 40% with mortality ranging from 30% to 60%. Differences may be due to patient selection and diagnostic criteria. The death risk of first bleeding seems to be higher than that of subsequent episodes (Christensen et al. 1981; D’Amico et al. 1986), indicating that the first bleeding episode causes a selection.

β1-Integrin Cytoplasmic Subdomains Involved in Dominant Negative Function

1998

The beta1-integrin cytoplasmic domain consists of a membrane proximal subdomain common to the four known isoforms ("common" region) and a distal subdomain specific for each isoform ("variable" region). To investigate in detail the role of these subdomains in integrin-dependent cellular functions, we used beta1A and beta1B isoforms as well as four mutants lacking the entire cytoplasmic domain (beta1TR), the variable region (beta1COM), or the common region (beta1 deltaCOM-B and beta1 deltaCOM-A). By expressing these constructs in Chinese hamster ovary and beta1 integrin-deficient GD25 cells (Wennerberg et al., J Cell Biol 132, 227-238, 1996), we show that beta1B, beta1COM, beta1 deltaCOM-B, a…

Une étude exploratoire des représentations de la gratuité et de ses effets sur le comportement des publics des musées et des monuments

2007

What perceptions of free admission to museums and monuments do French visiting and non-visiting audiences have ? What are the consequences of such perceptions on individuals' perceptions, visit-planning and behavior patterns in museums and monuments ? Research conducted between 2002 and 2003 tried to answer these questions by multiangulation applied to data production and analysis. Results show that free admission alter the perceptions of museums and monuments, it is secondary in planning and implementation of visit-planning, and finally it can spark a learning process from the visit that results in the appropriation of a free admission scheme. Conclusions are related to the need to create …

A Realistic Model to Support Rescue Operations After an Earthquake via UAVs

2022

In this paper, we consider the problem of completely flying over an area just hit by an earthquake with a fleet of Unmanned Aerial Vehicles (UAVs) to opportunely direct rescue teams. The cooperation between UAVs ensures that the search for possible survivors can be faster and more effective than the solutions currently implemented by civil protection. To study this scenario, we introduce the Cover by Multitrips with Priorities (CMP) problem, which tries to keep into account all the main real-life issues connected to the flight and coordination of the UAVs. We conduct a theoretical study to estimate the best number of UAVs and additional batteries, to give indications to the organization tha…

Circular law for sparse random regular digraphs

2020

Fix a constant $C\geq 1$ and let $d=d(n)$ satisfy $d\leq \ln^{C} n$ for every large integer $n$. Denote by $A_n$ the adjacency matrix of a uniform random directed $d$-regular graph on $n$ vertices. We show that, as long as $d\to\infty$ with $n$, the empirical spectral distribution of appropriately rescaled matrix $A_n$ converges weakly in probability to the circular law. This result, together with an earlier work of Cook, completely settles the problem of weak convergence of the empirical distribution in directed $d$-regular setting with the degree tending to infinity. As a crucial element of our proof, we develop a technique of bounding intermediate singular values of $A_n$ based on studyi…

Isozyme uniformity in a wild extinct insular plant,Lysimachia minoricensisJ.J. Rodr. (Primulaceae)

1999

Isozyme analyses were conducted to evaluate the genetic diversity of seed accessions of Lysimachia minoricensis (a Balearic endemic plant that became extinct in the wild) provided by 10 European botanical gardens. No isozyme variation was detected after examination of the electrophoretic patterns of 22 putative loci in more than 150 plants. The lack of genetic variation in L. minoricensis is probably due to the shortage of sample propagules originally recovered in the field before its extinction. Extant plants of L. minoricensis are believed to have originated from a single source, therefore limiting their use as seed stocks for restoration projects.
