ChemInform Abstract: Synthesis and Pharmacological Evaluation of 1-Methyl-5- [substituted-4(3H)-oxo-1,2,3-benzotriazin-3-yl]-1H-pyrazole-4-acetic Aci…

2010

Abstract Several new 1-methyl-5-[substituted-4-oxo-1,2,3-benzotriazin-3-yl]-1H-pyrazole-4-acetic acids and their ethyl ester derivatives were prepared. The compounds were tested for analgesic and antiinflammatory activities, acute toxicity, ulcerogenic effect, and as in vitro inhibitors of 3α-hydroxysteroid dehydrogenase (3α-HSD), since it is claimed that the inhibition of such an enzyme predicts in vivo antiinflammatory activity. Some compounds were more active than phenylbutazone in the phenylbenzoquinone and acetic acid peritonitis tests, and equiactive to the same drug in the carrageenin paw edema test. All the compounds inhibited the 3α-HSD, but no correlation was observed with the paw…

Absence of binding of human salivary glycoprotein to human gingival fibroblast-like cells in vitro.

1996

The aim of this study was to determine whether human high molecular weight salivary glycoprotein binds in vitro to human gingival fibroblast-like cells. Primary monolayer cultures of 2 human gingival fibroblast-like cell lines were incubated with a high molecular weight fraction of salivary glycoprotein which expressed blood group A activity and glycoprotein-cell binding probed using an FITC-conjugated mouse monoclonal antibody to human blood group A antigen. Surface fluorescence of protein-treated cells was found to be no greater than that of untreated or serum-treated control cultures. As significant binding of salivary glycoprotein to gingival fibroblast-like cells does not occur in vitr…

Synthesis, structural aspects and bioactivity of the marine cyclopeptide hymenamide C

2001

Abstract Head-to-tail proline containing cyclopeptide hymenamide C [cyclo(Leu-Trp-Pro 3 -Phe-Gly-Pro 6 -Glu); 1 ], isolated from a marine sponge and for which a preliminary immunomodulating activity was reported, was efficiently synthesized by a three-dimensional orthogonal solid-phase strategy (Fmoc/tBu/Allyl) via anchoring the ω -carboxyl function of the glutamic acid to the solid support (PAC–PEG–PS). The linear precursor was entirely assembled and subsequently cyclized on resin, yielding a major product identical to the natural hymenamide C and a minor one, a geometric isomer of hymenamide C ( 2 ), differing for the geometry of peptide linkages at Pro residues. Both the ‘proline-rich’ c…

Chemical conjugation of dexamethasone to a polyaspartamide and in vitro evaluation studies

2004

Two macromolecular conjugates of dexamethasone containing different drug amounts were synthesized using PHEA as the polymeric carrier and a succinic group as spacer. The content of linked drug was equal to 25.3% w/w (conjugate A) and 12.7% w/w (conjugate B). Both polymeric conjugates, unlike the free drug, were water-soluble and the amount of unlinked drug was evaluated to be approximately about 0.01% w/w. Both conjugates were relatively stable in vitro at pH 7.4 whereas in the presence of esterase only the conjugate B was able to release drug under the used experimental conditions. This dissimilar behavior has been attributed to the distinct macromolecular conformations assumed in aqueous …

Self-assembly properties of the proteinaceous coat secreted by the ?slime? variant of Neurospora crassa

1989

The proteinaceous extracellular material (PEM) synthesized by the cells of the ‘slime” strain of Neurospora crassa (see Martinez et al. 1989) was solubilized by treatment with urea or guanidine. Removal of these chemicals by dialysis, caused reassembly of the solubilized proteins into material with the same microscopic appearance as the original PEM. Polypeptide patterns from both native and reassembled structures were identical. Dialysis-mediated reassembly of the solubilized proteins appeared to be dependent on both concentration of the soluble macromolecules and time. Gel chromatography of PEM solubilized with different agents revealed two discrete populations of complexes with molecular…

