PLS-NIR determination of total sugar, glucose, fructose and sucrose in aqueous solutions of fruit juices

Abstract An analytical procedure has been developed for the direct determination of sugars in fruit juice samples. The method is based on the partial least-squares (PLS) treatment of first derivative near infrared (NIR) spectroscopic data obtained between 1200 and 2450 nm, using 1 mm pathlength cell and a multicomponent calibration set, including seven binary mixtures and 10 ternary mixtures of glucose, fructose and sucrose. The limit of detection values found are of the order of 0.2 g/100 ml for total sugar and 0.2, 0.4 and 0.5 g/100 ml for glucose, fructose and sucrose, respectively. Fludioxonil is a fungicide with a singular mode of action that is widely employed in the treatment of fruit crops. A competitive enzyme-linked immunosorbent assay (cELISA) has been developed and validated for the determination of residues of this fungicide in fruit juices. A collection of monoclonal antibodies (mAbs) against the fungicide fludioxonil has been produced and characterized using direct and indirect cELISAs. The high affinity achieved (IC50lower than 0.5 μg L-1) considerably improved that obtained with previous polyclonal antibodies. Four hundred seventy residents of Ridgewood, New Jersey, and 493 residents of Jyvaskyla, Finland, were randomly selected and interviewed about their attitudes concerning nuclear war. In both areas, a high proportion of the samples believed that some kind of nuclear incident is likely in the next decade. The vast majority stated that a nuclear war could not be won and that they expected to die in the event of war. About three fourths in both communities seldom discuss nuclear issues with anyone including their own children, and only one fourth of the U.S sample and about one half of the Finnish sample have ever done anything personally to oppose the arms race.
