Elongation speed is a key parameter in RNA polymerase II (RNA pol II) activity. It affects the transcription rate, while it is conditioned by the physicochemical environment it works in at the same time. For instance, it is well-known that temperature affects the biochemical reactions rates. Therefore in free-living organisms that are able to grow at various environmental temperatures, such as the yeast Saccharomyces cerevisiae, evolution should have not only shaped the structural and functional properties of this key enzyme, but should have also provided mechanisms and pathways to adapt its activity to the optimal performance required. Reactivation of the HMGA1 protoncogene is very frequent in human cancer, but still very little is known on the molecular mechanisms leading to this event. Prompted by the finding of putative E2F binding sites in the human HMGA1 promoter and by the frequent deregulation of the RB/E2F1 pathway in human carcinogenesis, we investigated whether E2F1 might contribute to the regulation of HMGA1 gene expression. In the rat pineal gland cAMP mediates nocturnal induction of the enzyme arylalkylamine N-acetyltransferase (AA-NAT) as well as of transcription factors such as inducible cAMP early repressor (ICER), Fos-related antigen-2 (Fra-2) and JunB. Cyclic AMP stimulates the phosphorylation of the DNA binding protein cAMP response element binding protein (CREB). While cAMP-induced CREB phosphorylation appears to be a prerequisite for AA-NAT and ICER gene expression, it is not known whether CREB phosphorylation accounts for the full cAMP response of the two genes. Ionizing radiation-induced E-selectin gene expression and tumor cell adhesion is inhibited by lovastatin and all-trans retinoic acid

E-selectin mediated tumor cell adhesion plays an important role in metastasis. Here we show that ionizing radiation (IR) induces E-selectin gene and protein expression in human endothelial cells at therapeutically relevant dose level. E-selectin expression is accompanied by an increase in the adhesion of human colon carcinoma cells to primary human umbilical vein endothelial cells (HUVEC). The HMG-CoA reductase inhibitor lovastatin impairs IR-stimulated E-selectin expression as analyzed at the level of the protein, mRNA and promoter. Dendritic cell-derived IL-12p40 homodimer contributes to susceptibility in cutaneous leishmaniasis in BALB/c mice

Abstract Protection against Leishmania major in resistant C57BL/6 mice is mediated by Th1 cells, whereas susceptibility in BALB/c mice is the result of Th2 development. IL-12 release by L. major-infected dendritic cells (DC) is critically involved in differentiation of Th1 cells. Previously, we reported that strain differences in the production of DC-derived factors, e.g., IL-1αβ, are in part responsible for disparate disease outcome. In the present study, we analyzed the release of IL-12 from DC in more detail. Stimulated DC from C57BL/6 and BALB/c mice released comparable amounts of IL-12p40 and p70. Placental alkaline phosphatase types in Germany

The phenotypes of placental alkaline phosphatase were determined in a sample of 231 Germans and 109 non-Germans. The observed gene frequencies of the German sample were \({\text{Pl}}^{{\text{S}}_{\text{1}} } {\text{ = 0}}{\text{.654}}\), \({\text{Pl}}^{{\text{F}}_{\text{1}} } {\text{ = 0}}{\text{.247}}\), and \({\text{Pl}}^{{\text{I}}_{\text{1}} } {\text{ = 0}}{\text{.097}}\). No association could be found with placental weight. Lower birth weight was correlated with an increase of the \({\text{Pl}}^{{\text{I}}_{\text{1}} } \) gene frequency.

International solidarity in the GDR and transnationality: an analysis of primary school materials for Namibian child refugees

As part of a solidarity project between the South West African People’s Organization (SWAPO) and the German Democratic Republic (GDR), approximately 430 Namibian children were brought to the GDR from 1979 to 1989 to be trained as an elite for a future liberated Namibia. The children attended school in the GDR until they were brought back to Namibia in August 1990. The school lessons intertwined topics about Namibia and SWAPO with the usual GDR school curriculum. The linchpin of this intertwining was the socialist ideal of international solidarity. This article uses an objective-hermeneutic analysis to show how the school materials produced transnationality.
