Abstract The filamentous fungus Neurospora crassa, a model microbial eukaryote, has a life cycle with many features that make it suitable for studying experimental evolution. However, it has lacked a general tool for estimating relative fitness of different strains in competition experiments. To remedy this need, we constructed N. crassa strains that contain a modified csr-1 locus and developed an assay for detecting the proportion of the marked strain using a post PCR high resolution melting assay. DNA extraction from spore samples can be performed on 96-well plates, followed by a PCR step, which allows many samples to be processed with ease. Furthermore, we suggest a Bayesian approach for…

Impact of Pressurized Liquid Extraction and pH on Protein Yield, Changes in Molecular Size Distribution and Antioxidant Compounds Recovery from Spiru…

2021

The research aims to extract nutrients and bioactive compounds from spirulina using a non-toxic, environmentally friendly and efficient method—Pressurized Liquid Extraction (PLE). In this work, Response Surface Methodology (RSM)–Central Composite Design (CCD) was used to evaluate and optimize the extraction time (5–15 min), temperature (20–60 °C) and pH (4–10) during PLE extraction (103.4 bars). The multi-factor optimization results of the RSM-CCD showed that under the pressure of 103.4 bars, the optimal conditions to recover the highest content of bioactive compounds were 10 min, 40 °C and pH 4. Furthermore, the compounds and antioxidant capacity of PLE and non-pressurized extraction extra…

Chemical composition, herbicidal and antifungal activity of Satureja cuneifolia essential oils from Spain

2016

The chemical composition of essential oils from Satureja cuneifolia growing in east Spain was analyzed by GC, GC/MS. Forty-five compounds accounting for 99.1% of the total oil were identified. Camphor (47.6%), followed by camphene (13.6%) were the main compounds. Their herbicidal and antifungal activity was tested in vitro against three weeds (Amaranthus hybridus, Portulaca oleracea and Conyza canadensis) and eleven common pathogenic or saprophytic fungi (Phytophthora citrophthora, P. palmivora, Pythium litorale, Verticillium dahlia, Rhizoctonia solani, Penicillium hirsutum, Colletotrichum gloeosporioides, Phaeoacremonium aleophilum, Phaemoniella chlamydospora, Cylindrocarpon liriodendri a…

Activation of a nuclear-localized SIPK in tobacco cells challenged by cryptogein, an elicitor of plant defence reactions.

2009

When a plant cell is challenged by a well-defined stimulus, complex signal transduction pathways are activated to promote the modulation of specific sets of genes and eventually to develop adaptive responses. In this context, protein phosphorylation plays a fundamental role through the activation of multiple protein kinase families. Although the involvement of protein kinases at the plasma membrane and cytosolic levels are now well-documented, their nuclear counterparts are still poorly investigated. In the field of plant defence reactions, no known study has yet reported the activation of a nuclear protein kinase and/or its nuclear activity in plant cells, although some protein kinases, e.…

World-wide variation in Drosophila melanogaster sex pheromone: behavioural effects, genetic bases and potential evolutionary consequences

1996

0016-6707 (Print) Journal Article Research Support, Non-U.S. Gov't; In Drosophila melanogaster, male wing vibration, a key element of courtship behaviour, is most efficiently induced by a female-specific contact pheromone cis, cis 7,11 heptacosadiene (7,11 HD), which is the main mature female cuticular hydrocarbon in the CS laboratory strain. A study of 63 strains from around the world revealed that flies from Sub-Saharan Africa and the Caribbean are unique in showing low levels 7,11 HD and high levels of the position isomer 5,9 HD. This difference maps to chromosome III, perhaps indicating a simple genetic control of the 7,11 HD:5,9 HD ratio. Females from strains with high levels of 7,11 H…

Drosophila cuticular hydrocarbons revisited : Mating status alters cuticular profiles

2010

Editor: Frederic Marion-Poll, INRA - Paris 6 - AgroParisTech, France.; International audience; Most living organisms use pheromones for inter-individual communication. In Drosophila melanogaster flies, several pheromones perceived either by contact/at a short distance (cuticular hydrocarbons, CHs), or at a longer distance (cis-vaccenyl acetate, cVA), affect courtship and mating behaviours. However, it has not previously been possible to precisely identify all potential pheromonal compounds and simultaneously monitor their variation on a time scale. To overcome this limitation, we combined Solid Phase Micro-Extraction with gas-chromatography coupled with mass-spectrometry. This allowed us (i…

Innovative alternative technologies to extract carotenoids from microalgae and seaweeds

2016

Marine microalgae and seaweeds (microalgae) represent a sustainable source of various bioactive natural carotenoids, including β-carotene, lutein, astaxanthin, zeaxanthin, violaxanthin and fucoxanthin. Recently, the large-scale production of carotenoids from algal sources has gained significant interest with respect to commercial and industrial applications for health, nutrition, and cosmetic applications. Although conventional processing technologies, based on solvent extraction, offer a simple approach to isolating carotenoids, they suffer several, inherent limitations, including low efficiency (extraction yield), selectivity (purity), high solvent consumption, and long treatment times, w…

Extraction of benzaldehyde from fermentation broth by pervaporation

1996

Abstract The application of pervaporation to extract benzaldehyde produced by microorganisms is considered. A model system was used to study the influence of different parameters and attempts to understand better the mass transfer of this flavour compound. Benzaldehyde was removed from a culture medium from which it was produced by Bjerkandera adusta . The performance of these processes is compared.

Efficient estimation of generalized linear latent variable models.

2019

Generalized linear latent variable models (GLLVM) are popular tools for modeling multivariate, correlated responses. Such data are often encountered, for instance, in ecological studies, where presence-absences, counts, or biomass of interacting species are collected from a set of sites. Until very recently, the main challenge in fitting GLLVMs has been the lack of computationally efficient estimation methods. For likelihood based estimation, several closed form approximations for the marginal likelihood of GLLVMs have been proposed, but their efficient implementations have been lacking in the literature. To fill this gap, we show in this paper how to obtain computationally convenient estim…
