tBuLi-Mediated One-Pot Direct Highly Selective Cross-Coupling of Two Distinct Aryl Bromides.

A Pd-catalyzed direct cross-coupling of two distinct aryl bromides mediated by tBuLi is described. The use of [Pd-PEPPSI-IPr] or [Pd-PEPPSI-IPent] as catalyst allows for the efficient one-pot synthesis of unsymmetrical biaryls at room temperature. The key for this selective cross-coupling is the use of an ortho-substituted bromide that undergoes lithium-halogen exchange preferentially.

Abstract Paths to the ligands (Cl/Br-CH2CH2CH2)2NR/O/S/Se, R Me, Bz, iPr, iBu, to the diGrignard reagents (Cl/BrMg-CH2CH2CH2)2NR/O/S, R Me, Bz, iPr, iBu and to the eight-membered group 15 heterocycles (Cl/Br/I-As/Sb/Bi(CH2CH2CH2)2NR/S, R Me, Bz, iBu are given. 13C-NMR, IR and Raman spectra are discussed; for the compound ClSb(CH2CH2CH2)2NMe the crystal structure analysis is given : the antimony atom is clearly Lewis acidic with ψ-trigonal-bipyramidal coordination and a transannular Sb⋯N interaction of 2.385(2) A. For such a configuration the values of the 13C-NMR chemical shifts of the α-CH2-groups (e.g. Cl-As/Sb/Bi(αCH2CH2CH2)2NMe 34.6, 25.9, 46.6 ppm) display clearly a sequence N > …

Palladium-catalyzed C--C bond formation involving secondary sp3-hybridized carbon is described. These reactions occur with secondary 1-bromoethyl arylsulfoxides and different arylboronic acids, to produce the corresponding arylated sulfoxides in moderate to high yields and with complete stereospecificity. Despite the presence of beta hydrogens in the substrate, the competitive beta-hydride elimination is not a significant side reaction when coordinating solvents are used. The reported cross-coupling involves secondary C(sp3)--C(sp2) bond formation: this is the first time that a mechanistic study has been carried out with such substrates. The efficient one-pot synthesis of several new tricyclic systems of type 1 and 2, obtained from the reaction of substituted 2-amino-3-cyanopyrroles and 3-amino-4-cyanopyrroles with BMMAs, is reported. The duration and yields of the reaction strongly depend on the reactivity of the starting pyrrole and on the size of the ring to be formed. Mechanist features of the reaction were investigated and proposed by studying also the reactivity of a 3-aminopyrrole-2,4-dicyano substituted. (R)-(+)-nor-Roefractine (1) was synthesized by the Bischler-Napieralski route, using asymmetric reduction of the 1, 2-didehydro precursor imine with sodium (S)-N-CBZ-prolinyloxyborohydride. Compound 1 was able to displace [3H]-raclopride (a D2 dopamine receptor-selective ligand) from its specific binding sites in rat striatum with selectivity vs [3H]-SCH23390 (D1 dopamine receptor-selective ligand).

Abstract In this work composite polymeric films or sponges, based on hyaluronic acid (HA) covalently crosslinked with α,β-poly(N-2-hydroxyethyl)(2-aminoethylcarbamate)- d , l -aspartamide (PE), have been prepared and characterized as local gene delivery systems. In particular, HA/PE scaffolds have been loaded with PE/DNA interpolyelectrolyte complexes, employing PE as a macromolecular crosslinker for HA and as a non-viral vector for DNA. In vitro studies showed that HA/PE films and sponges have high compatibility with human dermal fibroblasts and they give a sustained DNA release, whose trend can be easily tailored by varying the crosslinking ratio between HA and PE. Abstract The diastereoselectivity of the addition of several organometallic reagents to the carbonyl group of protected D - and L -erythrulose derivatives has been investigated. Tertiary alcohols are stereoselectively formed, the diastereomeric ratio being markedly dependent on the reagent type, solvent and temperature.

Abstract A new time-dependent Monte Carlo algorithm was developed to simulate isobar and isotherm three-phase batch hydrogenation of 2,4-dinitro-toluene on Pd/C catalysts. A new reaction mechanism was formulated, involving 9 and 27 toluene derivatives, in solution and adsorbed on the surface, respectively. In fact, three different ways of adsorption were considered for all surface derivatives. Hydrogels for potential colon drug release by thiol-ene conjugate addition of a new inulin derivative.

Inulin was chosen as a starting polymer for biocompatible, pH-sensitive and biodegradable hydrogels. Three INUDVSA-TT hydrogels were obtained by crosslinking inulin derivatives with trimethylolpropane tris(3-mercaptopropionate) under varying conditions. The resulting hydrogels were cell compatible, as demonstrated by MTS and trypan blue exclusion assays acting on Caco-2 cells, and were biodegraded by inulinase and esterase, thus suggesting their use as colonic drug delivery systems. 2-Methoxyestradiol, an anti-cancer drug, was soaked in INUDVSA-TT hydrogels and its in vitro release and apoptotic effect on Caco-2 cells were evaluated.

Supercritical carbon dioxide (scCO2) is an efficient reaction medium to perform the Baeyer-Villiger oxidation with hydrated silica-supported potassium peroxomonosulfate (h-SiO2.KHSO5) under flow-through conditions. Hydration modulates the reactivity of the active surface by softening the acidity of the KHSO4 present in the supported reagent. The reaction in scCO2 is much more efficient than in n-hexane under similar conditions, which is attributed to better transport and solvating properties of the supercritical medium with regard to n-hexane.
