Apurinic acid (APA) and apyrimidinic acid (APyA) prepared from deoxyribonucleic acid (DNA) by removing either purine or pyrimidine bases have been reported to inhibit enzymatic DNA-synthesis in vitro (Heicke, 1970). The present paper deals with in vivo effects of APA and APyA on mammalian cells in tissue culture.

A radioactive preludin preparation, C14 labelled on the 2-carbon of the tetrahydrooxazine ring, was injected into mice in doses of 100 mg/kg s.c. and rats in doses of 10 mg/kg i.v. The radioactivity of the tissues and excreta was followed for 48 hours. An activity concentration surpassing the average body concentration was obtained for a short time in the lungs and for a longer time in kidneys, liver and in the wall of stomach and small intestine. 95–99% of the total activity is found in the urine. The kidney excretion is virtually finished at the end of 8 hours; 1–5% appear after the 8th hour in faeces. No radioactivity was obtained from the expired CO2. The main excretion organs are kidne…

Evidence that ATP or a related purine is an excitatory neurotransmitter in the longitudinal muscle of mouse distal colon

Background and purpose: This study analysed the contribution of the purinergic system to enteric neurotransmission in the longitudinal muscle of mouse distal colon. Experimental approach: Motor responses to exogenous ATP and to nerve stimulation in vitro were assessed as changes in isometric tension. Adenosine negatively regulates duodenal motility in mice: role of A1 and A2A receptors

BACKGROUND AND PURPOSE Adenosine is considered to be an important modulator of intestinal motility. This study was undertaken to investigate the role of adenosine in the modulation of contractility in the mouse duodenum and to characterize the adenosine receptor subtypes involved. EXPERIMENTAL APPROACH RT-PCR was used to investigate the expression of mRNA encoding for A1, A2A, A2B and A3 receptors. Contractile activity was examined in vitro as changes in isometric tension. Surface molecularly imprinted silica for selective solid-phase extraction of biochanin A, daidzein and genistein from urine samples

Selective molecularly imprinted silica polymer (SiO2MIP) for extraction of biochanin A, daidzein and genistein was synthesized using the surface molecular imprinting technique with the silica gel as a support. Biochanin A (BCA) was used as a template, 3-aminopropyltriethoxysilane (APTES) as a functional monomer, and tetraethoxysilicane (TEOS) as a cross-linker. Non-imprinted polymer with the sol–gel process (SiO2NIP) was also prepared for comparison. An efficient sample clean-up and preconcentration procedure for phytoestrogens analysis in urine has been developed. It was based on a combination of solid phase extraction with hollow-fiber supported liquid membrane and molecularly imprinted beads (MIPs-HF-SLM-SPE). The molecularly imprinted polymers (MIPs) were synthesized by precipitation polymerization technique with biochanin A (BCA) as a template, giving narrowly dispersed microspheres with a regular shape.
