Role of Activity in Defining Metabolic and Contractile Adaptations After SCI
2016
Spinal cord injury (SCI) can lead to moderate to severe muscle paralysis, loss of lower-limb functionality and often results in a reduced physical activity. As a consequence, people with SCI demonstrate numerous metabolic and contractile transformations such as leg muscles atrophy, a transformation from slow, fatigue-resistant fibers to fast, fatigable fibers, a decreased vascularization. Appropriate exercise and most especially exercise using functional electrical stimulation (FES) is now well-known to have beneficial effects on muscle characteristics, force output, exercise capacity, but also bone mineral density and cardiovascular parameters. For example, increases in muscle mass and str…
Pharmacological Suppression of CNS Scarring by Deferoxamine Reduces Lesion Volume and Increases Regeneration in an In Vitro Model for Astroglial-Fibr…
2015
Lesion-induced scarring is a major impediment for regeneration of injured axons in the central nervous system (CNS). The collagen-rich glial-fibrous scar contains numerous axon growth inhibitory factors forming a regeneration-barrier for axons. We demonstrated previously that the combination of the iron chelator 2,2'-bipyridine-5,5'-decarboxylic acid (BPY-DCA) and 8-Br-cyclic AMP (cAMP) inhibits scar formation and collagen deposition, leading to enhanced axon regeneration and partial functional recovery after spinal cord injury. While BPY-DCA is not a clinical drug, the clinically approved iron chelator deferoxamine mesylate (DFO) may be a suitable alternative for anti-scarring treatment (A…
Effects of Neuromodulation on Gait
2018
In the last decades, non-invasive brain stimulation (NIBS) has evolved to become a valuable tool in both basic and clinical neuroscience. Various methods of transcranial magnetic stimulation (TMS) and transcranial electrical stimulation (TES) have been widely used for diagnostic, prognostic and even therapeutic applications in a broad range of neurological and psychiatric disorders. The rationale for using NIBS techniques lies in the possibility to modulate, in a targeted manner, the activity of different cerebral and cerebellar cortical regions, as well as the functional connections between these areas and distant brain regions also including subcortical structures. The neural circuitry in…
Social comparison, coping and depression in people with spinal cord injury
2006
The present study among 70 people with spinal cord injury examined the prevalence and correlates of identification (seeing others as a potential future) and contrast (seeing others in competitive terms) in social comparison as related to coping and depression. The most prevalent social comparison strategy was downward contrast (a positive response to seeing others who were worse-off), followed by upward identification (a positive response to perceiving better-off others as a potential future), downward identification (a negative response to perceiving worse-off others as a potential future), and upward contrast (a negative response to seeing others who were better-off). Those with less seve…
The adult spinal cord harbors a population of GFAP-positive progenitors with limited self-renewal potential
2013
Adult neural stem cells (aNSCs) of the forebrain are GFAP-expressing cells that are intercalated within ependymal cells of the subventricular zone (SVZ). Cells showing NSCs characteristics in vitro can also be isolated from the periaqueductal region in the adult spinal cord (SC), but contradicting results exist concerning their glial versus ependymal identity. We used an inducible transgenic mouse line (hGFAP-CreERT2) to conditionally label GFAP-expressing cells in the adult SVZ and SC periaqueduct, and directly and systematically compared their self-renewal and multipotential properties in vitro. We demonstrate that a population of GFAP+ cells that share the morphology and the antigenic pr…
Laparoscopic endopelvic sacral implantation of a Brindley controller for recovery of bladder function in a paralyzed patient
2008
Background: A number of techniques are being investigated to accomplish bladder control recovery in paralyzed patients using the neurostimulation, but currently, all techniques are based on the dorsal implantation of the electrodes using a laminectomy. Methods: On 27 April 2006 we performed a laparoscopic implantation of a Finetech-Brindley bladder controller on the endopelvic sacral roots in a Th8 completely paralyzed woman who had previously undergone the removal of a Brindley controller due to an arachnoiditis after extrathecal implantation with intradural sacral deafferentation. Results: We required about 3.5 h for the entire surgical procedure; no complications occurred and the patient…
Efecto del pedaleo de brazos sobre el sistema cardiorrespiratorio de las personas con tetraplejia. (Effect of armcrank pedaling on the cardiorespirat…
2010
Outcomes with intracytoplasmic sperm injection of cryopreserved sperm from men with spinal cord injury
2013
Erectile dysfunction, ejaculatory dysfunction and poor semen quality are the main causes of infertility in men with spinal cord injury (SCI). Different sperm retrieval techniques such as penile vibratory stimulation (PVS), electro-ejaculation (EEJ) or surgical sperm retrieval (SSR) associated or not with sperm cryopreservation can be offered to these patients to preserve their fertility. If fatherhood cannot be achieved naturally, assisted reproductive techniques can be offered to these patients using either fresh or frozen/thawed sperm. The aim of the study was to report in SCI patients from Dijon (Burgundy) and in the literature, intracytoplasmic sperm injection (ICSI) outcomes using froz…
Methylprednisolone neutralizes the beneficial effects of erythropoietin in experimental spinal cord injury
2005
Inflammation plays a major pathological role in spinal cord injury (SCI). Although antiinflammatory treatment using the glucocorticoid methyprednisolone sodium succinate (MPSS) improved outcomes in several multicenter clinical trials, additional clinical experience suggests that MPSS is only modestly beneficial in SCI and poses a risk for serious complications. Recent work has shown that erythropoietin (EPO) moderates CNS tissue injury, in part by reducing inflammation, limiting neuronal apoptosis, and restoring vascular autoregulation. We determined whether EPO and MPSS act synergistically in SCI. Using a rat model of contusive SCI, we compared the effects of EPO [500-5,000 units/kg of bod…
Dry needling technique decreases spasticity and improves general functioning in incomplete spinal cord injury: A case report
2018
Context: Spasticity in neurological disorders (i.e. stroke patients and cerebral palsy) is positively improved by dry needling. However, reports are scarce regarding the potential effects of dry needling in reducing spasticity and improving functionality in patients with an incomplete spinal cord injury. The aim of this case report was to study the immediate, short-term effects of dry needling treatment (10 weeks) on spasticity, dynamic stability, walking velocity, self-independence, and pain in a single patient with an incomplete spinal cord injury. Findings: The dry needling treatment resulted in immediate, short-time effects on basal spasticity in the upper (reduction from 2 to 0 point m…
