Analysis of mebendazole binding to its target biomolecule by laser flash photolysis

2016

[EN] Mebendazole (MBZ) and related anticancer benzimidazoles act binding the beta-subunit of Tubulin (TU) before dimerization with alpha-TU with subsequent blocking microtubule formation. Laser flash photolysis (LFP) is a new tool to investigate drug-albumin interactions and to determine binding parameters such as affinity constant or population of binding sites. The aim of this study was to evaluate the interactions between the nonfluorescent mebendazole (MBZ) and its target biomolecule TU using this technique. Before analyzing the MBZ@TU complex it was needed to determine the photophysical properties of MBZ triplet excited state ((3)MBZ*) in different media. Hence, 3MBZ* showed a transien…

Evaluation of beauvericin and enniatins in Italian cereal products and multicereal food by liquid chromatography coupled to triple quadrupole mass sp…

2013

In this study, 48 multicereal baby foods samples including 25 of pasta and 23 of multicereal baby foods from supermarkets of Campania region (Italy) were analysed for evaluating the presence of beauvericin (BEA) and enniatins (ENs) A, A1, B, B1 and B4. Subsequently to evaluate the risk exposure of Italian population and infant population over the consumption of pasta or multicereal baby food, was, respectively, evaluated. For the above mentioned evaluation, a method developed in our laboratory by liquid chromatography tandem mass spectrometry was used. A liquid phase dispersion procedure was optimised for the simultaneous extraction of BEA and the five ENs from multicereal baby food samples…

The algebra of symmetric analytic functions on L∞

2017

We consider the algebra of holomorphic functions on L∞ that are symmetric, i.e. that are invariant under composition of the variable with any measure-preserving bijection of [0, 1]. Its spectrum is identified with the collection of scalar sequences such that is bounded and turns to be separable. All this follows from our main result that the subalgebra of symmetric polynomials on L∞ has a natural algebraic basis.

In Vitro and in Vivo Evaluation of Water-Soluble Iminophosphorane Ruthenium(II) Compounds. A Potential Chemotherapeutic Agent for Triple Negative Bre…

2014

A series of organometallic ruthenium(II) complexes containing iminophosphorane ligands have been synthesized and characterized. Cationic compounds with chloride as counterion are soluble in water (70–100 mg/mL). Most compounds (especially highly water-soluble 2) are more cytotoxic to a number of human cancer cell lines than cisplatin. Initial mechanistic studies indicate that the cell death type for these compounds is mainly through canonical or caspase-dependent apoptosis, nondependent on p53, and that the compounds do not interact with DNA or inhibit protease cathepsin B. In vivo experiments of 2 on MDA-MB-231 xenografts in NOD.CB17-Prkdc SCID/J mice showed an impressive tumor reduction (…

Why small particle fixed dose triple therapy? An excursus from COPD pathology to pharmacological treatment evolution

2022

Although bronchodilators are the cornerstone in chronic obstructive pulmonary disease (COPD) therapy, the treatment with a single-agent bronchodilator may not provide adequate symptoms control in COPD. The combination of drugs with different mechanisms of action may be more effective in inducing bronchodilation and preventing exacerbations, with a lower risk of side-effects in comparison with the increase of the dose of a single molecule. Several studies comparing the triple therapy with the association of long-acting ß2 agonist (LABA)/inhaled corticosteroid (ICS) or long-acting muscarinic antagonist (LAMA)/LABA reported improvement of lung function and quality of life. A significant reduc…

Representation Theorems for Solvable Sesquilinear Forms

2017

New results are added to the paper [4] about q-closed and solvable sesquilinear forms. The structure of the Banach space $\mathcal{D}[||\cdot||_\Omega]$ defined on the domain $\mathcal{D}$ of a q-closed sesquilinear form $\Omega$ is unique up to isomorphism, and the adjoint of a sesquilinear form has the same property of q-closure or of solvability. The operator associated to a solvable sesquilinear form is the greatest which represents the form and it is self-adjoint if, and only if, the form is symmetric. We give more criteria of solvability for q-closed sesquilinear forms. Some of these criteria are related to the numerical range, and we analyse in particular the forms which are solvable…

An isoperimetric type problem for primitive Pythagorean hodograph curves

2012

An isoperimetric type problem for primitive Pythagorean hodograph curves is studied. We show how to compute, for each possible degree, the Pythagorean hodograph curve of a given perimeter enclosing the greatest area. We also discuss the existence and construction of smooth solutions, obtaining a relationship with an interesting sequence of Appell polynomials.

Some algebras of symmetric analytic functions and their spectra

2011

AbstractIn the spectrum of the algebra of symmetric analytic functions of bounded type on ℓp, 1 ≤ p < +∞, and along the same lines as the general non-symmetric case, we define and study a convolution operation and give a formula for the ‘radius’ function. It is also proved that the algebra of analytic functions of bounded type on ℓ1 is isometrically isomorphic to an algebra of symmetric analytic functions on a polydisc of ℓ1. We also consider the existence of algebraic projections between algebras of symmetric polynomials and the corresponding subspace of subsymmetric polynomials.

Tripled Fixed Point Results for T-Contractions on Abstract Metric Spaces

2014

In this paper we introduce the notion of T-contraction for tripled fi xed points in abstract metric spaces and obtain some tripled fi xed point theorems which extend and generalize well-known comparable results in the literature. To support our results, we present an example and an application to integral equations.

The GRP94 Inhibitor PU-WS13 Decreases M2-like Macrophages in Murine TNBC Tumors: A Pharmaco-Imaging Study with 99mTc-Tilmanocept SPECT

2021

Triple-negative breast cancer (TNBC) is the most aggressive subtype of breast cancers and is not eligible for hormone and anti-HER2 therapies. Identifying therapeutic targets and associated biomarkers in TNBC is a clinical challenge to improve patients’ outcome and management. High infiltration of CD206+ M2-like macrophages in the tumor microenvironment (TME) indicates poor prognosis and survival in TNBC patients. As we previously showed that membrane expression of GRP94, an endoplasmic reticulum chaperone, was associated with the anti-inflammatory profile of human PBMC-derived M2 macrophages, we hypothesized that intra-tumoral CD206+ M2 macrophages expressing GRP94 may represent innovative…
