Increase of sulfite tolerance in Oenococcus oeni by means of acidic adaptation

International audience; Sulfite is an antimicrobial agent used at the beginning of winemaking to avoid development of undesirable microorganisms. However, Oenococcus oeni, which is mainly responsible for the malolactic fermentation, has to grow in wine and therefore has to be resistant to sulfite. This study showed that acid-adapted cells of O. oeni survived better than non-adapted cells in the presence of a high sulfite concentration (30 mg l-1). Addition of a sub-lethal concentration of sulfite (15 mg l31) during the adaptation step in acidic medium increases the sulfite tolerance. Reactivity of Neutral Mo(S2C6H4)3 in Aqueous Media: an Alternative Functional Model of Sulfite Oxidase.

The kinetics of the reaction of neutral [Mo(S2C6H4)3] with hydrogen sulfite to produce the anionic Mo(V) complex, [Mo(S2C6H4)3]-, and sulfate have been investigated. It has been shown that [Mo(S2C6H4)3] acts as the electron-proton sink in the oxygenation reaction of HSO3(-) by water. Reaction rates, monitored by UV/vis stopped-flow spectrometry, were studied in THF/water media as a function of the concentration of HSO3(-) and molybdenum complex, pH, ionic strength, and temperature. The reaction exhibits pH-dependent HSO3(-) saturation kinetics, and it is first-order in complex concentration. A study of electrochemically-induced corrosion of low carbon steel in a medium modelling acid rain

Complementary electrochemical, spectrophotometric and electron microsopic investigations were made in addition to the conversion electron Mossbauer spectroscopic (CEMS) measurements to learn more about the mechanism of corrosion of low carbon steel samples in aqueous sulfate and sulfite containing sulfate solutions (pH 3.5, 6.5 and 8.5). The passivation of low carbon steel was studied in aqueous solution of 0.5 M Na2SO4+0.001 M NaHSO3 at pH=3.5 and 6.5. The found major components at pH-3.5 were: γ-FeOOH and Fe3C, and also FeSO4.H2O could be identified on the surface of the low carbon steel as a minor component. At pH-6.5, the passive film contained only amorphous iron(III)-oxide or oxyhydroxide.

Technological properties of Oenococcus oeni strains isolated from typical southern Italian wines.

Aims:  To isolate indigenous Oenococcus oeni strains suitable as starters for malolactic fermentation (MLF), using a reliable polyphasic approach. Methods and Results: Oenococcus oeni strains were isolated from Nero di Troia wines undergoing spontaneous MLF. Samples were taken at the end of alcoholic fermentation and during MLF. Wine samples were diluted in a sterile physiological solution and plated on MRS and on modified FT80.
