A novel and selective fluoride opening of aziridines by XtalFluor-E. synthesis of fluorinated diamino acid derivatives.

The selective introduction of fluorine onto the skeleton of an aminocyclopentane or cyclohexane carboxylate has been developed through a novel and efficient fluoride opening of an activated aziridine ring with XtalFluor-E. The reaction proceeded through a stereoselective aziridination of the olefinic bond of a bicyclic lactam and regioselective aziridine ring opening with difluorosulfiliminium tetrafluoroborate with the neighboring group assistance of the sulfonamide moiety to yield fluorinated diamino acid derivatives. The method based on the selective aziridine opening by fluoride has been generalized to afford access to mono- or bicyclic fluorinated substances.

Abstract The role of the trifluoromethyl group in reactivity, regio-, and stereoselectivity in cycloaddition reactions has been analyzed by studying the 1,3-dipolar cycloaddition (13DC) reactions of nitrone 4 with 3,3,3-trifluoro-1-nitropropene 7b and 3,3,3-trifluoro-2-nitropropene 14 using density functional theory (DFT) methods at the B3LYP/6-311G(d) level. The recent M06-2X functional has also been evaluated but, although the thermodynamic performance is improved, the kinetic data are not necessarily better than those obtained by the popular B3LYP functional for cycloaddition reactions. 4-(1-Cycloalken-1-yl)-1,2-dialkoxybenzenes 8, 10, and 11 and 5-(1-cycloalken-1-yl)-1,3-benzodioxoles 12−15 react with the semisquaric halides 5a and 5b in a dehydrative annulation process to give the annulated α-halobenzocyclobutenones 9a,b and 16a,b−21a,b in poor to good yields (20−76%). The reaction failed with alicyclic−aromatic dienes having no or only one alkoxy group in the benzene ring. The dehydrative annulation process could be extended to 4-(3,4-dimethoxyphenyl)-1,2-dihydronaphthalene (24), affording the highly annulated α-chlorobenzocyclobutenone 25 in 50% yield. Synthesis of Benzo[b]thioxanthenes Benzo[b]thioxanthenes, heterocyclic compounds related to tetracenes and tetracyclines can be obtained by the reaction of 2H-benzo[b]thiete (1) and 1,4-naphthoquinones (3a–g). The primary cycloadducts 4a–g undergo an autoxidation process leading to the quinones 6a–g. The dihydroxy compound 4e shows an additional isomerization by a tetrafold H transfer (4e 5e′). Another preparative route to benzo[b]thioxanthenes makes use of the cycloadditon reaction of 1 and 1,4-epoxynaphthalenes (7a–d). The selective bromination with copper(II) bromide of the pyrrole ring in 2-(2'-hydroxybenzoyl)pyrrole (II) in the heterogeneous phase is des- cribed giving in almost quantitative yield the 4,5-dibromo derivative (VI). The subsequent introduction of halogen into the phenol moiety was observed when the reaction was perfomed in the homogeneous phase with an excess of halogenating agent.
