AbstractAllogeneic hematopoietic stem cell transplantation can induce considerable tumor remissions in metastatic renal-cell carcinoma (RCC) patients. The precise effector mechanisms mediating these graft-versus-tumor reactions are unknown. We studied RCC-directed CD8+ T-cell responses in blood lymphocytes of healthy individuals matched with established RCC cell lines for HLA-class I. In 21 of 22 allogeneic mixed lymphocyte/tumor-cell cultures (MLTCs), RCC-reactive cytotoxic T-lymphocytes (CTLs) were readily obtained. Photoreduction of carbon dioxide to formic acid in aqueous suspension: a comparison between phthalocyanine/TiO2 and porphyrin/TiO2 catalysed processes

Composite materials prepared by loading polycrystalline TiO2 powders with lipophilic highly branched Cu(II)- and metal-free phthalocyanines or porphyrins, which have been used in the past as photocatalysts for photodegradative processes, have been successfully tested for the efficient photoreduction of carbon dioxide in aqueous suspension affording significant amounts of formic acid. The results indicated that the presence of the sensitizers is beneficial for the photoactivity, confirming the important role of Cu(II) co-ordinated in the middle of the macrocycles. Biogenic amines on fish tissue are formed as a result of bacterial contamination and spoilage during storage. A new method based on liquid chromatography (LC) and tandem mass spectrometry (MS/MS) using a triple quadrupole (QqQ) analyser was developed for the analysis of eight biogenic amines (cadaverine, histamine, phenylethylamine, putrescine, spermine, spermidine, tyramine and tryptamine) in fish tissues. Abstract The configurations and conformations of torreyol (6; “δ-cadinol”,10α-hydroxy-4-muurolene), α-cadinol (7; 10α-hydroxy-4-cadinene), T-muurolol (8; 10β-hydroxy-4-muurolene), and T-cadinol (9; 10β-hydroxy-4-cadinene) have been determined by an extensive PMR study using lanthanide-induced shifts. Characteristic mass spectrometric fragmentation patterns of muurolols and cadinols are briefly discussed.

High-resolution analysis of trace elements in crustose coralline algae from the North Atlantic and North Pacific by laser ablation ICP-MS

We have investigated the trace elemental composition in the skeleta of two specimens of attached-living coralline algae of the species Clathromorphum compactum from the North Atlantic (Newfoundland) and Clathromorphum nereostratum from the North Pacific/Bering Sea region (Amchitka Island, Aleutians). Samples were analyzed using Laser Ablation-Inductively Coupled Plasma Mass Spectrometry (LA-ICP-MS) yielding for the first time continuous individual trace elemental records of up to 69 years in length. Clumped isotope analysis of carbonates: comparison of two different acid digestion techniques

RATIONALE The kinetic nature of the phosphoric acid digestion reaction enables clumped isotope analysis of carbonates using gas source isotope ratio mass spectrometry (IRMS). In most laboratories acid digestions are performed at 25°C in sealed vessels or at 90°C in a common acid bath. Here we show that different Δ47 results are obtained depending on the digestion technique employed. METHODS Several replicates of a biogenic aragonite and NBS 19 were reacted with 104% H3PO4 in sealed vessels at 25°C and at 90°C using a common acid bath. Improving Fractionation of Human Milk Proteins through Calcium Phosphate Coprecipitation and Their Rapid Characterization by Capillary Electrophoresis

This work describes a simple sample pretreatment method for the fractionation of human milk proteins (into their two main groups, whey and caseins) prior to their analysis. The protein-extraction protocol is based on the addition of calcium phosphate to nonadjusted pH human milk. The combination of calcium ions with phosphate results in an effective coprecipitation of caseins. To assess the suitability of this fractionation protocol, the protein extracts were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), LC-MS/MS, and capillary electrophoresis (CE) analysis. Abstract Direct standardization (DS) and piecewise direct standardization (PDS) methods are applied to multivariate standardization of chemiluminescence signals using PLS model as the calibration model. The linear concentration interval of the chemiluminescence determination was determined. This interval was located using univariate robust calibration by least median of squares (LMS) method. Laurus nobilis L. Seed Extract Reveals Collateral Sensitivity in Multidrug-Resistant P-Glycoprotein-Expressing Tumor Cells.

The frequent failure of standard cancer chemotherapy requires the development of novel drugs capable of killing otherwise drug-resistant tumors. Here, we have investigated a chloroform extract of Laurus nobilis seeds. Fatty acids and 23 constituents of the volatile fraction were identified by gas chromotography/flame ionization detection (GC/FID) and gas chromatography/mass spectrometry (GC/MS), in good agreement with (1)H NMR (nuclear magnetic resonance) spectrum. Type I keratin cDNAs from the rainbow trout: independent radiation of keratins in fish

Five different type I keratins from a teleost fish, the rainbow trout Oncorhynchus mykiss, have been sequenced by cDNA cloning and identified at the protein level by peptide mass mapping using MALDI-MS. This showed that the entire range of type I keratins detected biochemically in this fish has now been sequenced. Three of the keratins are expressed in the epidermis (subtype Ie), whereas the other two occur in simple epithelia and mesenchymal cells (subtype Is). Among the Is keratins is an ortholog of human K18; the second Is polypeptide is clearly distinct from K18.
