How DNA lesions are turned into powerful killing structures: Insights from UV-induced apoptosis

Mammalian cells treated with ultraviolet (UV) light provide one of the best-known experimental systems for depicting the biological consequences of DNA damage. UV irradiation induces the formation of DNA photoproducts, mainly cyclobutane pyrimidine dimers (CPDs) and (6-4) pyrimidine-pyrimidone photoproducts [(6-4)PPs], that drastically impairs DNA metabolism, culminating in the induction of cell death by apoptosis. While CPDs are the most important apoptosis-inducing lesions in DNA repair proficient cells, recent data indicates that (6-4)PPs also signals for apoptosis in DNA repair deficient cells. Apurinic/apyrimidinic endonuclease (APE alias Ref-1) is a key enzyme in the base excision repair pathway. Besides its function in DNA repair, APE serves to maintain several transcription factors in an active reduced state such as c-Fos, c-Jun, NF-kappaB, p53 and HIF-1alpha, all of which have been shown to play a role in tumorigenesis. A novel spirochaete, Treponema sp. strain SPIT5T, was isolated from hindgut contents of the drywood termite Incisitermes tabogae (Snyder). The cells of strain SPIT5T were motile, helical in shape, 0.4-0.5 microm in diameter and generally 12-20 microm long. The strain is obligately anaerobic and ferments different mono-, di- and oligosaccharides by forming ethanol as the main liquid fermentation end product. Furthermore, strain SPIT5T was able to grow anaerobically with yeast extract as sole carbon and energy source. International audience; Collective dance improvisation (e.g., traditional and social dancing, contact improvisation) is a participatory, relational and embodied art form which eschews standard concepts in aesthetics. We present our ongoing research into the mechanisms underlying the lived experience of "togetherness" associated with such practices. Togetherness in collective dance improvisation is kinaesthetic (based on movement and its perception), and so can be simultaneously addressed from the perspective of the performers and the spectators, and be measured.
