A novel and simple method for the purification of extracellular levansucrase from Zymomonas mobilis.

A new and simple method for the purification of extracellular levansucrase from Zymomonas mobilis from highly viscous fermentation broth was developed. After incubation of the fermentation broth with a fructose-polymer cleaving enzyme preparation (Fructozyme, Novozymes, DK) for 48 h, levansucrase precipitated as aggregates and was redissolved in a 3 M urea solution. By ongoing size-exclusion chromatography on Sephacryl S-300 the final levansucrase preparation was purified 100-fold and exhibited a specific activity of 25-35 U/mg(protein). Cryopreservation of white mulberry (Morus alba L.) by encapsulation-dehydration and vitrification

Shoot apices of in vitro-grown plantlets of white mulberry, Morus alba L. cv Florio, were cryopreserved using either encapsulation-dehydration or vitrification. For encapsulation-dehydration, alginate beads containing apices were dehydrated for 1, 3, 5 or 7 days in a liquid medium containing various sucrose concentrations (0.5, 0.75, 1.0 or 1.25 M). Bead desiccation was performed using silica gel for either 0, 4, 6, 8, 9 or 14 h. For vitrification, apices were directly immersed for either 5, 15, 30 or 60 min in a vitrification solution (PVS2). Studies on sea urchin oocytes. II. Synthesis of RNA during oogenesis.

Abstract Isolated oocytes of the sea urchin Paracentrotus lividus actively incorporate 3H-uridine into RNA. Labeled RNA was analysed by sucrose gradient and acrylamide gel electrophoresis following cell fractionation. Much of the radioactivity is incorporated at the nucleolar level in the form of rRNA precursors. The kinetics of maturation of these latter suggests that this occurs at a slower rate than during embryogenesis. Other non-nucleolar RNA classes are also actively labelled and retained in the nucleus for many hours. These results are confirmed by an autoradiographic investigation.

Echovirus 1 Endocytosis into Caveosomes Requires Lipid Rafts, Dynamin II, and Signaling EventsV⃞

Binding of echovirus 1 (EV1, a nonenveloped RNA virus) to the α2β1 integrin on the cell surface is followed by endocytic internalization of the virus together with the receptor. Here, video-enhanced live microscopy revealed the rapid uptake of fluorescently labeled EV1 into mobile, intracellular structures, positive for green fluorescent protein-tagged caveolin-1. Partial colocalization of EV1 with SV40 (SV40) and cholera toxin, known to traffic via caveosomes, demonstrated that the vesicles were caveosomes. Discovering Copper for Methane C–H Bond Functionalization

The copper complex Tp(CF3)2,BrCu(NCMe) (1, Tp(CF3)2,Br = hydrotris((3,5-bis(trifluoromethyl)-4-bromo)-pyrazol-1-yl)borate) catalyzes the insertion of the CHCO2Et group (from ethyl diazoacetate N2CHCO2Et, EDA) into the C–H bonds of methane, in a homogeneous process that uses supercritical carbon dioxide (scCO2) as the reaction medium. Other light alkanes such as ethane, propane, and butane have been also functionalized with this copper-based catalyst, in the first example of the derivatization of the series of C1–C4 alkanes with this metal and a soluble catalyst.

Deformation of sulfur hexafluoride and floppiness of trifluoromethyl sulfur pentafluoride

International audience; With recent advances in space exploration and atmospheric chemistry there is an increased need for more spectroscopic tools to allow the of study complex species. One such tool is the theory of frame transformation of coupled rotor systems. In this article, the theory of frame transformation along with the concept of rotational energy surface is used to study the symmetry that occurs in trifluoromethyl sulfur pentafluoride due to the internal rotation of the CF3 radical and, more generally, to the extent of floppiness of SF5CF3. Other lower symmetries when a CF4 molecule is stuck on the various symmetry axes of an SF6 molecule are also discussed.

Detached synapses (synaptosomes), first isolated by the author in 1958 and identified as such in 1960, are sealed presynaptic nerve terminals often with a portion of the target cell--sometimes amounting to a complete dendritic spine--adhering to their external surface. They can be prepared in high yield from brain tissue and also in decreasing yield from spinal cord, retina, sympathetic ganglia, myenteric plexus and electric organs. Germination and storage protein mobilisation of Trifolium subterraneum Var 45C seeds under hypoxic treatment

International audience; The rate, speed and latency period of germination were defined for seeds of Trifolium subterraneum variety 45 C. The latency period was 12 hours on distilled water. The hypoxic stress was applied at the beginning (T12) and the end of latency period (T24). It consists on the addition of nitrogen (N2) under 1 bar pressure in hermetically closed boxes during 2 hours. The germination of treated seeds was then assessed on distilled water. The germination test during hypoxic treatment showed that the hypoxic stress delayed germination, slowing down its speed and decreasing the final percentage of germinated seeds. High Speed Stereovision Setup for Position and Motion Estimation of Fertilizer Particles Leaving a Centrifugal Spreader

EA SPE GEAPSI; International audience; A 3D imaging technique using a high speed binocular stereovision system was developed in combination with corresponding image processing algorithms for accurate determination of the parameters of particles leaving the spinning disks of centrifugal fertilizer spreaders. Validation of the stereo-matching algorithm using a virtual 3D stereovision simulator indicated an error of less than 2 pixels for 90% of the particles. The setup was validated using the cylindrical spread pattern of an experimental spreader.