Loss of Contact-Dependent Inhibition of Growth in Chemically Transformed Fibroblasts

1988

The plasma membrane has been recognized as an important regulatory unit of mammalian cells during determination, differentiation, and social behaviour of individual cells within various tissues (1–4). On the molecular level, plasma membrane glycoproteins and glycolipids have been shown to be involved in these processes (1–4). Density-dependent growth of non-transformed cells in vitro has been proposed to be regulated by secreted inhibitory compounds (5–7), by the cell’s shape (8) or by diffusion boundary layers (9). On the other hand, specific cell-cell interactions via cell membrane molecules were found to be of great importance for the contact-dependent inhibition of growth (10–16) and co…

EFFECTS OF CYCLOPROPENOID FATTY ACIDS (BAOBAB SEED OIL) ON THE FATTY ACID PROFILE OF LIPIDS FROM DIFFERENT TISSUES IN THE RAT

1996

Rats were fed for 8 weeks a diet containing either Baobab seed oil (FBO) rich in cyclopropenoid fatty acids (CPFA) or heated oil (HBO) devoid of CPFA. In FBO rats, compared to controls (CO), the monounsaturated fatty acid content of adipose tissue lipids, liver triacylglycerols, liver and adrenal cholesteryl esters and liver, kidney, adrenal phospholipids were highly decreased to the benefit of saturated fatty acid content. In cholesteryl esters and phospholipids, the arachidonic acid (20:4 n-6) content was markedly decreased, especially in adrenals. In adrenal cholesteryl esters, adrenic acid (22:4 n-6) content was decreased more than its precursor. The effects of HBO on the tissue lipid f…

Comparison of the Biosynthesis of Cellulose in vitro and in vivo in Cotton Bolls

1966

THE work of Hassid et al.1–3 on the cell-free synthesis of cellulose with an enzyme system isolated from mung bean seedlings and young cotton bolls has shown that the enzyme is apparently unable to distinguish guanosine diphosphate-D-glucose from guanosine diphosphate-D-mannose. Moreover, there was a notable decrease in the amount of the synthesized cellulose using enzymes from cotton bolls older than 15 days.

The immunosuppressive enzyme IL4I1 promotes FoxP3+regulatory T lymphocyte differentiation

2015

Abstract IL4I1 (interleukin-4-induced gene 1) is a phenylalanine oxidase produced mainly by APCs of myeloid origin, and converts phenylalanine (Phe) to phenylpyruvate, hydrogen peroxide, and ammonia. We have previously shown that IL4I1 is highly expressed by tumor-associated macrophages from various human cancers and facilitates immune evasion from the cytotoxic response in a murine tumor model. Indeed, IL4I1 inhibits T-cell proliferation via hydrogen peroxide toxicity on effector/memory T cells. Here, we explored the effect of IL4I1 on naive CD4(+) T-cell differentiation. We show that IL4I1 stimulates the generation of Foxp3(+) regulatory T (Treg) cells in vitro from human and mouse T cell…

Effect of plant growth regulators and basal media onin vitro shoot proliferation and rooting ofMyrtus communis L.

1996

The influence of macronutrients and growth regulators on in vitro shoot proliferation and rooting of an East Spanish population ofMyrtus communis L. were studied. Preincubation of field material on a medium without mineral salts prevented the browning from phenolic exudates. For multiplication, nodal segments of 5 mm fromin vitro produced shoots were cultured on Murashige and Skoog (MS), Schenk and Hildebrandt (SH) and Heller (H) media (full strength or diluted to 1/2 or 1/4), with 6-benzylaminopurine (BAP) at concentrations 4.4, 13.3 and 22.2 ΜM or kinetin (K) at concentrations 4.7, 14.0 and 23.2 ΜM. The optimum shoot proliferation was on quarter-strength MS medium with 4.4 ΜM BAP, whereas…
