Preparation of hepatitis C virus structural and non-structural protein fragments and studies of their immunogenicity

Abstract Plasmids pQE-60 and pQE-30 containing 6× His-tag sequence were used for expression of fragments of HCV structural and non-structural proteins in Escherichia coli (E. coli). The following fragments were used: core (1–98 aa), NS3 (202–482 aa), and tetramer of hypervariable region 1 (HVR1) of E2 protein. The constructed plasmids directed high levels of expression of HCV proteins in E. coli JM109. After purification by the metal-affinity chromatography on nickel–nitrilotriacetic acid (Ni–NTA) agarose, the His-tagged HCV proteins were used for immunization of BALB/c mice. A review on some discrete variational techniques for the approximation of essential boundary conditions

We review different techniques to enforce essential boundary conditions, such as the (nonhomogeneous) Dirichlet boundary condition, within a discrete variational framework, and especially techniques that allow to account for them in a weak sense. Those are of special interest for discretizations such as geometrically unfitted finite elements or high order methods, for instance. Some of them remain primal, and add extra terms in the discrete weak form without adding a new unknown: this is the case of the boundary penalty and Nitsche techniques. MAST-2D diffusive model for flood prediction on domains with triangular Delaunay unstructured meshes

Abstract A new methodology for the solution of the 2D diffusive shallow water equations over Delaunay unstructured triangular meshes is presented. Before developing the new algorithm, the following question is addressed: it is worth developing and using a simplified shallow water model, when well established algorithms for the solution of the complete one do exist? In-depth evaluation of software tools for data-independent acquisition based label-free quantification.

Label-free quantification (LFQ) based on data-independent acquisition workflows currently experiences increasing popularity. Several software tools have been recently published or are commercially available. Morphological determination of the phototrophic community composition of biological soil crusts in coastal sand dunes in northern Germany

This dataset comprises the microbial community composition of biological soil crusts in north-German sand dunes. For this we obtained enrichment cultures of phototrophic microorganisms, by placing fragments of biocrusts of the same Petri dishes as used for sequencing, in Petri dishes with Bold Basal (1N BBM) agarized medium (Bischoff and Bold 1963). Cultures were grown under standard laboratory conditions: with a 12-hour alteration of light and dark phases and irradiation of 25 μmol photons m-2 s-1 at a temperature 20 ± 5 ºС. Phytoplankton strategies and diversity under different nutrient levels and planktivorous fish densities in a shallow Mediterranean lake

Two mesocosm experiments were carried out to investigate the dynamic effects of nutrients (nitrogen and phosphorus) and planktivorous fish additions on phytoplankton strategies and diversity. The phylogenetic and functional approaches were used to understand phytoplankton ecology in shallow Mediterranean lakes. The experimental approach is new for the study of algal functional groups. Nutrient loading and fish stocks enhanced biomass of small algae but decreased phytoplankton diversity and species richness.
