Specific Features of the Coagulopathy Signature in Severe COVID-19 Pneumonia

2021

Rationale: COVID-19 displays distinct characteristics that suggest a unique pathogenesis. The objective of this study was to compare biomarkers of coagulopathy and outcomes in COVID-19 and non-COVID-19 patients with severe pneumonia. Methods: Thirty-six non-COVID-19 and 27 COVID-19 non-immunocompromised patients with severe pneumonia were prospectively enrolled, most requiring intensive care. Clinical and biological characteristics (including plasma biomarkers of coagulopathy) were compared. Results: At similar baseline severity, COVID-19 patients required mechanical ventilation (MV) for significantly longer than non-COVID-19 patients (p = 0.0049) and more frequently developed venous thrombot…

Antibodies to proteinase 3 mediate expression of vascular cell adhesion molecule-1 (VCAM-1).

1996

SUMMARY VCAM-1 was first identified as an adhesion molecule induced on human endothelial cells (HEC) by inflammatory cytokines such as IL-1, tumour necrosis factor (TNF), and lipopolysaccharide (LPS). The molecule binds to a variety of leucocytes, including B cells, T cells, basophils, eosinophils and monocytes. Vascular expression of VCAM-1 has been associated with a number of disease states, including rheumatoid arthritis and vasculitis. The detection of antineutrophil cytoplasmic antibodies (ANCA), especially to proteinase 3 (PR3), has become important in the diagnosis of Wegener’s granulomatosis (WG) and related vasculitides. Recently we were able to demonstrate a direct effect of anti-…

Redox signaling in acute pancreatitis

2015

Acute pancreatitis is an inflammatory process of the pancreatic gland that eventually may lead to a severe systemic inflammatory response. A key event in pancreatic damage is the intracellular activation of NF-κB and zymogens, involving also calcium, cathepsins, pH disorders, autophagy, and cell death, particularly necrosis. This review focuses on the new role of redox signaling in acute pancreatitis. Oxidative stress and redox status are involved in the onset of acute pancreatitis and also in the development of the systemic inflammatory response, being glutathione depletion, xanthine oxidase activation, and thiol oxidation in proteins critical features of the disease in the pancreas. On th…

Angiogenesis of oral lichen planus: a possible pathogenetic mechanism.

2008

Objective: Oral Lichen Planus (OLP) is a chronic inflammatory disease with an autoimmune inflammatory pathogenesis. The aim of the research is to compare the vascular endothelial growth factor (VEGF) and adhesion of molecules in the biopsy samples of patients affected by OLP, in order to research the presence of the angiogenetic phenomenon and to understand its pathogenetic mechanism. Materials and Methods: Thirty OLP patients and thirty healthy subjects were enrolled in a study. The immunohistochemical analysis of the VEGF and vascularendothelial adhesion molecules was carried out by means of primary antibodies and anti-CD34, anti-VEGF, antiCD106 antigen (VCAM-1) and anti-CD54 antigen (ICA…

Increased expression of ICAM-1, E-selectin, and VCAM-1 by cultured human endothelial cells upon exposure to haptens.

1992

Contact allergens induce several accessory signals which promote the activation of antigen-specific T cells. One of these signals is the increased expression of adhesion molecules on antigen-presenting cells and endothelial cells. Epicutaneous application of non-toxic doses of 2,4-dinitrofluorobenzene (DNFB) onto the skin of non-sensitized individuals elicited progressive staining for ICAM-1 on dermal microvascular endothelial cells. To elucidate the question of whether contact allergens can act directly on endothelial cells to elevate their expression of surface structures that bind leukocytes, confluent monolayers of human umbilical vein endothelial cells were incubated with the contact a…

Interaction of endothelial cells and neutrophilsin vitro: kinetics of thrombomodulin, intercellular adhesion molecule-1 (ICAM-1), E-selectin, and vas…

1999

SUMMARYRecently markers of endothelial cell activation or injury gained increasing interest as serological parameters of disease activation in vasculitides. Among these, soluble serum thrombomodulin, ICAM-1, VCAM-1 and E-selectin are of particular interest. However, only thrombomodulin showed the expected close correlation. The objective of this study was to investigate in vitro the kinetics of these endothelial cell receptors after interaction of unstimulated or cytokine-activated polymorphonuclear neutrophils (PMN) and endothelial cells in order to find evidence explaining these different clinical findings. Over the time period of up to 48 h of incubation the kinetics of thrombomodulin, I…

Peļu aktivitātes izmaiņas atklātā lauka testā pēc insulta modelēšanas

2020

Išēmisks insults var izraisīt gan neiroloģiskas izmaiņas, tai skaitā sensorimotorās funkcijas un izziņas traucējumus, gan uzvedības izmaiņas. Atklātā lauka tests tiek plaši izmantots, lai novērtētu un analizētu peļu spontāno jeb vispārējo lokomotoro aktivitāti. Bakalaura darba mērķis ir izvērtēt peļu aktivitāti sešus mēnešus pēc insulta modelēšanas, izmantojot atklātā lauka testu. Iegūtie rezultāti rāda, ka Stroke grupas peles uzrāda ievērojami lielāku aktivitāti, griežoties pretpulksteņrādītāja virzienā, salīdzinot gan ar Naive, gan ar Sham grupas pelēm, kas norāda uz insulta bojājumu kontralaterālajā pusē. Naive grupas pelēm ievērojami samazinās aktivitāte, salīdzinot ar pirmo testēšanas …

Chronic consumption of an inositol-enriched beverage ameliorates endothelial dysfunction and oxidative stress in type 2 diabetes

2015

The anti-diabetic properties of an inositol-enriched beverage (IEB) on the endothelial function and redox status in diabetic subjects were assessed. This was a 12-week, double-blind randomized controlled trial employing thirty-eight diabetic subjects that were divided into two intervention groups: one receiving an IEB and the other a sucrose-enriched beverage. Subjects consuming IEB exhibited a significant decrease in triacylglycerol (8.82%) and HbA1c (4.53%) levels. Continuous glucose monitoring revealed a significant net reduction of 2.51 and 7.11% during postprandial and overnight fasting periods after consumption of IEB, respectively. Moreover, IEB improved endothelial function by reduc…

Gastrin induces the interaction between human mononuclear leukocytes and endothelial cells through the endothelial expression of P-selectin and VCAM-…

2009

Gastric mucosal inflammation is frequently associated with hypergastrinemia, and a correlation exists between the level of gastrin and degree of gastritis. We have previously observed that gastrin promotes leukocyte-endothelial interactions and contributes to Helicobacter -induced inflammation in the rat mesentery. In the present study, we aimed to evaluate a possible proinflammatory activity of gastrin in humans. The interaction between human leukocytes [U-937 cells, peripheral blood polymorphonuclear (PMN), and peripheral blood mononuclear (PBMC) cells] and human umbilical vein endothelial cells (HUVEC) was analyzed in static and dynamic conditions. The endothelial expression of adhesion …

Motoro funkciju novērtēšana rotarod un kondziela testos pelēs išēmiska insulta modelī

2022
