Specific proteolysis by the tetanus toxin light chain of a vesicle-associated membrane protein (VAMP) involved in exocytosis is thought to underlie its intracellular blockade of neurotransmitter release. To substantiate this mechanism, recombinant light chain was expressed as a maltose binding protein-light chain fusion product in Escherichia coli. After purification of affinity chromatography and cleavage with factor Xa, the resultant light chain was isolated and its identity confirmed by Western blotting and N-terminal sequencing. Abstract Pentachlorophenol 4-monooxygenase (PCP4MO) from Sphingomonas chlorophenolica is a flavoprotein that hydroxylates PCP in the presence of NADPH and oxygen. In order to investigate the structure and function of active site, recombinant PCP4MO (rePCP4MO) was produced in Escherichia coli as a glutathione S-transferase (GST) fusion protein. Moreover, a tobacco etch virus (TEV) protease cleavage site (EKLYFQG) was introduced into GST-PCP4MO and a his-tagged TEV protease was employed. Tässä tutkielmassa tarkasteltiin käsitetestien synnyttämää keskustelua mekaniikan peruskurssilla. Käsitetestit ovat oleellinen osa Peer Instruction -opetusmenetelmää, joka pyrkii lisäämään opiskelijoiden roolia aktiivisina oppijina massakurssilla. Kurssin luennot rakentuivat vuorottelevista luennoitsijan alustuksista ja esitetyistä käsitetesteistä. Opiskelijat pohtivat ja vastasivat ensin testeihin itsenäisesti. In the current study, a new technique for the manufacturing of polymer nanocomposites (PNCs) with ultra-low filling content (<0.05 wt%) is presented. Specifically, this method has been validated through the production of polypropylene (PP) nanocomposites with Ag nanoparticles. The presented method first consisted in a metal nano-film deposition on PP pellet substrates by a physical vapor deposition (PVD) sputtering equipment. Subsequently, a percentage of the coated pellet was mixed to uncoated ones via an injection molding machine to produce the PNC in a single step. Quantitative data on the stability of mono-, di- and trimethyltin(IV)-carboxylate complexes (acetate, malonate, succinate, malate, oxydiacetate, diethylenetrioxydiacetate, tricarballylate, citrate, butanetetracarboxylate and mellitate) are reported at t=25°C and I → 0 mol l - 1 . Several mononuclear, mixed proton, mixed hydroxo and polynuclear species are formed in these systems. As expected, the stability trend is mono- > di- > trimethyltin(IV) and mono < di < tri < tetra < hexa for the organotin moieties and carboxylate ligands investigated, respectively.
