Tumor targeting using agents with slow pharmacokinetics represents a major challenge in nuclear imaging and targeted radionuclide therapy as they most often result in low imaging contrast and high radiation dose to healthy tissue. To address this challenge, we developed a polymer-based targeting agent that can be used for pretargeted imaging and thus separates tumor accumulation from the imaging step in time. Tässä opinnäytetyössä tarkastellaan, onko kokonaisarkkitehtuurin kypsyystasolla ja kokonaisarkkitehtuurituotteen laadulla keskinäistä yhteyttä. Tämän selvittämiseksi valittiin kirjallisuuslähteistä ne artikkelit, joissa kokonaisarkkitehtuurin laatua tutkitaan laatuattribuuttien (quality attributes) kautta. Näiden artikkeleiden perusteella muodostettiin kahdeksan laatuattribuuttia. Näiden laatuattribuuttien kuvauksia verrattiin kolmen kypsyysmallin (JHKA:n kypsyystasomalli, EAMM, DyAMM) kypsyystasokuvauksiin. Aplidin® is an antitumor agent in phase II clinical trials that induces apoptosis through the sustained activation of Jun N-terminal kinase (JNK). We report that Aplidin® alters glutathione homeostasis increasing the ratio of oxidized to reduced forms (GSSG/GSH). Aplidin® generates reactive oxygen species and disrupts the mitochondrial membrane potential. Exogenous GSH inhibits these effects and also JNK activation and cell death. We found two mechanisms by which Aplidin® activates JNK: rapid activation of Rac1 small GTPase and downregulation of MKP-1 phosphatase. In order to investigate the involvement of Ras and/or Rho proteins in the induction of the inducible isoform of nitric oxide synthase (NOS II) we used HMG-CoA reductase inhibitors (statins) and Clostridium difficile toxin B (TcdB) as pharmacological tools. Statins indirectly inhibit small G proteins by preventing their essential farnesylation (Ras) and/or geranylgeranylation (Rho). In contrast, TcdB is a glucosyltransferase and inactivates Rho-proteins directly.
