Are we sure we know how to measure 8-oxo-7,8-dihydroguanine in DNA from human cells?

The most commonly measured marker of oxidative DNA damage is 8-oxo-7,8-dihydroguanine (8-oxoGua) or its deoxyribonucleoside (8-oxodGuo). Published estimates of the concentration of 8-oxoGua/8-oxodGuo in DNA of normal human cells vary over a range of three orders of magnitude. Analysis by chromatographic methods (GC-MS, HPLC with electrochemical detection (ECD) or HPLC-MS/MS) is beset by the problem of adventitious oxidation of guanine during sample preparation. Objectives To assess whether antepartum measurement of the subpubic arch angle (SPAA) as a parameter of the bony pelvic outlet can predict vaginal delivery. Methods Between January 2007 and June 2011, 625 nulliparous women with a singleton pregnancy were recruited from the antenatal clinic to be interviewed and have a four-dimensional transperineal ultrasound examination. Ultrasound examinations were performed between 34 and 36 weeks of gestation. Volume ultrasound data were saved for offline analysis, blinded against all other data. Posidonia oceanica, the dominant Mediterranean seagrass, has been historically described as a species typically growing on mobile substrates whose development requires precursor communities. During more than 10 years of direct observations, we noticed that P. oceanica seedlings were often firmly anchored to rocky reefs, even at exposed sites. The species discrimination in the family Campulidae is largely based on morphometric characters (absolute measurements and ratios). To assess the variability of this characters we have studied the campulid species Hadwenius tursionis from four Mediterranean bottlenose dolphins Tursiops truncatus from the Western Mediterranean. In H. tursionis most absolute measurements differed significantly across the infrapopulations, showing high coefficients of variation. Only egg dimensions appeared to be fairly conservative. Several studies have investigated the differentiation of mitochondrial DNA in Eurasian, African and American cattle as well as archaeological bovine material. A global survey of these studies shows that haplogroup distributions are more stable in time than in space. All major migrations of cattle have shifted the haplogroup distributions considerably with a reduction of the number of haplogroups and/or an expansion of haplotypes that are rare or absent in the ancestral populations. The most extreme case is the almost exclusive colonization of Africa by the T1 haplogroup, which is rare in Southwest Asian cattle.
