Evidence for a role of inducible nitric oxide synthase in gastric relaxation of mdx mice

Alterations of gastric mechanical activity have been reported in mdx mouse, animal model for Duchenne muscular dystrophy. This study examined if alterations in the vasoactive intestinal polypeptide (VIP) system are present in mdx stomach. Gastric mechanical activity was recorded in vitro as changes of endoluminal pressure and neurally or pharmacologically evoked relaxations were analysed in mdxvs normal stomach. Reverse-transcription polymerase chain reaction was used to detect inducible nitric oxide synthase (iNOS) expression. Global end-diastolic volume (GEDV) as determined at the bedside by the transpulmonary technique is a reliable marker of cardiac preload in critically ill patients. As shown by numerous studies, GEDV is superior to cardiac filling pressures in various settings in anesthesia and intensive care medicine in patients undergoing mechanical ventilation. The objectives are as follows: To assess the efficacy as well as adverse events of emergency sclerotherapy (or band ligation) combined with pharmacological treatments versus sclerotherapy (or band ligation) alone or vasoactive drugs alone for acute variceal bleeding in cirrhotic patients based on the results of randomised clinical trials (RCTs).

NANC inhibitory neurotransmission in mouse isolated stomach: Involvement of nitric oxide, ATP and vasoactive intestinal polypeptide

The neurotransmitters involved in NANC relaxation and their possible interactions were investigated in mouse isolated stomach, recording the motor responses as changes of endoluminal pressure from whole organ. 2. Field stimulation produced tetrodotoxin-sensitive, frequency-dependent, biphasic responses: rapid transient relaxation followed by a delayed inhibitory component. 3. The inhibitor of the synthesis of nitric oxide (NO), L-NAME, abolished the rapid relaxation and significantly reduced the slow relaxation. The present immunohistochemical study demonstrates the multiplicity, histotopography and origin of peptidergic innervation in the mammalian heart. Neuropeptide Y (NPY) is the major representative of peptides in cardiac sympathetic efferents. Sympathetic afferents are characterized by the presence of tachykinins, calcitonin gene-related peptide and apparently also some opioid peptides. Predominant peptides of the vagal system are tachykinins. The intrinsic peptidergic system predominantly consists of vasoactive intestinal polypeptide/peptide histidine isoleucine. Chromogranin A in the Mammalian Merkel Cell: Cellular and Subcellular Distribution

Chromogranin-A (CGA), which accounts for more than half the soluble matrix protein in secretory granules of various neuroendocrine cells, has a wide spectrum of potential biological roles and is considered an important marker of the diffuse neuroendocrine system (DNES). Light and electron microscopic immunohistochemistry of mammalian skin revealed that Merkel cells are exclusively CGA-immunoreactive (ir) and that the immunoreaction is localized in the secretory granules. This finding supports the classification of the Merkel cell as a member of the DNES. The CGA immunoreactivity was restricted to Merkel cells of pigs and humans. Expression and regulation of mPer1 in immortalized GnRH neurons.

Hypothalamic GnRH (gonadotropin-releasing hormone) neurons play a critical role in the initiation and maintenance of reproduction competence. Using the mouse GnRH neuronal cell line, GT1-7, we have characterized the expression of the gene mPer1, a recognized key element of the mammalian circadian clockwork. Both mPer1 transcripts and the 136 kDa mPER1 gene product could be detected in these cells. Immunocytochemical analysis also confirmed expression of mPER1 both in vitro and in vivo in GnRH neurons.
