GAL4-responsive UAS- tau as a tool for studying the anatomy and development of the Drosophila central nervous system

To improve the quality of cytoplasmic labelling of GAL4-expressing cells in Drosophila enhancer-trap and transgenic strains, a new GAL4-responsive reporter UAS-tau, which features a bovine tau cDNA under control of a yeast upstream activation sequence (UAS), was tested. Tau, a microtubule-associated protein, is distributed actively and evenly into all cellular processes. Monoclonal anti-bovine Tau antibody reveals the axonal structure of the labelled cells with detail similar to that of Golgi impregnation. We demonstrate that the UAS-tau system is especially useful for studying processes of differentiation and reorganisation of identified neurones during postembryonic development.

Streptolysin O-permeabilized granulocytes shed L-selectin concomitantly with ceramide generation via neutral sphingomyelinase

Abstract Cleavage of membrane-associated L-selectin regulates leukocyte rolling on vascular endothelium at sites of inflammation. We report that rapid and massive shedding of L-selectin occurs from granulocytes attacked by the pore-forming bacterial toxin streptolysin O (SLO). Shedding was not induced by an SLO mutant that retained binding capacity but lacked pore-forming activity. Cells permeabilized with SLO exhibited a 1.5-fold increase in the activity of neutral sphingomyelinase, which was accompanied by increased ceramide formation. Identity Double-Proton Transfer in (3Z)-3-Hydroxy-1,4-di(quinolin-2-yl)but-3-en-2-one

Although there is a very fast (on the NMR timescale) double-proton transfer in (1Z,3Z)-3-hydroxy-4-quinolin-2-yl-1-quinolin-2(1H)-ylidenbut-3-en-2-one (the product of the condensation of ethyl oxalate with 2-lithiomethylquinoline), it is the only species present in chloroform solution. Comparison of the product of condensation of ethyl oxalate with 2-lithiomethyl derivatives of pyridine (recent studies) and quinoline (present studies) shows that benzoannulation considerably affects the tautomeric equilibrium. GIAO/DFT calculated chemical shifts of tautomeric species. 2-Phenacylpyridines and (Z)-2-(2-hydroxy-2-phenylvinyl)pyridines

1H, 13C and 15N NMR chemical shifts for 28 substituted 2-phenacylpyridines (ketimine forms) and their enolimine tautomers, (Z)-2-(2-hydroxy-2-phenylvinyl)pyridines, were calculated via the GIAO/DFT approach. Among four tested methods at the B3LYP level of theory, the 6–311G, 6–311++G and 6–311G** basis sets gave acceptable result for 13C NMR chemical shifts whereas the 6–311++G** basis set was the minimum needed for reproduction of 15N NMR chemical shifts. Satisfactory reproduction of 13C and 15N NMR chemical shifts for different tautomers revealed that intramolecular hydrogen bonding could be modeled reliably by these calculations when the geometry optimizations were done with the HF/3–21G…

A molecular electron density theory (MEDT) study of the role of halogens (X2= F2, Cl2, Br2and I2) on the aza-Michael-addition reactions

Using the Molecular Electron Density Theory (MEDT), a set of uncatalyzed and X2 molecule-catalyzed (X2 = F2, Cl2, Br2, or I2) aza Michael addition reactions (aza-MARs) between pyrrolidine (PYR) and methyl acrylate (MA) have been studied in the gas phase and in dichloromethane (DCM) medium from the kinetics and molecular mechanism aspects. Analysis of the conceptual DFT reactivity indices indicates that the coordination of the X2 molecules to the oxygen of the MA's carbonyl group increases both the softness and the electrophilic character of MA, thus explaining the catalytic role of the X2 molecules.
