The expression of the major protein kinase C substrate MARCKS (myristoylated alanine-rich C kinase substrate) is controlled by the stability of its mRNA. While the MARCKS mRNA is long living in quiescent fibroblasts (t1/2 = 14 h), its half-life time is drastically reduced (t1/2 = 2 h) in cells treated with phorbol esters to activate protein kinase C (PKC) or treated with growth factors. In a first step to study the underlying mechanism we identified both a cis-element on the MARCKS mRNA and the corresponding trans-acting factors. High yielding one-pot enzyme-catalyzed synthesis of UDP-glucose in gram scales

Abstract Uridine diphosphoglucose is an important cofactor of glucosylating enzymes. A simple and high yielding one-pot enzymatic synthesis of UDPG on a gram scale from glucose via hexokinase, phosphoglucomutase and UDPG pyrophosphorylase (UGPase) is described. Repetitive addition of substrate was used to avoid inhibition of UGPase. The approach allows recovery of active enzymes and their re-use. The synthesis of UDP-[4-13C]-glucose on a 0.5 g scale resulted in a final yield of 70% and a purity of >95% after chromatographic purification.

Superconducting properties of Nb thin films deposited on porous silicon templates

Porous silicon, obtained by electrochemical etching, has been used as a substrate for the growth of nanoperforated Nb thin films. The films, deposited by UHV magnetron sputtering on the porous Si substrates, inherited their structure made of holes of 5 or 10 nm diameter and of 10 to 40 nm spacing, which provide an artificial pinning structure. The superconducting properties were investigated by transport measurements performed in the presence of magnetic field for different film thickness and substrates with different interpore spacing. Effect of pH and nitrite concentration on nitrite oxidation rate

The effect of pH and nitrite concentration on the activity of the nitrite oxidizing bacteria (NOB) in an activated sludge reactor has been determined by means of laboratory batch experiments based on respirometric techniques. The bacterial activity was measured at different pH and at different total nitrite concentrations (TNO 2). The experimental results showed that the nitrite oxidation rate (NOR) depends on the TNO 2 concentration independently of the free nitrous acid (FNA) concentration, so FNA cannot be considered as the real substrate for NOB. Fast and accurate analysis and design of substrate integrated waveguide (SIW) filters

This paper describes the application of the boundary integral-resonant mode expansion (BI-RME) method to the accurate modelling of substrate integrated waveguides (SIW). The new formulation fully exploits the particular geometry of this class of structures, thus providing a very efficient analysis method to be used within CAD tools of SIW filters. The advantages of this method have been proved with the successful design of two filters operating in the microwave and millimeter-wave bands.

Magneto-optical effects in multilayers illuminated by total internal reflection

This paper describes the magneto-optical effects of metallic multilayers under the condition of total internal reflection. In the framework of Green’s dyadic technique, we detail a practical and at time-consuming scheme to compute accurately the optical properties of anisotropic multilayers deposited on a substrate. We present numerical simulations which account for the variation of the angle of incidence at a fixed wavelength and for the variation of the wavelength at fixed angle of incidence. The Kerr rotation is found to increase significantly around the critical angle for total reflection. The affinity for oxygen of polyphenoloxidase in grapes

The influence of oxygen on the reaction of polyphenoloxidase in grapes was determined. Using crude extracts of grapes with a sufficiently high polyphenoloxidase activity on a substrate of pyrocatechol, apparentKm values for oxygen, at 25° C were calculated as 1.1 × 10−4mol (or 9% 02) for Sultana, and 1 × 10−4mol (or 8% O2) for Doradillo grapes. The importance of these figures for wine making is discussed.

Rational selection of non-Saccharomyces wine yeasts for mixed starters based on ester formation and enological traits

Thirty-eight yeast strains belonging to the genera Candida, Hanseniaspora, Pichia, Torulaspora and Zygosaccharomyces were screened for ester formation on synthetic microbiological medium. The genera Hanseniaspora and Pichia stood out as the best acetate ester producers. Based on the ester profile Hanseniaspora guilliermondii 11027 and 11102, Hanseniaspora osmophila 1471 and Pichia membranifaciens 10113 and 10550 were selected for further characterization of enological traits. Abstract This work aims at evaluating the mechanical performance of joints in an AA6016 alloy, manufactured by means of two alternative processes: friction stir welding and adhesive bonding.
