Transporter (TAP)- and proteasome-independent presentation of a melanoma-associated tyrosinase epitope.

2000

The melanosomal protein tyrosinase is considered as a target of specific immunotherapy against melanoma. Two tyrosinase-derived peptides are presented in association with HLA-A2.1 [Wolfel et al., Eur. J. Immunol., 24, 759-764 (1994)]. Peptide 1-9 (MLLAVLYCL) is generated from the putative signal sequence. The internal peptide 369-377 is posttranslationally converted at residue 371, and its presentation is dependent on functional TAP transporters and proteasomes [Mosse et al., J. exp. Med.187, 37-48 (1998)]. Herein, we report on the processing and transport requirements for the signal sequence-derived peptide 1-9 that were studied in parallel to those for peptide 369-377. After infection of …

Differentiating Multiple Sclerosis From AQP4-Neuromyelitis Optica Spectrum Disorder and MOG-Antibody Disease With Imaging.

2023

Background and ObjectivesRelapsing-remitting multiple sclerosis (RRMS), aquaporin-4 antibody–positive neuromyelitis optica spectrum disorder (AQP4-NMOSD), and myelin oligodendrocyte glycoprotein antibody–associated disease (MOGAD) may have overlapping clinical features. There is an unmet need for imaging markers that differentiate between them when serologic testing is unavailable or ambiguous. We assessed whether imaging characteristics typical of MS discriminate RRMS from AQP4-NMOSD and MOGAD, alone and in combination.MethodsAdult, nonacute patients with RRMS, APQ4-NMOSD, and MOGAD and healthy controls were prospectively recruited at the National Hospital for Neurology and Neurosurgery (L…

Volcanism at Slab Tear Faults: the Diamante-Enotrio-Ovidio volcanic complex (offshore north-west Calabria)

2018

Several arc-shape sectors form the central and southern part of the Italian peninsula, representing the emerged portion of the continental crust disjoined by slab tear faults (e.g., Olevano-Antrodoco, Ortona-Roccamonfina Tindari-Letojanni faults). These sectors are characterized by different drift velocities and tectonic patterns, while above to them important volcanic phases developed, such as the Vulture volcano (onshore) and the Vulcano-Lipari-Salina alignment (offshore). In the Tyrrhenian basin such kind of volcanism is still poorly investigated and understood though it could be revealed by low-resolution geophysical anomalies and volcanic seafloor morphologies. This is probably due to …

Oligodendrocytes myelinate neuronal axons in the central nervous system (CNS) facilitating rapid transmission of action potentials by saltatory conduction. Myelin basic protein (MBP) is an essential component of myelin and its absence results in severe hypomyelination in the CNS of rodents. Mbp mRNA is not translated immediately after exit from the nucleus in the cytoplasm, but is transported to the plasma membrane in RNA transport granules in a translationally silenced state. We have previously identified the small non-coding RNA 715 (sncRNA715) as an inhibitor of Mbp translation associated with RNA granules. Argonaute (Ago) proteins and small RNAs form the minimal core of the RNA induced …

Kinetic Properties of Cl−Uptake Mediated by Na+-Dependent K+-2Cl−Cotransport in Immature Rat Neocortical Neurons

2007

GABA, the main inhibitory neurotransmitter in the adult nervous system, evokes depolarizing membrane responses in immature neurons, which are crucial for the generation of early network activity. Although it is well accepted that depolarizing GABA actions are caused by an elevated intracellular Cl−concentration ([Cl−]i), the mechanisms of Cl−accumulation in immature neurons are still a matter of debate. Using patch-clamp, microfluorimetric, immunohistochemical, and molecular biological approaches, we studied the mechanism of Cl−uptake in Cajal-Retzius (CR) cells of immature [postnatal day 0 (P0) to P3] rat neocortex. Gramicidin-perforated patch-clamp and 6-methoxy-N-ethylquinolinium-microfl…

Survey of the mean pressure exerted by a wide range of tractors on the soil

2010

In order to predict the mean pressure exerted by a vehicle tyre on the soil, in 1984 Plackett suggested measuring the contact area of a tyre on a hard surface. The model proposed by Hallonborg in 1996, based on the “super ellipse theory”, provided the best results in terms of prediction of the tyre-soil contact area. In this study 82 4WD tractors with different power, mass and age of manufacture were surveyed. Relying on their technical features, the contact area of the front and rear tyres of each tractor and, therefore, their pressure on the soil was calculated, in order to assess the attention paid by manufacturers to soil compaction. The results of this survey showed that the mean press…

Arc and forearc rifting in the Tyrrhenian subduction system

2022

The evolution of forearc and backarc domains is usually treated separately, as they are separated by a volcanic arc. We analyse their spatial and temporal relationships in the Tyrrhenian subduction system, using seismic profiles and numerical modelling. A volcanic arc, which included the Marsili volcano, was involved in arc-rifting during the Pliocene. This process led to the formation of an oceanic backarc basin (~ 1.8 Ma) to the west of the Marsili volcano. The eastern region corresponded to the forearc domain, floored by serpentinised mantle. Here, a new volcanic arc formed at ~ 1 Ma, marking the onset of the forearc-rifting. This work highlights that fluids and melts induce weakening of…

Improvement of the photo-stability of polystyrene-block-polybutadiene-block-polystyrene through carbon nanotubes

2015

Abstract The photo-stability of Polystyrene-Polybutadiene-Polystyrene (SBS) based nanocomposites containing bare multi-walled carbon nanotubes (CNTs) and carbon nanotubes bearing carboxylic functional groups (CNTs-COOH) in comparison to that of pristine SBS has been studied. The photo-oxidation of pristine SBS occurs through crosslinking reactions and oxidized species formation and both these processes begin at early stage of exposure. The formation of crosslinking, formerly in polybutadiene phase, assessed by spectroscopical (FTIR), mechanical, dynamic mechanical and rheological analysis, leads to occurrence of internal mechanical stresses in the solid state and the SBS samples become prem…

Influence of substrate and temperature on the biodegradation of polyester-based materials: Polylactide and poly(3-hydroxybutyrate-co-3-hydroxyhexanoa…

2020

[EN] The extended use of polymers from renewable resources such as aliphatic polyesters or polyhydroxyalkanoates boosted the necessity to understand their behaviour in an end-of-life scenario. Although they can be degraded in reasonable shorter times than traditional polymers, understanding the degradation mechanisms under dissimilar conditions will contribute to further developments in this field. This work aimed to study the effect of temperature and substrate in the degradation of polylactide (PLA) and poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) (PHBH) in a simulated laboratory scale to ascertain their contribution, separately or in combination. For this purpose, nine parallel degradat…

The influence of solvent on conformational properties of peptides with Aib residue—a DFT study

2017

The conformational propensities of the Aib residue on the example of two model peptides Ac-Aib-NHMe (1) and Ac-Aib-NMe2 (2), were studied by B3LYP and M06-2X functionals, in the gas phase and in the polar solvents. To verify the reliability of selected functionals, we also performed MP2 calculations for the tested molecules in vacuum. Polarizable continuum models (PCM and SMD) were used to estimate the solvent effect. Ramachandran maps were calculated to find all energy minima. Noncovalent intramolecular interactions due to hydrogen-bonds and dipole attractions between carbonyl groups are responsible for the relative stabilities of the conformers. In order to verify the theoretical results,…
