This work deals with the synthesis, the chemical characterization of dibutyltin(IV) complex of caffeic acid (Bu2Sn(IV)HCAF, caf1) and its cytotoxic action on tumor cells. The coordination environment at the tin center was investigated by FTIR, (119)Sn{(1)H} cross polarization magic angle spinning, electrospray ionization mass spectroscopy in the solid state and UV-vis, fluorescence and (1)H, (13)C and (119)Sn NMR spectroscopy in solution phases. Establishment and functional validation of a structural homology model for human DNA methyltransferase 1

Changes in DNA methylation patterns play an important role in tumorigenesis. The DNA methyltransferase 1 (DNMT1) protein represents a major DNA methyltransferase activity in human cells and is therefore a prominent target for experimental cancer therapies. However, there are only few available inhibitors and their high toxicity and low specificity have so far precluded their broad use in chemotherapy. Antitumor Effects of a Combined 5-Aza-2′Deoxycytidine and Valproic Acid Treatment on Rhabdomyosarcoma and Medulloblastoma in Ptch Mutant Mice

Abstract Patched (Ptch) heterozygous mice develop medulloblastoma (MB) and rhabdomyosarcoma (RMS) resembling the corresponding human tumors. We have previously shown that epigenetic silencing of the intact Ptch allele contributes to tumor formation in this model. Here, we investigated whether targeting of epigenetic silencing mechanisms could be useful in the treatment of Ptch-associated cancers. The Histone Deacetylase Inhibitor JAHA Down-Regulates pERK and Global DNA Methylation in MDA-MB231 Breast Cancer Cells

The histone deacetylase inhibitor N-1-(ferrocenyl)-N-8-hydroxyoctanediamide (JAHA) down-regulates extracellular-signal-regulated kinase (ERK) and its activated form in triple-negative MDA-MB231 breast cancer cells after 18 h and up to 30 h of treatment, and to a lesser extent AKT and phospho-AKT after 30 h and up to 48 h of treatment. Also, DNA methyltransferase 1 (DNMT1), 3b and, to a lesser extent, 3a, downstream ERK targets, were down-regulated already at 18 h with an increase up to 48 h of exposure. Nitrous oxide emission in a University of Cape Town membrane bioreactor: The effect of carbon to nitrogen ratio

Abstract The effect of the carbon-to-nitrogen (C/N) ratio in the influent on the nitrous oxide (N 2 O) emission from a University of Cape Town Membrane BioReactor pilot plant was investigated. The membrane was located in a separate tank to single out the production of N 2 O due to the biological processes from N 2 O stripping as a result of the extra aeration needed for the mitigation of membrane fouling. The experimental campaign was divided into two phases, each characterized by a different C/N ratio (namely, 10 and 5 mgCOD/mgTN, Phase I and Phase II, respectively). The effect of the solids and hydraulic retention time on moving bed membrane bioreactor performance

Abstract The aim of the present paper was to investigate the effect of solids (SRT) and hydraulic (HRT) retention time on Integrated Fixed Film Activated Sludge (IFAS) University of Cape Town (UCT) membrane Bioreactor (MBR). In particular, three different pairs of SRT and HRT values were analysed, namely, Phase I 56 d/30 h, Phase II 31 d/15 h and Phase III 7 d/13 h.
