Enzymes involved in the dynamic equilibrium of core histone acetylation ofPhysarum polycephalum
1992
DEAE-Scpharose chromatography of extracts from plasmodia of the myxomyccte PI~.~suru~~t ,~/.~crpl~~ho~~ revealed the presence of multiple histone acetyltransferases and histonc deacctylascs. A cyloplasmic histonc acctyltransferase B, specific for histonc H4, and two nuclear acetyltransferases Al and A2 were identilied; Al acetylates all core hislones with a preference for l-13 and H2A. whereas A2 is specific for H3 and also slightly for H2B. Two hislone deacetylases. HDI and HD2, could be discriminated. They differ with respect to subslralc speciliciiy and pH dependence. For the first time the substrate specificity of histonc deacetylascs was determined using HPLC-purilicd individual core h…
Gcn5p is involved in the acetylation of histone H3 in nucleosomes.
1997
Abstract Enzymatic extracts from a gcn5 mutant and wild-type strains of Saccharomyces cerevisiae were chromatographically fractionated and the histone acetyltransferase activities compared. When free histones were used as substrate, extracts from wild-type cells showed two peaks of activity on histone H3 but extracts from gcn5 mutant cells showed only one. With nucleosomes as substrate, the histone acetyltransferase activities present in extracts from the gcn5 mutant strain were not able to modify H3 whereas wild-type cell extracts acetylated intensely this histone. The activity that acetylated nucleosome-bound H3 behaved as a 170-kDa complex. We suggest that Gcn5p represents a catalytic su…
Past price “memory” in the housing market: testing the performance of different spatio-temporal specifications.
2017
ABSTRACTRecent methodological developments provide a way to incorporate the temporal dimension when accounting for spatial effects in hedonic pricing. Weight matrices should decompose the spatial effects into two distinct components: bidirectional contemporaneous spatial connections; and unidirectional spatio-temporal effects from past transactions. Our iterative estimation approach explicitly analyses the role of time in price determination. The results show that both spatio-temporal components should be included in model specification; past transaction information stops contributing to price determination after eight months; and limited temporal friction is exhibited within this period. T…
Precarious Sovereignty in a Post-Liberal Europe : the Covid-19 Emergency in Estonia and Finland
2020
The paper addresses a puzzle resulting from the current global state of alert: the coronavirus pandemic brought us back to the world of the allegedly sovereign nation states with borders and national governments in charge, yet in fact, this retrieved sovereignty looks very vulnerable and precarious. We explain this controversy through a triad of concepts—sovereignty, governmentality, and post-liberalism—that we apply to an analysis of a corona-imposed state of emergency in Estonia and Finland. Based on comparative case study research, we posit that sovereignty is precarious in post-liberalism due to its large dependence on the technologies of responsibilization and agency. From a biopolitic…
Influence of indigenous Hanseniaspora uvarum and Saccharomyces cerevisiae from sugar-rich substrates on the aromatic composition of loquat beer
2022
The demand for unique and exclusive food products and beverages is constantly on the increase. One of the products that mostly evolved to encounter market dynamics in the last decade is craft beer. For a long time, craft breweries have included fruit in beer production to enrich flavour and aroma profile of different beer styles. In this study, for the first time, the use of Saccharomyces and non-Saccharomyces yeast strains isolated from high -sugar matrices (manna and fermented honey by-products) were investigated to diversify fruit craft beer pro-duction, in order to improve the fermentation process and highlight the complexity of aroma profiles generated during alcoholic fermentation. Tw…
Effect of ethanol on membrane fluidity of protoplasts fromSaccharomyces cerevisiae andKloeckera apiculata grown with or without ethanol, measured by …
1994
Direct measurements of membrane fluidity by fluorescence anisotropy of protoplasts fromKloeckera apiculata andSaccharomyces cerevisiae, a low and a high ethanol tolerant strain respectively, are presented. The comparison of the behaviour of the two strains grown with or without ethanol enabled us to demonstrate the existing relationship between ethanol tolerance and membrane fluidity.
Fructan Biosynthesis by Intra- and Extracellular Zymomonas mobilis Levansucrase after Simultaneous Production of Ethanol and Levan
2003
Summary The chemical composition of the Zymomonas mobilis biomass and the culture liquid after ethanol and levan synthesis were studied. The activities of intra- and extracellular levansucrase produced by the Z. mobilis strain 113 ”S” under optimum conditions both for levan and fructooligosaccharide (FOS) synthesis were also determined. It was shown that levan production relates to the reduction of the carbohydrate and lipid content in the biomass by increasing the nucleic acid and protein content. The levan producing activity of cellular levansucrase after ethanol and levan synthesis was approximately 30–40% of the total activity in the second fermentation stage. It was established that th…
Die Bildung höherer Alkohole bei Aminosäuremangelmutanten von Saccharomyces cerevisiae III. Höhere Alkohole als Nebenprodukte der Aminosäuresynthese
1975
Summary The formation of higher alcohols as byproducts of the synthesis of threonine, isoleucine and valine was investigated using isogenic haploid and diploid mutant strains of Saccharomyces cerevisiae . Genetic markers were ade 2, leu 1. The growth rate, the formation of ethanol, the amino acid metabolism and the formation of higher alcohols were determined during fermentations in a synthetic medium. n-Propanol-1, 2-methylbutanol-l and isobutanol were synthesized parallel to ethanol formation during exponential and stationary growth phase. However, the formation of 3-methylbutanol-l from leucine was restricted to the exponential phase. A comparison of haploid and diploid strains revealed,…
Attachment of yeast to modified stainless steel wire spheres, growth of cells and ethanol production
1999
Abstract The immobilization of yeast Saccharomyces cerevisiae, their growth and ethanol production were investigated using untreated and modified stainless steel wire spheres (WS) as carriers. The carrier surface was modified by oxidation, by treatment with titanium (IV) chloride (TiCl4) or by γ-aminopropyltrietoxysilane (AS) in an attempt to raise the efficiency of the immobilization of the yeast cells. The influence of the cell fixation method on culture growth and ethanol synthesis was investigated. The immobilization of cells to carrier surface was checked by scanning electron microscopy (SEM). More closely attachment of yeast cells was seen on the aminated wire surface. It was establis…
