Light and electron microscopical demonstration of methylene blue accumulation sites in taste buds of fish and mouse after supravital dye injection
1995
Electron microscopical data regarding methylene blue staining of taste buds in the epithelia of the goldfish lip and the cirumvallate papilla of the mouse tongue after supravital dye application are presented for the first time. The ultrastructural details were compared with the corresponding light microscopical findings. The dye was applied in different concentrations by injection or in crystalline from directly to the surface of the tissues. Both methylene blue and tissue were simultaneously fixed by immersion in a paraformaldehyde-glutaraldehyde solution with the addition of phosphomolybdic acid. The ensuing dye precipitate was further stabilized by ammonium heptamolybdate. On the light …
Phenothiazines interfere with dopaminergic neurodegeneration in Caenorhabditis elegans models of Parkinson's disease
2010
Oxidative stress is involved in the pathogenesis of various neurodegenerative disorders, conventional antioxidant strategies have yet been of limited success. We have employed transgenic Caenorhabditis elegans expressing DsRed2 in dopaminergic neurons and CFP pan-neuronally, to characterize in larval and adult animals the effects of rotenone and 1-methyl-4-phenyl-pyridinium (MPP(+)) on the dopaminergic system. Investigating the antioxidant phenothiazine and different derived antipsychotic drugs, it was found that free phenothiazine exerted strong neuroprotection at the cellular level and resulted in a better performance in behavioral assays, whereas apomorphine and other dopamine agonists o…
Update on topical carbonic anhydrase inhibitors
2001
Topical carbonic anhydrase inhibitors are a novel addition to the armamentarium of medical glaucoma treatment; dorzolamide has been available since 1995 and brinzolamide since 1998. They lower intraocular pressure by inhibiting carbonic anhydrase, a key enzyme for aqueous humor formation. Intraocular pressure-lowering activity of the substances appears to be the same and is similar to that of most other agents, but it does not reach the activity of the unselective beta-blocker timolol or the prostaglandin latanoprost. On concomitant treatment, additivity is reached with all other topical agents. A possible improvement of blood flow may offer an additional benefit, but its significance for t…
Effect of the linkage location in double branched organic dyes on the photovoltaic performance of DSSCs
2015
Two novel double branched D–π–A organic dyes (DB dyes) are synthesized to investigate the influence of the linkage location in DB dyes on the performance of dye-sensitized solar cells (DSSCs), where phenothiazine is introduced as a donor, thiophene–benzotriazole unit as the π-bridge and cyanoacrylic acid as the electron-acceptor. The photophysical, electrochemical and photovoltaic properties of the dyes are systematically investigated. The results show that the location of the linkage unit has a small effect on the physical and electrochemical properties of the dyes. However, when the dyes are applied in DSSCs, an obvious decline of short-circuit current (Jsc) and open-circuit voltage (Voc)…
Photoinduced chemiluminescence of pharmaceuticals
2005
Abstract A screening test for the forward development of chemiluminescence systems able to determine pharmaceutical compounds is reported. The test is based on the on-line photodegradation of the drugs by using a photoreactor consisting of 697 cm × 0.5 mm PTFE tubing helically coiled around an 8 W low-pressure mercury lamp. Photodegraded pharmaceuticals are detected by direct chemiluminescence of the resulting photofragments and their subsequent reaction with potassium permanganate in sulphuric acid medium as oxidant. The screening comprised 97 compounds with different molecular structures and relevant members of the most important families of pharmaceuticals are tested (amino acids, carbox…
Der Pulmonalarteriendruck des Menschen bei Kreislaufvernderungen im Phenothiazin-Schlaf
1957
Randomized Trial of Brinzolamide/Brimonidine Versus Brinzolamide Plus Brimonidine for Open-Angle Glaucoma or Ocular Hypertension
2014
Introduction Fixed-combination intraocular pressure (IOP)—lowering medications simplify treatment regimens for patients requiring 2 ocular hypotensive agents to maintain sufficiently low IOP. The aim of this study was to evaluate the safety and efficacy of fixed-combination brinzolamide 1%/brimonidine 0.2% (BBFC) versus concomitant administration of brinzolamide 1% plus brimonidine 0.2% (BRINZ + BRIM) in patients with open-angle glaucoma or ocular hypertension. Methods This was a prospective, phase 3, multicenter, double-masked, 6-month trial. Patients who had insufficient IOP control with monotherapy or who were receiving 2 IOP-lowering medications were randomized 1:1 to receive twice-dail…
The antioxidative, non-psychoactive tricyclic phenothiazine reduces brain damage after experimental traumatic brain injury in mice.
2014
Abstract Oxidative stress due to free radical formation is an important mechanism of secondary brain damage following traumatic brain injury (TBI). Phenothiazine has been found to be a strong antioxidant in eukaryotic cells in vitro and in invertebrates in vivo. The present study was designed to determine the neuroprotective potency of unsubstituted phenothiazine in a paradigm of acute brain injury. Thirty minutes after pneumatic, controlled cortical impact (CCI) injury, C57BI6 mice were randomly assigned to “low dose” (3 mg/kg, LD) or “high dose” (30 mg/kg, HD) s.c. phenothiazine or vehicle treatment. Brain lesion, neurofunctional impairment, body weight, and markers of cerebral inflammati…
Brinzolamide/brimonidine fixed-dose combination bid as an adjunct to a prostaglandin analog for open-angle glaucoma/ocular hypertension.
2019
Purpose: To evaluate the additive intraocular pressure–lowering effect of twice-daily brinzolamide 1%/brimonidine 0.2% fixed-dose combination (BBFC) as an adjunct to a prostaglandin analog (PGA) in patients with open-angle glaucoma or ocular hypertension insufficiently controlled with PGA monotherapy. Methods: In this Phase 4, double-masked trial, patients aged ⩾18 years, with a mean intraocular pressure of ⩾19 and <32 mm Hg in at least one eye were randomized (1:1) to receive BBFC + PGA ( n = 96) or vehicle + PGA ( n = 92) for 6 weeks. The primary endpoint was the mean change in diurnal intraocular pressure from baseline (averaged over 09:00 and 11:00 h) at Week 6. Results: The mean diu…
Metal-free organic dyes with di(1-benzothieno)[3,2-b:2′,3′-d]pyrrole as an auxiliary donor for efficient dye-sensitized solar cells: Effect of the mo…
2019
Abstract A series of novel di(1-benzothieno)[3,2-b:2′,3′-d]pyrrole (DBTP)-based organic dyes (WL5-10) with D-D-π-A or D-D-A-π-A configurations are designed and the relationship between the properties and molecular configurations of these organic dyes are studied systematically. WL5,7–8 with D-D-π-A configuration incorporating triphenylamine or phenothiazine as donor and DBTP as auxiliary donor and WL9-10 with D-D-A-π-A configuration incorporating benzothiadiazole as an auxiliary acceptor are synthesized to study the effect of the molecular engineering on the photovoltaic performance. WL5,7–8 exhibit similar absorption spectra and high molar extinction coefficient. Especially, the dye WL5 wi…
