Simultaneous quantification of the main organic acids and carbohydrates involved in tomato flavour using capillary zone electrophoresis

A capillary zone electrophoresis (CZE) procedure for the simultaneous determination of the major organic acids (oxalate, malate and citrate) and carbohydrates (fructose, glucose and sucrose) in Lycopersicon fruits is reported. Comparison of this method with routine HPLC methods indicates that the CZE method offers several attractive features (speed, resolution, sensitivity and cost) which significantly improve the determination of these compounds. Detection limits were better than 1.6 µg ml−1 for organic acids and from 13 to 24 µg ml−1 for carbohydrates; repeatabilities were better than 2.1% for migration times and between 1.4 and 7.3% for peak areas. Direct ATR-FTIR determination of sucrose in beet root.

A simple and fast procedure has been developed for the direct determination of sucrose in beet root samples through attenuated total reflectance Fourier transform infrared absorbance measurements (ATR-FTIR) at 1056 cm(-1) with a baseline established between 1187 and 887 cm(-1). The method only requires a previous crushing or liquefaction of samples and it is free from matrix effects and from the interference of minoritary sugars and fermentation molecules. PLS-NIR determination of total sugar, glucose, fructose and sucrose in aqueous solutions of fruit juices

Abstract An analytical procedure has been developed for the direct determination of sugars in fruit juice samples. The method is based on the partial least-squares (PLS) treatment of first derivative near infrared (NIR) spectroscopic data obtained between 1200 and 2450 nm, using 1 mm pathlength cell and a multicomponent calibration set, including seven binary mixtures and 10 ternary mixtures of glucose, fructose and sucrose. The limit of detection values found are of the order of 0.2 g/100 ml for total sugar and 0.2, 0.4 and 0.5 g/100 ml for glucose, fructose and sucrose, respectively. Quantification of low levels of amorphous content in sucrose by hyperDSC.

A method was developed for the quantification of low levels of amorphous content in sucrose with hyperDSC. The method was based on the fact that the change of specific heat at the glass transition is linearly proportional to the amorphous content. It was found out that as annealing time increased, the glass transition temperature moved to a higher temperature and the change of specific heat increased. DeltaC(p) for annealed totally amorphous sucrose was 0.761+/-0.012 Jg(-1) degrees C(-1). Synthetic mixtures with various proportions of crystalline and amorphous sucrose were prepared. Formation of Levan from Raffinose by Levansucrase ofZymomonas mobilis

Levansucrase (EC 2.4.1.10.) of Zymomonas mobilis 113S can perform the polymerisation of fructose moiety from raffinose to levan concomitantly with a release of non-catabolised melibiose into the medium. The kinetic parameters of the levansucrase-catalysed reaction provide even higher reaction velocities on raffinose as compared to sucrose, particularly at low substrate concentrations. Fructan Biosynthesis by Intra- and Extracellular Zymomonas mobilis Levansucrase after Simultaneous Production of Ethanol and Levan

Summary The chemical composition of the Zymomonas mobilis biomass and the culture liquid after ethanol and levan synthesis were studied. The activities of intra- and extracellular levansucrase produced by the Z. mobilis strain 113 ”S” under optimum conditions both for levan and fructooligosaccharide (FOS) synthesis were also determined. It was shown that levan production relates to the reduction of the carbohydrate and lipid content in the biomass by increasing the nucleic acid and protein content. Flame-burned stainless steel wire spheres as carriers ofZymomonas mobilis cells and extracellular levansucrase

In the present work, the use of flame-burned WS as carriers of Z. mobilis and extracellular levansucrase and the effect of the cell fixation method by dehydration on system productivity were investigated. Lyophilization and convective drying of Z. mobilis biomass at 30°C to a moisture content of 10–14% gave the best results for the repeated batch fermentations of a sucrose medium to obtain levan and ethanol. Significant correlation between the product formation and the concentration of free cells in the fermentation medium was established. Sugar Beet Juice Fermentation byZymomonas mobilis Attached to Stainless Steel Wire Spheres

The fermentation of sugar beet juice as well as juice syrup medium by Zymomonas mobilis inoculum attached to stainless steel wire spheres was investigated. A semi-synthetic sucrose medium enriched with mineral salts and yeast extract was used as the control. It was established that raw sugar beet juice ensured good Zymomonas mobilis culture growth and slightly decreased ethanol synthesis applying both flame-burned and TiCl 4 -treated wire spheres as carriers (Q x = 0.05-0.06 g/l × h: Q eth = 1.02-1.22 g/l × h). Hydrophobicity of bacteria Zymomonas mobilis under varied environmental conditions

Abstract Changes in the cell surface hydrophobicity (CSH) of bacteria Zymomonas mobilis 113S were examined in response to varied environmental conditions (temperature and phase of growth, concentration or type of carbon source, the presence of amphiphilic compounds). The values of CSH were elevated with a decreased growth rate over the time of cultivation up to 20–22% at the stationary phase.
