SOV approach for integrable quantum models associated to general representations on spin-1/2 chains of the 8-vertex reflection algebra

The analysis of the transfer matrices associated to the most general representations of the 8-vertex reflection algebra on spin-1/2 chains is here implemented by introducing a quantum separation of variables (SOV) method which generalizes to these integrable quantum models the method first introduced by Sklyanin. More in detail, for the representations reproducing in their homogeneous limits the open XYZ spin-1/2 quantum chains with the most general integrable boundary conditions, we explicitly construct representations of the 8-vertex reflection algebras for which the transfer matrix spectral problem is separated. Low-temperature spectrum of correlation lengths of the XXZ chain in the antiferromagnetic massive regime

We consider the spectrum of correlation lengths of the spin-$\frac{1}{2}$ XXZ chain in the antiferromagnetic massive regime. These are given as ratios of eigenvalues of the quantum transfer matrix of the model. The eigenvalues are determined by integrals over certain auxiliary functions and by their zeros. The auxiliary functions satisfy nonlinear integral equations. We analyse these nonlinear integral equations in the low-temperature limit. In this limit we can determine the auxiliary functions and the expressions for the eigenvalues as functions of a finite number of parameters which satisfy finite sets of algebraic equations, the so-called higher-level Bethe Ansatz equations. Space and Time Averaged Quantum Stress Tensor Fluctuations

We extend previous work on the numerical diagonalization of quantum stress tensor operators in the Minkowski vacuum state, which considered operators averaged in a finite time interval, to operators averaged in a finite spacetime region. Since real experiments occur over finite volumes and durations, physically meaningful fluctuations may be obtained from stress tensor operators averaged by compactly supported sampling functions in space and time. Protein delivery by subviral particles of human cytomegalovirus

Direct protein delivery is an emerging technology in vaccine development and gene therapy. We could previously show that subviral dense bodies (DB) of human cytomegalovirus (HCMV), a beta-herpesvirus, transport viral proteins into target cells by membrane fusion. Thus these non-infectious particles provide a candidate delivery system for the prophylactic and therapeutic application of proteins. Here we provide proof of principle that DB can be modified genetically. A 55 kDa fusion protein consisting of the green fluorescent protein and the neomycin phosphotransferase could be packed in and delivered into cells by recombinant DB in a functional fashion. AAV Vector–mediated RNAi of Mutant Huntingtin Expression Is Neuroprotective in a Novel Genetic Rat Model of Huntington's Disease

We report the characterization of a new rapid-onset model of Huntington's disease (HD) generated by adeno-associated virus (AAV) vector–mediated gene transfer of N-terminal huntingtin (htt) constructs into the rat striatum. Expression of exon 1 of mutant htt containing 70 CAG repeats rapidly led to neuropathological features associated with HD. In addition, we report novel data relating to neuronal transduction of AAV vectors that modulated the phenotype observed in this model. Quantitative reverse transcriptase–polymerase chain reaction (RT–PCR) revealed that AAV vector–mediated expression in the striatum increased by >100-fold as compared to the endogenous htt level. Efficacy of recombinant adenovirus as vector for allergen gene therapy in a mouse model of type I allergy

DNA-based immunization represents an attractive alternative approach to the current treatment of allergic diseases by specific immunotherapy with allergen extracts. In this study, we used a replication-deficient adenovirus vector (AdCMV), to examine the in vivo efficacy of preventive and therapeutic genetic immunization in a mouse model of type I allergy. Primary immunization with a recombinant adenovirus expressing the model antigen beta-galactosidase (AdCMV-(beta)gal) induced a Th1 immune response (predominance of IgG2a antibodies, high frequency of IFN-gamma producing T cells) and large numbers of cytotoxic T lymphocytes. Mutilation of RNA phage Qβ virus-like particles: from icosahedrons to rods

Icosahedral virus-like particles (VLPs) of RNA phage Qbeta are stabilized by four disulfide bonds of cysteine residues 74 and 80 within the loop between beta-strands F and G (FG loop) of the monomeric subunits, which determine the five-fold and quasi-six-fold symmetry contacts of the VLPs. In order to reduce the stability of Qbeta VLPs, we mutationally converted the amino acid stretch 76-ANGSCD-81 within the FG loop into the 76-VGGVEL-81 sequence. It led to production in Escherichia coli cells of aberrant rod-like Qbeta VLPs, along with normal icosahedral capsids. The length of the rod-like particles exceeded 4-30 times the diameter of icosahedral Qbeta VLPs.

Prevention of long-term IgE antibody production by gene gun-mediated DNA vaccination.

Background Vaccination with allergen-encoding DNA represents a promising approach for the treatment of allergic diseases. Objective In a mouse model of type I allergy, we analyzed the ability of biolistic transfection to inhibit antigen-specific IgE production and to modulate T H 2 responses. Methods BALB/c mice were vaccinated by means of gene gun–mediated DNA immunization with plasmid vector pCMV-βGal, encoding β-galactosidase as a model allergen. Cell surface display of rat invariant γ chain: detection by monoclonal antibodies directed against a C-terminal γ chain segment

A series of 14 monoclonal antibodies (mAb) directed against the C-terminal part of the rat invariant gamma chain (amino acid 142-216) was generated using distinct fusion proteins that contain this gamma segment for immunization and hybridoma screening. Additional fusion protein were prepared carrying discrete regions of the gamma chain. Employing these reagents confirmed that the obtained mAb do indeed recognize the C-terminal portion of the invariant chain, as demonstrated by Western blot analysis. Continuous spectrum for a two phase eigenvalue problem with an indefinite and unbounded potential

Abstract We consider a two phase eigenvalue problem driven by the ( p , q ) -Laplacian plus an indefinite and unbounded potential, and Robin boundary condition. Using a modification of the Nehari manifold method, we show that there exists a nontrivial open interval I ⊆ R such that every λ ∈ I is an eigenvalue with positive eigenfunctions. When we impose additional regularity conditions on the potential function and the boundary coefficient, we show that we have smooth eigenfunctions.
