Nuclear and Cytoplasmic Soluble Proteins Extraction from a Small Quantity of Drosophila’s Whole Larvae and Tissues

The identification and study of protein’s function in several model organisms is carried out using both nuclear and cytoplasmic extracts. For a long time, Drosophila’s embryos have represented the main source for protein extractions, although in the last year, the importance of collecting proteins extracts also from larval tissues has also been understood. Here we report a very simple protocol, improved by a previously developed method, to produce in a single extraction both highly stable nuclear and cytoplasmic protein extracts from a small quantity of whole Drosophila’s larvae or tissues, suitable for biochemical analyses like co-immunoprecipitation.
Technical Note: Comparison of Techniques for Evaluating the Relative Preference by Sheep Among Saltbush Clones

This research compared 4 field methods of evaluating the relative preference by sheep of 28 clones of saltbush (Atriplex halimus L.). The methods were as follows. 1) Leaf dots (LD): 8 leaves per shrub were marked on the lower surface with a small dot using a water-resistant, nontoxic ink. 2) Twig marks (TM): 2 current-year twigs per shrub were marked with 3 lines using the same ink approximately in the middle of the basal, median, and apical thirds. 3) Branch length (BL): 2 branches per shrub were marked with ink at the base of the current year's growth. The Italian Hydrographic Service publishes yearly maximum rainfall depths with 1-, 3-, 6-, 12-, and 24-hr duration, recorded by recording rain gauges. It also publishes selected short-duration (t<1 hr) rainfall amounts. It is impossible to carry out a statistical analysis of the short-duration rainfalls for each recording rain gauge because the published data correspond to different record durations and to different recording rain gauges from year to year. Therefore, there is a practical need to derive empirical relationships between the 1-, 3-, 6-, 12-, and 24-hr duration amounts and shorter duration (t<1 hr) rainfall amounts. Stress separation in the photoelastic study of centrifugal stresses

This technical note refers to the problem of stress separation in the photoelastic analysis of plane models under centrifugal stresses. Two methods are described in order to determine the sum of principal stresses. These methods, which are based on the compatibility equation, reduce the determination of the sum of principal stresses to the solution of a Laplace's or Poisson's equations. As an example of application, the separation of stresses in a rotating disk with two eccentric holes is shown and comparison with the stresses obtained by using the shear-difference method is made.

Abstract In the paper the authors present the results of several experimental tests aimed to determine the Coulomb friction coefficient in sheet metal forming operations at the varying of the sheet metal material and for different operative conditions. INTRODUCTION Head-mounted mixed-reality technologies may enable advanced intraoperative visualization during visceral surgery. In this technical note, we describe an innovative use of real-time mixed reality during robotic-assisted transanal total mesorectal excision. TECHNIQUE Video signals from the robotic console and video endoscopic transanal approach were displayed on a virtual monitor using a head-up display. mBISON: Finding miRNA target over-representation in gene lists from ChIP-sequencing data

Background Over-representation of predicted miRNA targets in sets of genes regulated by a given transcription factor (e.g. as defined by ChIP-sequencing experiments) helps to identify biologically relevant miRNA targets and is useful to get insight into post-transcriptional regulation. Findings To facilitate the application of this approach we have created the mBISON web-application. mBISON calculates the significance of over-representation of miRNA targets in a given non-ranked gene set.
