Selective binding to monoamine oxidase A: in vitro and in vivo evaluation of (18)F-labeled β-carboline derivatives.

2015

In this study we synthesized four different (18)F-labeling precursors for the visualization of the monoamino oxidase A using harmol derivatives. Whereas two are for prosthetic group labeling using [(18)F]fluoro-d2-methyl tosylate and 2-[(18)F]fluoroethyl-tosylate, the other three precursors are for direct nucleophilic (18)F-labeling. Additionally the corresponding reference compounds were synthesized. The syntheses of [(18)F]fluoro-d2-methyl-harmol and 2-[(18)F]fluoroethyl-harmol were carried out using harmol as starting material. For direct nucleophilic (18)F-labeling of the tracers carrying oligoethyled spacers (PEG), a toluenesulfonyl leaving group was employed. The radiolabeling, purifi…

Fluorescent Hg2+ Sensors: Synthesis and Evaluation of a Tren-Based Starburst Molecule Containing Fluorinated 1,2,4-Oxadiazoles.

2010

A new tren-based starburst molecule containing fluorinated 1,2,4-oxadiazoles as fluorophores has been synthesized and its sensing behavior toward several metal cations has been investigated by UV/Vis, fluorescence, 1 H NMR and 19 F NMR spectroscopy. Selective sensing for Hg 2+ ions through a PET-based mechanism was evidenced, suggesting application as fluorescent sensor for Hg 2+ of the off-on type.

Selective derivatisation of resorcarenes: Part 7. The reason for the diastereoselectivity of Mannich reactions with chiral amines

2001

The formation of tetrakis(benzo-3,4-dihydro-1,3-oxazines) 2 by condensation of resorcarenes 1 with various primary amines and an excess of formaldehyde is not only regioselective but, in the case of (R)- or (S)-1-phenylethylamine and their para-substituted (Br, Me) derivatives, also completely diastereoselective. The reaction with the structurally similar (R)-1-aminoindane and (S)-1-(1-naphthyl)ethylamine, on the other hand, produced only a slight excess of one of the two possible epimers 2 and with (R)-1-cyclohexylethylamine no diastereoselectivity was found at all. The presence and the ratio of both diastereomeric structures can be clearly determined by NMR spectroscopy. Inspection of par…

Diastereoselective formation of homochiral flexible perylene bisimide cyclophanes and their hybrids with fullerenes†‡

2021

Cyclophanes of different ring sizes featuring perylene-3,4:9,10-tetracarboxylic acid bisimide (PBI) linked by flexible malonates were designed, synthesized, and investigated with respect to their structural, chemical and photo-physical properties. It is predominantly the number of PBIs and their geometric arrangement, which influence dramatically their properties. For example, two-PBI containing cyclophanes reveal physico-chemical characteristics that are governed by strong co-facial π–π interactions. This is in stark contrast to cyclophanes with either three or four PBIs. Key to co-facial π–π stackings are the flexible malonate linkers, which, in turn, set up the ways and means for diaster…

N-Substituted and ring opened saccharin derivatives selectively inhibit transmembrane, tumor-associated carbonic anhydrases IX and XII.

2017

A series of N-substituted saccharins incorporating aryl, alkyl and alkynyl moieties, as well as some ring opened derivatives were prepared and investigated as inhibitors of the metalloenzyme carbonic anhydrase (CA, EC 4.2.1.1). The widespread cytosolic isoforms CA I and II were not inhibited by these sulfonamides whereas transmembrane, tumor-associated ones were effectively inhibited, with KIs in the range of 22.1-481nM for CA IX and of 3.9-245nM for hCA XII. Although the inhibition mechanism of these tertiary/secondary sulfonamides is unknown for the moment, the good efficacy and especially selectivity for the inhibition of the tumor-associated over the cytosolic, widespread isoforms, make…

3H-1,2-benzoxathiepine 2,2-dioxides: a new class of isoform-selective carbonic anhydrase inhibitors

2017

Abstract A new chemotype with carbonic anhydrase (CA, EC 4.2.1.1) inhibitory action has been discovered, the homo-sulfocoumarins (3H-1,2-benzoxathiepine 2,2-dioxides) which have been designed considering the (sulfo)coumarins as lead molecules. An original synthetic strategy of a panel of such derivatives led to compounds with a unique inhibitory profile and very high selectivity for the inhibition of the tumour associated (CA IX/XII) over the cytosolic (CA I/II) isoforms. Although the CA inhibition mechanism with these new compounds is unknown for the moment, we hypothesize that it may be similar to that of the sulfocoumarins, i.e. hydrolysis to the corresponding sulfonic acids which therea…

Preparation of novel high permeability and antifouling polysulfone-vanillin membrane

2020

International audience; A novel high-performance nanofiltration membrane was fabricated by a simple and scalable route involving in situ cross-linking of hydrophilic, cheap, and environmentally friendly vanillin as antifouling agent with polysulfone (PSf) for salt rejection performance. Vanillin acts as a porogen, which induces a negative surface charge on the membrane surface due to the presence of polar functional groups like alcohol and aldehyde. The surface properties, including charge, morphology, and hydrophilicity, were investigated in detail using analytical instruments. The nanofiltration performance of the fabricated PSf-vanillin membranes was dependent on the percentage of vanill…

Selective photooxidation of ortho-substituted benzyl alcohols and the catalytic role of ortho-methoxybenzaldehyde

2016

It has been recently reported by Palmisano et al. (2015) [1] that the oxidation of 2-methoxybenzyl alcohol (2-MBA) to 2-methoxybenzaldehyde (2-MBAD) proceeds in water under near-UV light with an unexpected catalytic effect of 2-MBAD. In order to investigate the catalytic role of aldehyde in photolytic oxidation of ortho-substituted benzyl alcohols (OSBAs), reactivity runs were carried out with 2-methylbenzyl alcohol (2-MeBA), 2-nitrobenzyl alcohol (2-NBA), 2-hydroxybenzyl alcohol (2-HBA) and 2-chlorobenzyl alcohol (2-CIBA) in the absence and in the presence of their corresponding aldehyde. None of those alcohols showed a measurable oxidation rate even in the presence of their aldehydes but …

INFLUENCE OF THE EXPERIMENTAL SYSTEM AND OPTIMIZATION OF THE SELECTIVITY FOR THE ELECTROCARBOXYLATION OF CHLOROACETONITRILE TO CYANOACETIC ACID

2004

The electrocarboxylation of chloroacetonitrile (ClACN) to cyanoacetic acid in undivided cells equipped with sacrificial anodes was investigated both in a bench-scale electrochemical batch reactor and in a continuous batch recirculation reaction system equipped with a parallel-plate electrochemical cell. Selectivity and faradaic yields higher than 90% were obtained by working under anhydrous conditions with low halide concentrations and at more negative potentials. It was found that the instantaneous selectivity in the carboxylate compound depends mainly on the [CO 2 ]/[ClACN] ratio at high substrate concentration and is close to 100% for low ClACN concentration. These results were consisten…
