Stromal cell-derived factor-1alpha (SDF-1alpha) is a potent chemoattractant for hematopoietic progenitor cells (HPC), suggesting that it could play an important role during their migration within or to the bone marrow (BM). The integrin VLA-4 mediates HPC adhesion to BM stroma by interacting with CS-1/fibronectin and VCAM-1. It is required during hematopoiesis and homing of HPC to the BM. Išēmisks insults ir viens no biežākajiem nāves un invaliditātes cēloņiem pasaulē. Rezultātā tiek būtiski bojāti smadzeņu rajoni, kas izraisa motoros traucējumus. Lai izprastu išēmiska smadzeņu bojājuma izraisītās motorās komplikācijas cilvēkiem, pielieto preklīniskos uzvedības testus izmēģinājuma dzīvniekiem. Maģistra darba mērķis bija, izmantojot preklīnisku insulta modeli, izpētīt muskuļu koordinācijas un muskuļu spēka izmaiņas pelēm, pielietojot kondziela un rotarod testus, divu mēnešu periodā. Endothelial cells (EC) are very responsive to proinflammatory cytokines, e.g. interleukin-1 (IL-1) and tumor necrosis factor alpha (TNF alpha), as well as to bacterial lipopolysaccharide. EC are stimulated by these substances to secrete chemotactic factors and to increase expression of cell adhesion molecules (CAM), leading to dramatically altered interactions with leukocytes, e.g. granulocytes and monocytes. Peļu aktivitātes izmaiņas atklātā lauka testā pēc insulta modelēšanas

Išēmisks insults var izraisīt gan neiroloģiskas izmaiņas, tai skaitā sensorimotorās funkcijas un izziņas traucējumus, gan uzvedības izmaiņas. Atklātā lauka tests tiek plaši izmantots, lai novērtētu un analizētu peļu spontāno jeb vispārējo lokomotoro aktivitāti. Bakalaura darba mērķis ir izvērtēt peļu aktivitāti sešus mēnešus pēc insulta modelēšanas, izmantojot atklātā lauka testu. Iegūtie rezultāti rāda, ka Stroke grupas peles uzrāda ievērojami lielāku aktivitāti, griežoties pretpulksteņrādītāja virzienā, salīdzinot gan ar Naive, gan ar Sham grupas pelēm, kas norāda uz insulta bojājumu kontralaterālajā pusē. SUMMARY VCAM-1 was first identified as an adhesion molecule induced on human endothelial cells (HEC) by inflammatory cytokines such as IL-1, tumour necrosis factor (TNF), and lipopolysaccharide (LPS). The molecule binds to a variety of leucocytes, including B cells, T cells, basophils, eosinophils and monocytes.
