Fibril formation and toxicity of the non-amyloidogenic rat amylin peptide.

Full-length native rat amylin 1-37 has previously been widely shown to be unable to form fibrils and to lack the toxicity of the human amylin form leading to its use as a non-amyloidogenic control peptide. A recent study has suggested that rat amylin 1-37 forms amyloidogenic β-sheet structures in the presence of the human amylin form and suggested that this property could promote toxicity. Using TEM analysis we show here fibril formation by synthetic rat amylin 1-37 and 8-37 peptides when the lyophilized HPLC purified peptides are initially dissolved in 20 mM Tris-HCl. Dissolution of synthetic rat amylin 1-37 and 8-37 peptides in H(2)O or phosphate buffered saline failed to produce fibrils.…
Type II keratin cDNAs from the rainbow trout: implications for keratin evolution.

From a teleost fish, the rainbow trout Oncorhynchus mykiss, we have cloned and sequenced cDNAs encoding five different type II keratins. The corresponding protein spots, as separated by 2D-PAGE of trout cytoskeletal preparations, have been identified by peptide mass mapping using MALDI mass spectrometry. Three of the sequenced keratins are expressed in the epidermis (subtype IIe), and two in simple epithelia and mesenchymal cells (subtype IIs). The IIs keratins are both orthologs of human K8. This leaves unsequenced only the trace component S3 of the biochemically established trout keratin catalog. Plasticizer extraction of Taxol infusion solution from various infusion devices.

Taxol solution extracts the plasticizer DEHP (di(2-ethylhexyl)phthalate) from polyvinyl chloride (PVC) materials. In order to minimize patient exposure to DEHP, Taxol solutions should be prepared and administered in PVC-free materials. Particulate matter may form in Taxol infusion solution over time, so that in-line filtration with microporous membranes not greater than 0.22 microns is advisable. The purpose of this study was to evaluate the suitability of various administration- and in-line filter-sets for Taxol application. Gain of function mutations in the H3K27 methyltransferase EZH2 represent a promising therapeutic target in germinal center lymphomas. In this study, we assessed the frequency and distribution of EZH2 mutations in a large cohort of patients with follicular lymphoma (FL) (n = 366) and performed a longitudinal analysis of mutation during the disease progression from FL to transformed FL (tFL) (n = 33). Mutations were detected at 3 recurrent mutation hot spots (Y646, A682, and A692) in 27% of FL cases with variant allele frequencies (VAF) ranging from 2% to 61%. Studies on zymogenicity and solubilization of chitin synthase from Candida albicans

The zymogenic form of the chitin synthase present in mixed membrane preparations was extracted by digitonin treatment. The residual extracted membranes exclusively retained the basal activity. Trypsin activation of the zymogenic form of the enzyme did not modify the digitonin solubilization characteristics of the original zymogenic form, suggesting significant differences between ‘in vivo’ activation of chitin synthase and that carried out by trypsin ‘in vitro’.
Humic acid reduces gonadotropin activity and hormonal sensitivity of frog oocytes

The specific stimulatory effect of sturgeon Acipenser güldenstädti Br. gonadotropic hormone (GTH) on frog Rana temporaria L. oocyte maturation in vitro was investigated in relation to humic acid (HA) concentrations from 12.5 to 50 mg/l. HA was observed to bind to both the follicular membrane of the oocytes and the GTH molecule, reducing the oocytes' hormone sensitivity and maturation ability. It was also shown that HA inactivated GTH, lowering its specific ability to stimulate oocyte maturation.

Abstract In this paper, the effects of model (commercial) and natural (extracted from peat) humic substances on the membrane of liposomes formed with egg yolk lecithin (EYL) are presented. In our research, mass concentrations of fulvic and humic acids were used, which in relation to lecithin varied from 0% to 13%. In the integrin family, the collagen receptors form a structurally and functionally distinct subgroup. Two members of this subgroup, α1β1 and α2β1 integrins, are known to bind to monomeric form of type I collagen. However, in tissues type I collagen monomers are organized into large fibrils immediately after they are released from cells. Here, we studied collagen fibril recognition by integrins. By an immunoelectron microscopy method we showed that integrin α2I domain is able to bind to classical D-banded type I collagen fibrils. Modular Small Diameter Vascular Grafts with Bioactive Functionalities.

We report the fabrication of a novel type of artificial small diameter blood vessels, termed biomimetic tissue-engineered blood vessels (bTEBV), with a modular composition. They are composed of a hydrogel scaffold consisting of two negatively charged natural polymers, alginate and a modified chitosan, N,O-carboxymethyl chitosan (N,O-CMC).
