Vitrification of Immature Porcine Oocytes: Effects of Lipid Droplets, Temperature, Cytoskeleton, and Addition and Removal of Cryoprotectant

Three experiments were conducted to investigate the effects of single-step and stepwise exposure to and removal of cryoprotectant, of temperature, and of a cytoskeletal relaxant on the development of germinal vesicle porcine oocytes to the M-II stage. In experiment I, noncooled cumulus-oocyte complexes (COCs) were treated using single-step/stepwise exposure to ethylene glycol (EG) and removal at 23 or 42 degrees C. Stepwise exposure to EG and dilution at 42 degrees C were found to have a positive effect on the COC developmental rate. Vitrification of human mature oocytes in clinical practice.

Oocyte cryopreservation has long been the focus of unsuccessful efforts to perfect its clinical application. Lately, vitrification has been shown to provide high degrees of success by different research groups. This review attempts to address the efficiency of oocyte vitrification in clinical practice through an analysis of reports of IVF cycles in which embryo transfers have been conducted with vitrified metaphase-II human oocytes.

Cryopreservation of Limonium serotinum apical meristems from in vitro plantlets using droplet-vitrification

Abstract In this study in vitro shoot tips of a Sicilian genotype of Limonium serotinum were successfully cryopreserved using the droplet-vitrification technique. Growth recovery of cryopreserved shoot tips was possible only when samples were pretreated for 16 h in liquid medium with 0.3 M sucrose, then for 5 h in liquid medium with 0.7 M sucrose before performing the cryopreservation protocol. Comparing encapsulation-dehydration and droplet-vitrification for cryopreservation of sugarcane (Saccharum spp.) shoot tips

Abstract In this study, in vitro shoot tips of two sugarcane clones were successfully cryopreserved using encapsulation-dehydration and droplet-vitrification with two vitrification solutions, PVS2 and PVS3. For both clones, encapsulation-dehydration induced significantly higher recovery, reaching 60% for clone H70-144 and 53% for clone CP68-1026, compared with droplet-vitrification in which recovery was 33–37% for clone H70-144 and 20–27% for clone CP68-1026. Optimal conditions included preculture of encapsulated shoot apices for 24 h in liquid medium with 0.75 M sucrose and dehydration with silica gel to 20% moisture content (fresh weight basis) before direct immersion in liquid nitrogen. …

The structural effect of trehalose confined in water-containing sodium bis(2-ethylhexyl)sulfosuccinate (AOT) reversed micelles at water to AOT molar ratio W = 5 and 10 as a function of the trehalose to AOT molar ratio T (0 < T < 0.1) has been investigated by small-angle neutron scattering (SANS). SANS data analysis is consistent with the hypothesis that trehalose is encapsulated within the quite spherical hydrophilic micellar cores of water-containing reversed micelles, causing an increase of the aggregate size and a decrease of the polydispersion. The in vivo survival rate of rabbit morulae after vitrification in a mixture of dimethyl sulphoxide and ethylene glycol solution without protein supplement (WPS) was compared with two types of protein supplements: rabbit serum (RS) and bovine serum albumin (BSA). Significant dif- ferences were observed in the percentage of transferable embryos (undamaged embryos after devit- rification, 80.4 % versus 93.2 and 92.1 %, WPS, BSA and RS, respectively, P < 0.05) and live born rate (40.9 % versus 56.1 %, WPS and BSA, respectively, P < 0.05). Halogen (Cl, F) release during explosive, effusive, and intrusive phases of the 2011 rhyolitic eruption at Cordón Caulle volcano (Chile)

We investigate sulphur, chlorine and fluorine release during explosive, effusive and intrusive phases of the 2011 Cordon Caulle eruption, with a focus on halogen devolatilization. Petrological analysis shows halogen release to have been promoted by isobaric crystallization in slowly-cooled magma that was emplaced in a lava flow and sub-vent intrusion. Fluorine in particular mobilized only after extensive groundmass crystallization and incipient devitrification. By 2017, the gas emitted from vent-proximal fumaroles had hydrothermal compositions, with HCl/HF ratios decreasing with decreasing temperature. In certain areas rhyolitic tuffs of the Upper Rotliegend include a greenish, talc-like substance, called “Hygrophyllit”. It has been found too at the base of the Rotliegend in the Saarland. It is shown, that Hygrophyllit corresponds to decomposed pumice, related to the acid permian volcanism. X-ray, DTA and chemical investigations yielded, that Hygrophyllit consists essentially of a mixed layer mica-Ca-montmorillonite with random interstratification and previously about 33 % expandable layers.
