Effect of methyl jasmonate in gene expression, and in hormonal and phenolic profiles of holm oak embryogenic lines before and after infection with Ph…
2022
The dieback syndrome affecting Quercus ilex and other oak species impels the search for tolerant plant genotypes, as well as methods of plant immunization against such infections. Elicitation treatments can be an effective strategy to activate plant defense response and embryogenic lines represent a promising tool to generate new tolerant genotypes and also to study early markers involved in defense response. The aim of the presented work was to investigate changes in gene expression, and in hormonal and phenolic profiles induced in three holm oak embryogenic lines (ELs) elicited with methyl jasmonate (MeJA) before and after infection with the oomycete Phytophthora cinnamomi, which is the m…

Perspectives of the encapsulation technology in the nursery activity of Citrus
2007
The genetic and phytosanitary certification has to guarantee the quality of the propagation material also in Citrus. Traditional methods can be assisted by the techniques of in vitro culture, which are fundamental for the mass production of high quality plants. The encapsulation technology represents an innovative tool to combine the advantages of micropropagation with handling ease, storability, reduced size of the propagules, mechanization potentiality, transportability of the gamic seeds. In this work somatic embryos obtained from in vitro anther culture of the cultivar Mandarino Tardivo di Ciaculli (Citrus reticulata Blanco) were encapsulated in a sodium alginate matrix and synthetic se…
Somatic embryogenesis through in vitro anther culture ofCitrus sinensisL. Osbeck ‘Moro’
2019
In many crops, anther culture is the most used method to induce gametic embryogenesis, aimed to regenerate homozygous plants. However, also somatic embryogenesis can be obtained by this method, when somatic tissue is involved in regeneration process. Many factors can affect this procedure, such as genotype, pre-treatments applied to floral buds, pollen developmental stage, donor plant states, culture media composition and growth culture conditions. Anthers of Citrus sinensis L. Osbeck, cv. Moro, were collected at the vacuolate stage, and after a chilling (4°C) pre-treatment of 7 days, were placed on the same medium, evaluating different temperature stresses applied after the placing in cult…
Preliminary research on conversion of encapsulated somatic embryos of Citrus reticulata Blanco, cv. Mandardino Tardivo di Ciaculli.
2006
Somatic embryogenesis was obtained through anther culture of Citrus reticulata, cv. Mandarino Tardivo di Ciaculli. The work was carried out to evaluate the response of somatic embryos inside a sodium alginate coating to different storage periods, and to the effects of the germicide PPM (1 ml l(-1) stop) and the fungicide Thiophanate-methyl (100 mg l(-1) stop). The effect of these alone or in combination, added to the artificial endosperm on the performance of the encapsulated somatic embryos was tested. After 45 days of culturing, sprouting, root development and conversion were recorded. The encapsulated somatic embryos can be conveniently stored at 4 degrees C, but not for a period longer …
Rearing of Prays citri on callus derived from lemon stigma and style culture
2000
A new method for rearing the citrus flower moth (Prays citri Mill.) (Lepidoptera, Yponomeutidae) on lemon [Citrus limon (L.) Burm.] callus is reported. In the present research callus (an undifferentiated mass of plant cells that can be grown under sterile conditions on an artificial medium in vitro) was induced from lemon stigma and style explants cultured on Murashige and Skoog (MS) medium supplemented with 500 mg l -1 malt extract, 13.3 μM 6-benzylaminopurine, and 146 mM sucrose. Also somatic embryogenesis and plant regeneration were obtained from the cultures of styles and stigmas of lemon. Adults were obtained from larvae on infested flowers collected in the field. Different oviposition…
Control of in vitro somatic embryogenesis of the spindle tree ( Euonymus europaeus L.) by the sugar type and the osmotic potential of the culture med…
1999
In vitro somatic embryogenesis was achieved from zygotic embryo explants of a woody angiosperm species, the spindle tree, cultivated on various culture media differing in their sugar type and concentration, or in the applied osmotic potential. The highest frequency of somatic embryogenesis was obtained with a 350 mM sucrose, or a 89 mM glucose concentration in the culture medium. Experiments with culture media differing only in osmotic potential indicated that a minimal threshold osmotic potential is required to stimulate the emergence of somatic embryos. Elevated concentrations of glucose have an inhibitory effect, independent of their osmotic effect, while elevated concentrations of sucr…
Morphological and anatomical observations of abnormal somatic embryos from anther cultures of Citrus reticulata
2010
A morphological and anatomical study of regenerants obtained from mandarin anther culture was carried out. Beside morphologically normal somatic embryos, abnormal structures were originated in the course of somatic embryogenesis. Anatomical anomalies can be found at several growth stages, causing the formation of slender stems, stubby structures, non-functional leaves. When too long, some structures are subject to shedding, with the formation of various abscission zones. Most of them are subject to degeneration, although many are capable of further, localized, morphogenesis. A thorough knowledge of morphology and anatomy of normal and abnormal regenerants could make possible to select and s…
Stimulation of root and somatic embryo production in Euconymus europaeus L. by an inhibitor of polyamine biosynthesis
1995
In vitro formation of roots and somatic embryos is obtained from cotyledon explants of a Spindle tree (Euonymus europaeus L.) cultured on two different media: a medium inducing callus formation and the production of roots, and a medium inducing callus formation, root and somatic embryo production. We studied the effects of α-difluoromethylornithine (DFMO), a specific, irreversible inhibitor of ornithine decarboxylase (ODC) on root and somatic embryo production, growth and titers of putrescine in Euonymus explants and explant-derived calli. Early changes in putrescine levels were detected in both cultures before the visible emergence of roots or somatic embryos. DFMO rapidly inhibited putres…
Correlating stress and somatic embryogenesis for NBT: myth or reality?
2018
Global warming and growing demography have increased demand for agronomic resources, leading to increasing lack of land suitable for agriculture and provoking several abiotic stresses which, added to biotic ones, result in physiological and metabolic disorders that impact on crop yield when most needed. Reducing this impact is a major scientific and agronomic challenge and biotechnology would be an efficient alternative. However, to reduce risks of somaclonal variation among regenerants it is better to produce them by somatic embryogenesis directly from explants or gametes, or indirectly from callus or cell suspensions. Thus, globular embryos regenerate and develop through to the heart, tor…
