New 4-substituted acyloxyproline derivatives with different hydrophobic properties of the acyl group were easily synthesized and used as catalysts in the direct asymmetric aldol reaction between cyclic ketones (cyclohexanone and cyclopentanone) and several substituted benzaldehydes. Reactions were carried out using water, this being the best reaction medium examined. Screening of these catalysts showed that compounds bearing the most hydrophobic acyl chains [4-phenylbutanoate and 4-(pyren-1-yl)butanoate] provided better results. Rearrangement under acidic conditions of the pyrrolylbenzotriazinone 7 afforded the pyrrolylbenzamides 10 and 11. By thermal rearrangement instead, the first fully aromatic derivative of the pyrrolo[3,4-c]isoquinoline ring system 9 was obtained.

Abstract In this report we will describe the preparation and the biological activity of a novel class of heterocyclic arotinoids endowed with potent cytotoxic and apoptotic acitivity. Structure­activity relationship studies revealed that the different stereochemistry at the C9 double bond of retinoids seems associated with a different biological activity: potent apoptotic activity for the cis-isomers, whereas differentiating activity for the trans structures. An interesting modified Wittig procedure that allows easily to arotinoids will also be described. Versatile oxidative approach to carbazoles and related compounds using MoCl5.

The unique oxidizing power of molybdenum pentachloride provides an easy to perform, versatile, and high yielding method to construct carbazoles and the corresponding dibenzo analogues of thiophene, furan, and selenophene. The coupling reaction tolerates a variety of functional groups. The synthesis is highly modular. By this approach a precursor for the naturally occurring carbazole koenigicine was prepared.

Ab Initio Study of Stereo- and Regioselectivity in the Diels−Alder Reaction between 2-Phenylcyclopentadiene and α-(Methylthio)acrylonitrile

Ab initio molecular orbital calculations have been used to investigate the Diels−Alder reaction between 2-phenylcyclopentadiene and α-(methylthio)acrylonitrile. Geometry optimizations were performed with the RHF/3-21G, RHF/3-21G*, and RHF/6-31G* calculations while the correlation energy has been calculated at MP2 and MP3 levels. Four asynchronous transition structures corresponding to the formation of different stereoisomers and regioisomers associated with the four reaction channels have been located. The condensation reaction under acid condition of the benzylic alcohols 1, 2 and 3 with the hexadeutero dimethoxybenzenes 4, 5 and 6 leads to the expected hexadeutero bis(dimethoxyphenyl)-2-methylpropanes 7, 8 and 9, respectively. However, the presence of both dodecadeutero and unlabelled 1-bis(2, 4-dimethoxyphenyl)-2-methylpropanes 10 and 11 indicates that 9 undergoes a rapid degenerate dealkylation-alkylation reaction.

Ring-Opened Analogues of Serotonine, II: Synthesis of 3-(2-Aminophenyl)propanamine and 1-(Piperidin-1-yl)-3-(2-aminophenyl)propan-3-one and Their Derivatives The synthesis of the title compounds was achieved by Mannich reaction of suitably substituted acetophenones. The preparation of 3-(2-aminophenyl)propanamine and its derivatives requires complete hydrogenation of the carbonyl group of the Mannich bases.

Enantioselective LaIII-pyBOX-Catalyzed Nitro-Michael Addition to (E)-2-Azachalcones

A [La(OTf)3] complex with a new pyBOX ligand bearing a bulky 1-naphthylmethyl substituent at the 4′-position of the oxazoline ring catalyzes the conjugate addition of nitroalkanes to a broad range of (E)-2-azachalcones, providing the expected nitro-Michael products with good yields and enantiomeric excesses up to 87 %. The optical purity of the products can be increased by a single crystallization. A plausible stereochemical model to account for the observed stereochemistry has been proposed.

A DFT study of the [3 + 2] versus [4 + 2] cycloaddition reactions of 1,5,6-trimethylpyrazinium-3-olate with methyl methacrylate.

The reaction between 1,5,6-trimethylpyrazinium-3-olate and methyl methacrylate (MMA) yielding a lactone-lactam has been studied using the DFT method at the B3LYP/6-31G(d) level. It is concluded that formation of the lactone-lactam is a domino process involving three consecutive reactions: (i) a 1,3-dipolar cycloaddition (13DC) reaction between the pyrazinium-3-olate and MMA yielding a [3 + 2] cycloadduct (CA); (ii) a skeletal rearrangement, which converts the [3 + 2] CA into a formal [4 + 2] CA, possessing a diazabicyclo[2.2.2]octane structure; and finally, (iii) an S(N)2 reaction, promoted by halide anion, with concomitant nucleophilic attack of the created carboxylate anion on an iminium …

Convenient Synthesis of 3-Cinnamoyl-2-styrylchromones: Reinvestigation of the Baker-Venkataraman Rearrangement

An efficient and straightforward, one-pot sequence gives access to highly functionalized 3-cinnamoyl-2-styrylchromones in excellent yields. The low solubility of the target molecules allows convenient isolation. The formation of an α,α-dicinnamoylated acetophenone, as a consequence of a two-fold Baker-Venkataraman sequence, has to be anticipated.
