Steatosis is the first step in the development of non-alcoholic fatty liver disease (NAFLD). However, the mechanisms involved in its pathogenesis are not fully understood. Many nuclear receptors (NRs) involved in energy homeostasis and biotransformation constitute a network connecting fatty acids, cholesterol and xenobiotic metabolisms; therefore, multiple NRs and their ligands may play a prominent role in liver fat metabolism and accumulation. On the Optimization of Self-Organizing Maps by Genetic Algorithms

Publisher Summary This chapter reviews the research on the genetic optimization of self-organizing maps (SOMs). The optimization of learning rule parameters and of initial weights is able to improve network performance. The latter, however, requires chromosome sizes proportional to the size of the SOM and becomes unwieldy for large networks. The optimization of learning rule structures leads to self-organization processes of character similar to the standard learning rule. A particularly strong potential lies in the optimization of SOM topologies, which allows the study of global dynamical properties of SOMs and related models, as well as to develop tools for their analysis. Gelation of smectic liquid crystal phases with photosensitive gel forming agents.

This paper studies the potential of 3 newly synthesized low molecular weight gelling agents (2 of them with azo groups) with the semicarbazide group as a H-bonding motif to gel the smectic phase of a commercial chiral smectic C material. To detect gelation two methods were used: the stabilization of the director pattern, which measures the interaction of the physical gel network with the LC-director, and the suppression of the onset of electrohydrodynamic instabilities, which measures the increase of viscosity. Depletion of cytosolic GSH decreases the ATP levels and viability of synaptosomes from aged mice but not from young mice

The effect of glutathione depletion on the viability of freshly isolated synaptosomes from whole brain was investigated in young and aged mice. Aging did not influence the GSH and ATP levels and the viability of these synaptosomes. However depletion of glutathione caused by the cytosolic glutathione inhibitor diethyl maleate (1 mM) resulted in a significant decline, after 60 min of incubation, in ATP levels and viability in the synaptosomes from aged mice but not in those from young mice. Postmenopausal hormone replacement therapy modifies skeletal muscle composition and function: a study with monozygotic twin pairs

We investigated whether long-term hormone replacement therapy (HRT) is associated with mobility and lower limb muscle performance and composition in postmenopausal women. Fifteen 54- to 62-yr-old monozygotic female twin pairs discordant for HRT were recruited from the Finnish Twin Cohort. Habitual (HWS) and maximal (MWS) walking speeds over 10 m, thigh muscle composition, lower body muscle power assessed as vertical jumping height, and maximal isometric hand grip and knee extension strengths were measured. Npl3 stabilizes R-loops at telomeres to prevent accelerated replicative senescence.

Abstract Telomere shortening rates must be regulated to prevent premature replicative senescence. TERRA R‐loops become stabilized at critically short telomeres to promote their elongation through homology‐directed repair (HDR), thereby counteracting senescence onset. Using a non‐bias proteomic approach to detect telomere binding factors, we identified Npl3, an RNA‐binding protein previously implicated in multiple RNA biogenesis processes. Using chromatin immunoprecipitation and RNA immunoprecipitation, we demonstrate that Npl3 interacts with TERRA and telomeres. Aneuploidy as a consequence of senescence and ovariectomy in the golden hamster (Mesocricetus auratus).

The hypothesis of the preferred X-chromosome loss in elder human females was reevaluated in the golden hamster: early castration of females proved that the increase of aneuploid cells is correlated with the loss of the ovaries. But here, and in old females, aneuploidy consisted of random loss of excess of chromosomes, in no case an X-chromosome.

NECAB2 participates in an endosomal pathway of mitochondrial stress response at striatal synapses

Synaptic signaling depends on ATP generated by mitochondria. Due to extensive connectivity, the striatum is especially vulnerable to mitochondrial dysfunction and thus requires efficient mitochondrial quality control. We found that the neuronal calcium-binding protein NECAB2 ensures synaptic function in the striatum by increasing mitochondrial efficiency. NECAB2 associates with early endosomes and mitochondria at striatal synapses. Loss of NECAB2 dysregulates proteins of the endosomal ESCRT machinery and oxidative phosphorylation. ABO genotyping by PCR-RFLP and cloning and sequencing

A refined PCR-RFLP based method was established to genotype ABO blood groups. The main objective of this study was to make the techniques also suitable for working with degraded DNA. Specific primer design was carried out to choose fragments shorter than 200 bp as necessary in forensic and archaeological applications. Four fragments of exon 6 and 7 of the ABO gene were amplified and digested by in total 7 restriction endonucleases.
