Role of Levo-tetrahydropalmatine and its metabolites for management of chronic pain and opioid use disorders.

2021

Abstract Background Opioids have been prescribed to reduce suffering from pain and to enhance quality of life. Due to the addictive potential and the lack of other effective alternatives to treat severe acute and chronic pains, opioids remain a serious public health issue. While, opioids directly influence the drug-seeking behavior, tolerance and withdrawal processes, through neuroadaptation, the brain's endogenous opioid system also adapts in the presence of chronic pain and could contribute to the difficulty of treatment. Despite the seemingly obvious interaction between the presence of pain and opioid-abuse, little is known about the underlying mechanisms in the brain. Purpose To review …

Impact of adolescent methamphetamine use on social cognition: A human-mice reverse translation study.

2021

Background: Methamphetamine dependence is associated with social cognition deficits that may underpin negative social outcomes. However, there are considerable inter-individual differences in social cognition within people with methamphetamine dependence, with age of onset of methamphetamine use being a potential contributing factor. Materials and methods: We conducted two sequential studies examining the link between age of onset of methamphetamine use (adolescence versus young adulthood) and performance in social cognition tests: (1) a human cross-sectional study in 95 participants with methamphetamine dependence varying in age of onset (38 with adolescent onset and 57 with adult onset) a…

The embodiment of wastewater data for the estimation of illicit drug consumption in Spain

2021

Data obtained from wastewater analysis can provide rapid and complementary insights in illicit drug consumption at community level. Within Europe, Spain is an important country of transit of both cocaine and cannabis. The quantity of seized drugs and prevalence of their use rank Spain at the top of Europe. Hence, the implementation of a wastewater monitoring program at national level would help to get better understanding of spatial differences and trends in use of illicit drugs. In this study, a national wastewater campaign was performed for the first time to get more insight on the consumption of illicit drugs within Spain. The 13 Spanish cities monitored cover approximately 6 million inh…

Co-Citation Analysis of Articles Published in Substance Abuse Journals: Intellectual Structure and Research Fields (2001-2012).

2016

The purpose of this study is to introduce a new methodology in the field of substance abuse, namely, co-citation analysis, which uses the bibliographic references of publications to establish the main thematic areas being researched and to identify the seminal documents that have contributed to establishing the intellectual foundation of the discipline at the present time.We identified all bibliographic references that were cited in documents published in the substance abuse journals included in the Journal Citation Reports in the 2001-2012 period, generating a co-citation matrix. This matrix was used to perform a co-citation network analysis. The co-citation network analysis led to the iden…

Estimating population size in wastewater-based epidemiology. Valencia metropolitan area as a case study.

2017

Wastewater can provide a wealth of epidemiologic data on common drugs consumed and on health and nutritional problems based on the biomarkers excreted into community sewage systems. One of the biggest uncertainties of these studies is the estimation of the number of inhabitants served by the treatment plants. Twelve human urine biomarkers —5-hydroxyindoleacetic acid (5-HIAA), acesulfame, atenolol, caffeine, carbamazepine, codeine, cotinine, creatinine, hydrochlorothiazide (HCTZ), naproxen, salicylic acid (SA) and hydroxycotinine (OHCOT)— were determined by liquid chromatography-tandem mass spectrometry (LC–MS/MS) to estimate population size. The results reveal that populations calculated fr…

Extensive molecular analysis of patients bearing CFTR-related disorders.

2012

Cystic fibrosis transmembrane conductance regulator (CFTR)–related disorders (CFTR-RDs) may present with pancreatic sufficiency, normal sweat test results, and better outcome. The detection rate of mutations is lower in CFTR-RD than in classic CF: mutations may be located in genes encoding proteins that interact with CFTR or support channel activity. We tested the whole CFTR coding regions in 99 CFTR-RD patients, looking for gene mutations in solute carrier (SLC) 26A and in epithelial Na channel (ENaC) in 33 patients who had unidentified mutations. CFTR analysis revealed 28 mutations, some of which are rare. Of these mutations, RT-PCR demonstrated that the novel 1525-1delG impairs exon 10 s…

Substance use disorders as risk factors for suicide in an Eastern and a Central European city (Tallinn and Frankfurt/Main)

2009

Cultural and regional differences on the well-known elevated suicide risk in substance use disorders have not been clarified yet. Therefore, the suicide risk associated with substance use disorders in a society of transition and in a socially and economically stable society should be identified and compared. Data from two population-based matched case-control studies were used to analyse the association between alcohol and other substance consumption and the risk of suicide. Data in Frankfurt were obtained by a semi-structured interview including the Structured Clinical Interview for DSM-IV Axis I (SCID-I) in 163 suicides that occurred in 1999 and 2000, and data from Tallinn were collected …

Opioid epidemic spread from Northern and Eastern Europe to Mediterranean Area.

2021

The addiction to illicit opioid and the misuse of prescription synthetic opioids pain relievers and fentanyl analogs generated an opioid epidemic in North America over the last two decades that affected public health with a constantly rising number of overdoses deaths. This health treat moved to Europe with a significant increase starting from 2015 involving mainly norther and eastern countries and finally also the Mediterranean area. The "lock down" isolation and economic recession caused by COVID-19 pandemic showed a resurgence in opioid use and harms.
