Es wurden mit einem Lichtstreuphotometer die Orientierungs- und Dichtefluktuationen in den amorphen Polymeren «Polycarbonat» (Poly(oxycarbonyloxy-1,4-phenylenisopropyliden-1,4-phenylen)) und Polystyrol in Abhangigkeit von der Temperatur untersucht. Ferner fuhrten wir erganzende Messungen an Oligomeren des Polystyrols und an der flussig-kristallinen Verbindung 4-Butyl-N-(p-methoxybenzyliden)anilin (MBBA) in der isotropen Phase durch. Aus der Hohe der depolarisierten Lichtstreuung ergab sich, das eine Orientierungskorrelation, wie sie fur einige Bundelmodelle vorgeschlagen wurde, nicht auftritt. The effect of combining sodium butyrate (NaB), a histone deacetylase inhibitor, and 7-hydroxy-staurosporine (UCN-01) on cytotoxicity in human cervical carcinoma cells was evaluated.HeLa and CaSki cells were treated using NaB alone or in combination with staurosporine (STS) or its analog UCN-01. Cytotoxicity was determined by flow cytometry and morphological assays. Apoptotic pathways were characterized by Western blotting and immunostaining. CaSki cells were also xenografted into nude mice to assess the in vivo effects of NaB/UCN-01 combination.Treatment with NaB and STS or UCN-01 resulted in enhanced apoptosis of cancer cells. Use of CDC2 from etoposide-treated cells as substrate to assay CDC25 phosphatase activity

International audience; Cyclin-dependent kinases (CDKs) regulate the key transition of the cell cycle in all organisms. In response to Etoposide (VP-16) induced DNA damage, cells undergo a G2-phase arrest resulting in the accumulation of inactive CDK1 (CDC2) kinase complexes. Here we report that upon Etoposide treatment CDC2 is phosphorylated on tyrosine 15 and is dephosphorylated and activated in vitro by recombinant CDC25 phosphatase. We also show that inactive CDC2 kinase from Etoposide-treated cells can be used as a substrate in a sensitive two-step assay of CDC25 phosphatase. In the last years evidence was presented for the activation of dendritic cells (DC) under stimulation with contact sensitizers. Most data were obtained for murine Langerhans cells (LC) whereas in man blood-derived dendritic cells were found to be a more suitable model to study the mechanism of DC activation by haptens. The first observation was the upregulation of MHC class II molecules on murine Langerhans cells in vivo1 followed by their migration from the epidermis into regional lymph nodes2. National audience; The Celluloïd taste observed in some mould-ripened cheeses is bound to the presence of styrene in these cheeses. The accumulation of this molecule was only detected on cultures of some strains of Penicillium camemberti. It seems to be bound to a deregulation of oxidative metabolism of tneee strains.; L'apparition du goût de Celluloïd dans les fromages à pâte molle à croûte fleurie est liée à la présence de styrène dans ces fromages. L'accumulation de cette molécule n'est observée que sur des cultures de souches particulières de Penicillium camemberti. Elle semble liée à un dérèglement du métabolisme oxydatif de ces souches.

Tyrosinases, which are widely distributed among animals, plants and fungi, are involved in many biologically essential functions, including pigmentation, sclerotization, primary immune response and host defence. In the present study, we present a structural and physicochemical characterization of two new tyrosinases from the crustaceans Palinurus elephas (European spiny lobster) and Astacus leptodactylus (freshwater crayfish). In vivo, the purified crustacean tyrosinases occur as hexamers composed of one subunit type with a molecular mass of approx. 71kDa. Oxazole ring occurs in numerous natural peptides, but conformational properties of the amino acid residue containing the oxazole ring in place of the C-terminal amide bond are poorly recognized. A series of model compounds constituted by the oxazole-amino acids occurring in nature, that is, oxazole-alanine (L-Ala-Ozl), oxazole-dehydroalanine (ΔAla-Ozl), and oxazole-dehydrobutyrine ((Z)-ΔAbu-Ozl), was investigated using theoretical calculations supported by FTIR and NMR spectra and single-crystal X-ray diffraction. It is now well-established that the surface chemistry and “stealth” surface functionalities such as poly(ethylene glycol) (PEG) chains of nanocarriers play an important role to decrease unspecific protein adsorption of opsonizing proteins, to increase the enrichment of specific stealth proteins, and to prolong the circulation times of the nanocarriers. At the same time, PEG chains are used to provide colloidal stability for the nanoparticles.
