Detection of antigen-specific T-cells with MHC/peptide-tetramer-complexes

2002

Abstract Tetramer analysis is a novel technique in immunological research that has dramatically changed our knowledge of the immune response to viral and bacterial pathogens, to tumors and in autoimmune disease. Through the formation of major histocompatibility complex (MHC)/peptide-tetrameric complexes it can provide accurate counts of antigen-specific T-cells and it allows their phenotypical and functional analysis. We review the strengths of this method and use the analysis of peripheral blood lymphocytes (PBL) from a patient with melanoma as a paradigm for the detection of tetramer-binding T-cells. The implementation of tetramer-guided analysis in immunomonitoring allows the ex vivo tes…

MZ-35, a new layered pentasil borosilicate synthesized in the presence of large alkali cations

2013

Abstract A new layered borosilicate has been synthesized in the presence of cesium and sodium cations and its structure has been solved by a combination of automated diffraction tomography (ADT) and X-ray powder diffraction (XRPD). MZ-35 has a composition NaCs 2 [BSi 7 O 16 (OH) 2 ](OH) 2 ·4H 2 O and features space group P-4m2. The unusually small unit cell ( a 7.3081 A, c 10.7520 A) is shared by two random-stacked configurations of the structure: a network of connected pentasil units related to the layer of RUB-18 and a bidimensional checkerboard of intersecting ladders of 4-membered rings. The two configurations are related by the simple face-sharing inversion of a hydroxyl-bearing tetrah…

Safety, tolerability, pharmacokinetics, pharmacodynamics, and exploratory efficacy of the novel enzyme replacement therapy avalglucosidase alfa (neoG…

2019

This multicenter/multinational, open-label, ascending-dose study (NCT01898364) evaluated safety, tolerability, pharmacokinetics, pharmacodynamics, and exploratory efficacy of repeat-dose avalglucosidase alfa (neoGAA), a second-generation, recombinant acid α-glucosidase replacement therapy, in late-onset Pompe disease (LOPD). Patients ≥18 years, alglucosidase alfa naïve (Naïve) or previously receiving alglucosidase alfa for ≥9 months (Switch), with baseline FVC ≥50% predicted and independently ambulatory, received every-other-week avalglucosidase alfa 5, 10, or 20 mg/kg over 24 weeks. 9/10 Naïve and 12/14 Switch patients completed the study. Avalglucosidase alfa was well-tolerated; no deaths…

Assortative mating and female clutch investment in black grouse

1999

Variation in female behaviour has only recently received attention in studies of sexual selection. It has been suggested that females may invest differentially in their offspring in relation to the quality of their mate. This may lead to females that mate with high-quality and/or attractive males laying larger clutches. Females may also differ in their ability to choose between males. For example, females in good physical condition may make better choices. If physical condition and clutch size are positively correlated, this hypothesis could also produce a relationship between male attractiveness and female clutch size. We found, in lekking black grouse, Tetrao tetrix, that females mated to…

Spacer-separated sialyl LewisX cyclopeptide conjugates as potential E-selectin ligands.

2006

Completely protected sialyl LewisX azide was synthesized from a neolactosamine azide precursor carrying a 3-O-allyloxycarbonyl group as the temporary protecting group. After its Pd(0)-catalyzed deprotection and stereoselective alpha-fucosylation, the obtained LewisX azide was subjected to O-deacetylation in the galactose unit and subsequent regio- and stereoselective sialylation. Reduction of the anomeric azido group afforded the sialyl LewisX amine building block. Two molecules of this tetrasaccharide ligand were conjugated to a preformed cyclooctapeptide containing two equidistant l-asparagine units equipped with carboxy-terminated tetraethyleneglycol side chains to give, after deprotecti…

MTHFR 677C → T genotype modulates the effect of a 5-year supplementation with B-vitamins on homocysteine concentration: The SU.FOL.OM3 randomized con…

2018

Aims To study how MTHFR 677C→T genotype modulates the effect of supplementation with B-vitamins on total homocysteine (tHcy) and B-vitamin concentrations. Methods 2381 patients with a personal history of cardiovascular disease were randomly assigned to one of four groups: 1) B-vitamins alone (560 μg of 5-methyl-THF, 3 mg of vitamin B6 and 20 μg of vitamin B12), 2) n-3 fatty acids alone (600 mg of EPA and DHA in a 2:1 ratio), 3) B-vitamins and n-3 fatty acids, and 4) placebo. Participants were followed up for 4.7 years. At baseline and annually thereafter, biological parameters were assessed. Multivariate and linear mixed models were fit to study the interaction between B-vitamins and MTHFR …

4,4-Di-fluoro-2,3;5,6-bis-(tetra-methylene)-4-bora-3a,4a-di-aza-s-indacene (LD540).

2014

The title compound, C18H21BF2N2, is a lipophilic dye based on a BODIPY fluorophore backbone, which was developed for microscopic imaging of lipid droplets; the molecule has a planar BODIPY core [dihedral angle between the pyrrole rings = 2.3 (3)°] and two tetramethylene substituents at the 2,3- and 5,6-positions in a half-chair conformation. One of the tetramethylene substituents is disordered over two two sets of sites with site occupancies of 0.5. In the crystal, pairs of C—H...F interactions link the molecules into inversion dimers. Neighbouring dimers are linked by further C—H...F interactions, forming an infinite array. C—H...π and π–π [centroid–centroid distance = 4.360 (3) Å] interac…

Top-down effects of a lytic bacteriophage and protozoa on bacteria in aqueous and biofilm phases

2014

Lytic bacteriophages and protozoan predators are the major causes of bacterial mortality in natural microbial communities, which also makes them potential candidates for biological control of bacterial pathogens. However, little is known about the relative impact of bacteriophages and protozoa on the dynamics of bacterial biomass in aqueous and biofilm phases. Here, we studied the temporal and spatial dynamics of bacterial biomass in a microcosm experiment where opportunistic pathogenic bacteria Serratia marcescens was exposed to particle‐feeding ciliates, surface‐feeding amoebas, and lytic bacteriophages for 8 weeks, ca. 1300 generations. We found that ciliates were the most efficient enem…
